In vitro Incubation with Rumen Fluid and Histological Staining of Select Cell Wall Components in Alfalfa (Medicago sativa L.) Stems.

The accumulation of cell wall components as plants grow limits forage nutrition for ruminants. In particular, lignin deposition in plant tissues impedes the breakdown of the cell wall carbohydrates cellulose and hemicellulose, thereby reducing the amount of energy available from forage. The highly digestible pectin component of the cell wall, however, is readily available for degradation. Information about lignin and pectin deposition in different stem cell types may provide mechanistic insights into the factors underlying variation in stem digestibility among alfalfa lines. In vitro incubatio

Tracing Soil CO2 Fluxes under Drying-Rewetting Cycles: Isotopic Insights from an Automatic Soil Incubation System.

Climate change is expected to increase the intensity and frequency of droughts and heavy rainfall events globally, with significant consequences on the terrestrial carbon cycle. One of the most critical yet highly variable components of the carbon cycle is the soil CO2 pulse triggered by precipitation events in dryland ecosystems. To examine the processes underlying the soil CO2 pulse under changing precipitation patterns, we developed a unique Online Automatic Soil Incubation System (OASIS) that allows (1) accurate manipulation of drying and rewetting regimes; (2) continuous monitoring of soi

Differences in Siderophore Production During the Growth of Phytoplankton in Southern Ocean Incubation Experiments Under High- and Low-Iron Conditions.

Iron is an important micronutrient required by marine microorganisms. The Southern Ocean is an ecosystem where primary production is limited by the availability of iron, but how microorganisms in this region adapt to low-iron conditions or respond to episodic iron inputs is understudied. We tested how external iron additions would impact bacterial production of siderophores during phytoplankton growth using shipboard incubation experiments under a range of iron conditions. Similar heterotrophic bacteria communities and phytoplankton biomass were observed whether iron was added from the contine

Mercury fractionation and methylation in Yangtze River Estuary wetland sediments: Insight from thermal decomposition analysis and microcosm incubation.

Understanding mercury (Hg) compounds and methylating microbes in sediments is crucial for evaluating Hg risk in aquatic environments, and simple techniques need to be developed. Wetland sediments from the Yangtze River Estuary (collected in 2019 and 2024) were examined using 4-step thermal decomposition and sequential extraction techniques to determine Hg fractions, and microcosm experiments were performed to evaluate the contributions of microbial communities to methylmercury (MeHg) production. The results indicated that more than 65 % of the total Hg (THg: 39.9-106.5 ng/g) was released at 17

Impact of labile organic carbon and manganese oxide on chromium and vanadium subsurface mobility: evidence from laboratory incubation experiments.

Chromium (Cr) and vanadium(V) are redox-active, geogenic contaminants observed to co-occur in groundwater in the North Carolina (NC) Piedmont region. On a landscape-scale, factors controlling Cr and V solubilization to groundwater in the Piedmont are understood to be largely associated with the regional geology. However, the specific mechanisms mediating (bio)geochemical interactions among heterogeneous geologic materials and redox active chemical inputs in the subsurface are poorly understood. The specific goal of this research was to elucidate the chemical controls on the solubilization of C

Effect of heterozygosity, ploidy and incubation temperature on post-cranial axial skeletal meristics and deformities in Atlantic salmon (Salmo salar).

The teleostean post-cranial axial skeleton is a highly specialized structure for an aquatic mode of life. However, there is limited knowledge regarding parental contributions, early-life environmental impacts on its meristic variation and if reduced heterozygosity challenges its development. To address this, the present study used isogenic homozygous and heterozygous lines of Atlantic salmon (Salmo salar) combined with ploidy manipulation (triploidization) to manipulate parental contributions, and incubation temperature (4 vs. 8°C) as an early-life variable, and reared the fish to ~150 g for a

Investigation of thermal performance of phase change materials-integrated solar yogurt incubation system and its effect on product's characteristics.

AIM: Glomerular involvement in HIV-positive patients is quite heterogeneous. In the present paper we reviewed 73 renal biopsies performed during a period of more than 20 years in a single Nephrology Unit, Milan, Northern Italy, in order to evaluate the aspects of single types of glomerular lesions (including HIV associated nephropathy-HIVAN), grouped according to histological patterns and clinical presentation. Moreover, in the group of non-HIVAN patients, the possible differences in histological characteristics from non-HIV lesions were investigated. MATERIALS AND METHODS: Renal tissues were

Plastic use as nesting material can alter incubation temperature and behaviour but does not affect yellow-legged gull chicks.

