Effects of vehicles and elicitation concentration in contact dermatitis testing. I. Experimental contact sensitization in humans.

A study was made to evaluate the effects of vehicle and challenge concentration on response of human subjects to potential allergens. In the vehicle studies the modified Draize test was used to test the response of subjects to cinnamic aldehyde and to costus oil, administered at two skin sites, in petrolatum and in 95% ethyl alcohol. In two tests of costus oil, alcohol proved to be more effective in eliciting a response than petrolatum; on the other hand, in one test with cinnamic aldehyde, no difference in results was obtained with these two vehicles. In the concentration studies, subjects kn

Feedback modalities in human-cobot collaboration: experimental evaluation of performance, user experience, and physiological responses.

Collaborative robots (cobots) are increasingly deployed in industrial as well as non-industrial domains to support human-centered operation. While physical safety and task efficiency have received considerable attention, less is known about how feedback modality influences operator experience and physiological responses under different collaboration demands. This study examines the effects of feedback modalities in two human-cobot collaboration scenarios representing distinct coordination structures: a low-collaboration turn-taking task and a high-collaboration synchronous shared-control task.

Patterns of Inflammation in Experimental Autoimmune Uveitis and Their Correlation to Optical Coherence Tomography Findings in Human Uveitis.

Experimental autoimmune uveitis (EAU) in rats is a pivotal model for understanding the immunological mechanisms of human uveitis and developing therapies. In humans, optical coherence tomography (OCT) allows for the in vivo detection of characteristic findings in active uveitis, as well as sequelae of inflammation. The objective of this study was to correlate OCT findings in patients with uveitis with retinal histologies from two rat models of EAU caused by T cells with different autoantigen specificities and well-known underlying immunological pathomechanisms. Patients with various noninfecti

Interaction of the NSAID cyclooxygenase inhibitor, acemetacin, with human serum albumin: an experimental and computational modelling investigation.

Acemetacin (ACM), a nonsteroidal anti-inflammatory drug (NSAID) used to treat chronic pain and inflammation, is known for its improved gastrointestinal tolerance compared to other NSAIDs. Clarifying how ACM associates with human serum albumin (HSA) is crucial for understanding its transport, distribution, and bioavailability. The ACM binding to HSA was examined through a combination of spectroscopic, microscopic, and computational methods. Fluorescence quenching confirmed spontaneous and moderate binding, with the binding constant (Ka) values of 7.52±0.03 × 104, 7.15±0.00 × 104, and 5.54±0.08

Experimental evaluation of a real-time implementation of compensatory reserve measurement in a human model of hemorrhagic shock.

INTRODUCTION: The leading cause of preventable traumatic death is hemorrhage. Early detection of hemorrhagic shock remains a critical challenge. For the early prediction of hemorrhagic shock-related cardiovascular decompensation, our team has developed the compensatory reserve measurement (CRM) algorithm. CRM uses a photoplethysmography waveform to quantify the body's capacity to compensate during hypovolemia. This study focuses on the development and use of an application that can predict CRM in real-time (CRMRT) during simulated hypovolemia experiments. METHODS: The CRMRT application was de

In vivo Cytokine Adsorption Reveals Distinct Neutrophil Mobilization Pathways during Experimental Human Endotoxemia.

INTRODUCTION: During acute inflammation, such as experimental human endotoxemia, circulating neutrophil counts initially decrease, followed by pronounced neutrophilia. Banded (CD16dim/CD62Lbright) and hypersegmented (CD16bright/CD62Ldim) neutrophils, which are absent in the blood during homeostasis, are rapidly mobilized. Banded and mature neutrophils are thought to be recruited from the bone marrow, while the origin of hypersegmented neutrophils remains disputed and underlying recruitment mechanisms remain to be established. METHODS: Fifteen volunteers received an intravenous bolus of bacter

Experimental human colonisation with non-toxigenic Clostridioides difficile: a placebo-controlled randomised clinical trial.

Clostridioides difficile infections remain a major global healthcare burden, underscoring the need for novel therapies. Human colonisation models provide mechanistic insight into C. difficile colonisation and facilitate identification of novel intervention targets. We conducted a placebo-controlled, randomised clinical trial (NCT05693077) administering non-toxigenic C. difficile (NTCD) capsules to healthy participants to assess safety and colonisation as primary endpoints, and microbiota susceptibility as a secondary endpoint. A total of 69 healthy participants (18-45 years), not previously co

Involvement of impaired carnitine-induced fatty acid oxidation in experimental and human diabetic kidney disease.

Diabetic kidney disease (DKD) is the leading cause of end-stage kidney disease. Kidney tubular cells have a high energy demand, dependent on fatty acid oxidation (FAO). Although carnitine is indispensable for FAO, the pathological role of carnitine deficiency in DKD is not fully understood. We showed here that ectopic lipid accumulation owing to impaired FAO increased in patients with DKD and inversely correlated with kidney function. Organic cation/carnitine transporter 2-deficient (OCTN2-deficient) mice exhibited systemic carnitine deficiency with increased renal lipid accumulation. Cell dea

Pathogenic Features, Experimental Models, and Molecular Tools of Human Fungal Pathogens: Who's on WHO's Radar?

With the prevalence of immunocompromising infections, the emergence of new pathogenic species, rising multidrug resistance, hospital outbreaks, and high mortality rates, once neglected fungal pathogens are now taking over the global healthcare arena, necessitating breakthroughs in diagnostics and therapeutics. The World Health Organization (WHO) recently published a list of critical and high-priority fungal pathogens, underlining the need and significance of broadening the focus to include fungal infections, among other life-threatening infectious diseases. This is also a wake-up call to the s
