Dysregulation of microRNAs in autoimmune diseases: Pathogenesis, biomarkers and potential therapeutic targets.

MicroRNAs (miRNAs) are small, single-stranded, endogenous non-coding RNAs that repress the expression of target genes via post-transcriptional mechanisms. Due to their broad regulatory effects, the precisely regulated, spatial-specific and temporal-specific expression of miRNAs is fundamentally important to various biological processes including the immune homeostasis and normal function of both innate and adaptive immune response. Aberrance of miRNAs is implicated in the development of various human diseases, especially cancers. Increasing evidence has revealed a dysregulated expression patte

Design of miniprotein inhibitors targeting complement C9 to block membrane attack complex assembly.

The abnormal formation of the membrane attack complex (MAC) is intrinsically linked to a range of acute and chronic immune diseases. The insertion of complement C9 into the membrane is the final step and kinetic bottleneck of MAC formation. However, research on blocking the MAC formation of C9 is currently limited. Given its broad, flat, and polar functional interface, complement C9 is a challenging target for rational design. Here, we utilize deep learning-based methods for protein scaffold generation, sequence design, and complex structure prediction to de novo design mini-protein inhibitors

STAT1 and STAT3 mutations: important lessons for clinical immunologists.

The transcription factors signal transducer and activator of transcription (STAT) 1 and STAT3 fulfill fundamental functions in nonimmune and immune cells. The description and follow-up of patients with germline mutations that result in either loss-of-function or gain-of-function have contributed to our understanding of the pathophysiology of these regulators. Depending on the type of mutations, clinical symptoms are complex and can include infection susceptibility, immune dysregulation as well as characteristic nonimmune features. Areas covered: In this review, we provide an overview about mec

Intramuscular iron-dextran and susceptibility of neonates to bacterial infections. In vitro studies.

An increased incidence of E. coli sepsis has been observed in neonates given intramuscular iron-dextran for prevention of iron deficiency. Mechanisms for this apparent effect on susceptibility to infection were investigated by comparing phagocytic and antibacterial functions in paired samples of venous blood from 7 infants, median age 5 days, before and after iron-dextran. Post-treatment sera had increased inhibitory effects on leucocyte chemotaxis and markedly reduced bacteriostatic effects agaainst E. coli. The clinical relevance of the effects on chemotaxis is uncertain. The reduction in se

Ciamexone, a highly selective immunomodulator--a tool for autoimmune diseases?

Both organ-specific diseases such as insulin dependent diabetes mellitus as well as non organ-specific disorders such as rheumatoid arthritis are thought to be autoimmune in origin. Both T-cell and B-cell mediated immune responses are involved in these diseases. More or less specific immunosuppressants are therefore widely used drugs in the treatment of autoimmune diseases which, however, suppress the immune reactions not only against autoantigens but also against foreign antigens. Cyclosporine (Cyclosporin A) has been a tremendous step forward in a more specific direction but it creates probl

Association of different cell types and inflammation in early acne vulgaris.

Acne vulgaris, one of the most common skin diseases, is a chronic cutaneous inflammation of the upper pilosebaceous unit (PSU) with complex pathogenesis. Inflammation plays a central role in the pathogenesis of acne vulgaris. During the inflammatory process, the innate and adaptive immune systems are coordinately activated to induce immune responses. Understanding the infiltration and cytokine secretion of differential cells in acne lesions, especially in the early stages of inflammation, will provide an insight into the pathogenesis of acne. The purpose of this review is to synthesize the ass

Crystal Formation in Inflammation.

The formation and accumulation of crystalline material in tissues is a hallmark of many metabolic and inflammatory conditions. The discovery that the phase transition of physiologically soluble substances to their crystalline forms can be detected by the immune system and activate innate immune pathways has revolutionized our understanding of how crystals cause inflammation. It is now appreciated that crystals are part of the pathogenesis of numerous diseases, including gout, silicosis, asbestosis, and atherosclerosis. In this review we discuss current knowledge of the complex mechanisms of cr

The ocular surface: from physiology to the ocular allergic diseases.

Allergic conjunctivitis (AC) is an inflammation of the conjunctiva secondary to an immune response to exogenous antigens, usually called allergens. In fact, AC is a syndrome that involves the entire ocular surface, including conjunctiva, lids, cornea, and tear film. The signs and symptoms of AC have a meaningful effect on comfort and patient health, and could be influenced by environment, genetics and immune regulation mechanisms, all of which work together in a complex immunological homeostasis. Dysregulation in such immune responses could turn into a variety of ocular allergic diseases (OAD)

Staphylococcus aureus: the multi headed hydra resists and controls human complement response in multiple ways.

The Gram positive human pathogen Staphylococcus aureus causes a spectrum of human diseases including pneumonia, tissue and skin infections, endocarditis, pneumonia and sepsis. The increasing number of resistant bacteria and the threat of methicillin resistant S. aureus (MRSA) urge for the need to develop new antibacterial compounds. A prerequisite for development of such anti microbial compounds is a better understanding of the complex immune crosstalk between the pathogenic bacterium and its human host. To this end proteins staphylococcal proteins that contribute to innate immune evasion espe

WASP: a key immunological multitasker.