Optimal incubation temperature is crucial for embryos' development and survival. With the increasing use of plastics in gulls' nests, it is essential to understand how their incorporation affects incubation temperature, parental behaviour, and hatching success. Considering this, we conducted an experiment where plastic was introduced into yellow-legged gulls (Larus michahellis) nests. The experiment comprised three groups: a control group, a group with low amount of plastic, and a third with a high amount of plastic. This design allowed us to investigate the effects of plastic on 1) the heart

Prolonged incubation of frozen-thawed equine spermatozoa for in vitro fertilization: A preliminary study using low temperature and INRA96 medium.

A protocol for conventional in vitro fertilization (IVF) in horses using fresh semen has been described, using a prolonged incubation in FERT-TALP medium (22 h) at 38.2°C in the presence of penicillamine, hypotaurine and epinephrine (PHE). Our work aimed to develop a protocol that maintains quality parameters in frozen-thawed equine spermatozoa incubated for 22 h in the presence of PHE using different media (FERT-TALP and INRA96) and incubation temperatures (30 and 38.2°C). Twelve frozen ejaculates from four stallions were thawed and then incubated in either FERT-TALP or INRA96 with PHE at 30

Adapting the in vitro rumen incubation method to evaluate the effect of a plant extract on the exsheathment inhibition of Haemonchus contortus infective larvae.

This study adapted the in vitro rumen incubation (IVRI) method to evaluate the biological activity of a Gymnopodium floribundum leaves extract against the exsheathment of Haemonchus contortus infective larvae (L3), and to determine the role of plant polyphenols on the biological activity. The incubation protocol followed the IVRI method, adding polyethylene glycol (PEG) as a polyphenol-blocking agent. The L3 were incubated in ruminal liquor (RL), ruminal liquor with PEG (RL+PEG), ruminal liquor with G. floribundum extract (RLE), and ruminal liquor with G. floribundum extract and PEG (RLE+PEG).

Hybrid Coatings Based on Polyvinylpyrrolidone/Polyethylene Glycol Enriched with Collagen and Hydroxyapatite: Incubation Studies and Evaluation of Mechanical and Physiochemical Properties.

Coating materials offers an intriguing solution for imparting inert implants with additional bioactive characteristics without changing underlying parameters such as mechanical strength. Metallic implants like endoprostheses or polymeric implants can be coated with a thin layer of bioactive film capable of stimulating bone-forming cells to proliferate or release a drug. However, irrespective of the final implantation site of such a coating biomaterial, it is necessary to conduct detailed mechanical and physicochemical in vitro analyses to determine its likely behavior under biological conditio

Effects of lighted incubation and foraging enrichment during rearing on individual fear behavior, corticosterone, and neuroplasticity in laying hen pullets.

Environmental conditions during incubation and rearing can influence stress responsivity of laying hen pullets throughout their lifespan, and therefore have important implications for their welfare. In this study, a 12:12h green LED light-dark cycle during incubation and larvae provisioning as enrichment during rearing were tested as strategies to optimize early-life conditions and thereby decrease stress responsivity in ISA Brown laying hens. A combination of parameters was measured to indicate neuronal, physiological, and behavioral changes that may affect fear and stress. The proteins calbi

Body Size, Cerebral Blood Flow, Ambient Temperature, and Relative Brain Temperatures in Newborn Infants under Incubator Care.

Subtle changes in body temperature affect the outcomes of ill newborns. However, the temperature profile of neonatal brains remains largely unknown. In open-cot care, increased cerebral perfusion is correlated with higher superficial brain temperatures. This study investigated the dependence of brain temperature (relative to rectal temperature) on ambient temperature, body size, cerebral perfusion, and metabolism in infants receiving incubator care. Rectal, scalp, and brain temperatures, superior vena cava flow, and brain oxygenation were assessed using echocardiography, thermo-compensatory te

Comparison of the Direct Identification and Short-Term Incubation Methods for Positive Blood Cultures via MALDI-TOF Mass Spectrometry.

Timely pathogen identification in bloodstream infections is crucial for patient care. A comparison is made between positive blood culture (BC) pellets from serum separator tubes using a direct identification (DI) method and colonies on agar plates from a short-term incubation (STI) method with a matrix-assisted laser desorption/ionization Biotyper for the evaluation of 354 monomicrobial BCs. Both the DI and STI methods exhibited similar identification rates for different types of bacteria, except for Gram-positive and anaerobic bacteria. The DI method's results aligned closely with the STI met