The Wiskott-Aldrich syndrome protein (WASP) is an important regulator of the actin cytoskeleton that is required for many haematopoietic and immune cell functions, including effective migration, phagocytosis and immune synapse formation. Loss of WASP activity leads to Wiskott-Aldrich syndrome, an X-linked disease that is associated with defects in a broad range of cellular processes, resulting in complex immunodeficiency, autoimmunity and microthrombocytopenia. Intriguingly, gain of function mutations cause a separate disease that is mainly characterized by neutropenia. Here, we describe recen

Maternal acceptance of the fetus: true human tolerance.

Induction and maintenance of immunologic tolerance in humans remains a desirable but elusive goal. Therefore, understanding the physiologic mechanisms of regulation of immune responses is highly clinically relevant for immune-mediated diseases (e.g., autoimmunity and asthma/allergy) and for cell and organ transplantation. Acceptance of the fetus, which expresses paternally inherited alloantigens, by the mother during pregnancy is a unique example of how the immune system reshapes a destructive alloimmune response to a state of tolerance. Understanding the complex mechanisms of fetomaternal tol

Regulated cell death in autoimmune diseases: molecular mechanisms and therapeutic opportunities.

Autoimmune diseases develop from a breakdown in immune tolerance and arise through complex interactions among genetic susceptibility, environmental factors, immune dysregulation, and tissue damage. Although apoptosis has traditionally been recognized as a central mechanism for maintaining immune homeostasis, increasing evidence has identified several non-classical forms of regulated cell death (RCD) that also contribute to autoimmune disease pathogenesis. These pathways include ferroptosis, pyroptosis, necroptosis, PANoptosis, NETosis, parthanatos, and cuproptosis. Unlike classical apoptosis,

Neuro-immune interactions in tumors, immune-related disorders, and infections.

The nervous and immune systems interact to form a complex network that sustains homeostasis and finely tunes immune responses. These interactions significantly influence the pathogenesis and progression of various diseases, such as tumors, immune-related disorders, and infections. Consequently, these interactions have become promising therapeutic targets. However, insufficient mechanistic research and the lack of candidate drugs have restricted the application of therapeutic strategies targeting neuro-immune interactions. In this review, we summarize three modes of neuro-immune interactions: l

Glucocorticoids-All-Rounders Tackling the Versatile Players of the Immune System.

Glucocorticoids regulate fundamental processes of the human body and control cellular functions such as cell metabolism, growth, differentiation, and apoptosis. Moreover, endogenous glucocorticoids link the endocrine and immune system and ensure the correct function of inflammatory events during tissue repair, regeneration, and pathogen elimination via genomic and rapid non-genomic pathways. Due to their strong immunosuppressive, anti-inflammatory and anti-allergic effects on immune cells, tissues and organs, glucocorticoids significantly improve the quality of life of many patients suffering

Mitochondrial perturbation adapts the proteome from early to advanced immune responses.

Dysregulated immunity, a hallmark of many human diseases, co-occurs with mitochondrial dysfunction and is commonly associated with misprimed primary immune signaling. While transcriptionally well-characterized, the impact of mitochondria on the host response at the protein level is less clear. Using in vitro and in vivo approaches including proteotranscriptomics, our data suggest that OXPHOS promotes expression of early, cell autonomous immune proteins whereas mitochondrial perturbation favors mediators of cell extrinsic responses like inflammation. This response is independent of immune cues,

Immunological Dynamics of Helminth Infections: From Host Defence and Immune Evasion Mechanisms to Vaccine Strategies.

Helminthiasis refers to infections caused by parasitic worms, which represent a significant global health burden, particularly in tropical and subtropical regions. The pathophysiology of helminth infections is highly complex, largely due to the intricate interactions between the host's immune system and the parasite's sophisticated immune evasion strategies. These parasites can modulate host immunity to ensure their long-term survival, often inducing regulatory responses that hinder both natural immunity and vaccine development. However, in recent decades, new vaccine candidates have emerged t

Inflammasomes and adaptive immune responses.

A fundamental concept in immunology is that the innate immune system initiates or instructs downstream adaptive immune responses. Inflammasomes are central players in innate immunity to pathogens, but how inflammasomes shape adaptive immunity is complex and relatively poorly understood. Here we highlight recent work on the interplay between inflammasomes and adaptive immunity. We address how inflammasome-dependent release of cytokines and antigen activates, shapes or even inhibits adaptive immune responses. We consider how distinct tissue or cellular contexts may alter the effects of inflammas

Fundamental mechanistic insights from rare but paradigmatic neuroimmunological diseases.

The pathophysiology of complex neuroimmunological diseases, such as multiple sclerosis and autoimmune encephalitis, remains puzzling - various mechanisms that are difficult to dissect seem to contribute, hampering the understanding of the processes involved. Some rare neuroimmunological diseases are easier to study because their presentation and pathogenesis are more homogeneous. The investigation of these diseases can provide fundamental insights into neuroimmunological pathomechanisms that can in turn be applied to more complex diseases. In this Review, we summarize key mechanistic insights
