Bioactivity and laundering resistance of five commercially available, factory-treated permethrin-impregnated fabrics for the prevention of mosquito-borne diseases: the need for a standardized testing and licensing procedure.

Personal protective measures against hematophagous vectors constitute the first line of defense against arthropod-borne diseases. In this regard, a major advance has been the development of residual insecticides that can be impregnated into clothing. Currently, however, information on specific treatment procedures, initial insecticide concentrations, arthropod toxicity, residual activity, and laundering resistance is either fragmentary or non-existent, and no World Health Organization Pesticides Evaluation Scheme or other guidelines exist for the standardized testing and licensing of insectici

Increase in insecticide susceptibility after sublethal exposure to deltamethrin in the German cockroach (Blattodea: Ectobiidae).

The German cockroach, Blattella germanica (L.), is a major urban insect pest primarily controlled by insecticides, including pyrethroid sprays, fipronil baits, and indoxacarb baits. However, widespread pyrethroid resistance increases the probability that cockroaches will survive treatments in the field. Many insecticides are applied concurrently or repetitively on a periodic basis, meaning survivors have the chance to be re-treated, but any lasting physiological effects from initial survival may impact the performance of subsequent treatments. We investigated the effects of sublethal deltameth

Molecular and kinetic characterization of two acetylcholinesterases with particular focus on the roles of two amino acid substitutions (Y390N and F392W) in Bemisia tabaci.

Two acetylcholinesterases (AChEs) are present in Bemisia tabaci (BtAChE1 and BtAChE2). A conserved AChE mutation conferring organophosphate (OP) resistance (F392W in BtAChE1) is saturated in field populations despite its potential fitness cost, and a highly conserved amino acid residue forming the backdoor of AChE is substituted with a unique amino acid (Y390N in BtAChE1) in B. tabaci. Thus, the roles and relationships of the two amino acid substitutions in the evolutionary adaptation of B. tabaci remain to be elucidated, and little information is available on the catalytic and molecular prope

Inheritance patterns, cross-resistance and synergism in Spodoptera frugiperda (Lepidoptera: Noctuidae) resistant to emamectin benzoate.

BACKGROUND: Fall armyworm, Spodoptera frugiperda (J. E. Smith), is a relevant global pest due to severe damage caused on agricultural crops and its capacity to evolve resistance to insecticides. Here, we selected a strain of S. frugiperda resistant to emamectin benzoate under laboratory conditions to understand the inheritance patterns, cross-resistance and synergism involved in the resistance. RESULTS: The emamectin benzoate-resistant (Ben-R) strain was isolated by using F2 screen in a field population collected in Lucas do Rio Verde, Mato Grasso state, Brazil. After ten generations of selec

Sublethal and transgenerational effects of spinetoram on the biological traits of Plutella xylostella (L.) (Lepidoptera: Plutellidae).

Spinetoram, a spinosyn insecticide is used to manage lepidopteran pests, including diamondback moth, Plutella xylostella. In addition to determining the lethal effects, identifying low and/or sublethal effects of an insecticide is crucial to understanding the total impact of an insecticide. We assessed the low lethal and sublethal effects of spinetoram on two successive generations of P. xylostella. The initial bioassay results showed that spinetoram exhibited high toxicity against P. xylostella with an LC50 of 0.114 mg L-1 after 48 h exposure. The low lethal (LC25) and sublethal (LC10) concen

Intraspecific Transcriptome Variation and Sex-Biased Expression in Anopheles arabiensis.

The magnitude and functional patterns of intraspecific transcriptional variation in the anophelines, including those of sex-biased genes underlying sex-specific traits relevant for malaria transmission, remain understudied. As a result, how changes in expression levels drive adaptation in these species is poorly understood. We sequenced the female, male, and larval transcriptomes of three populations of Anopheles arabiensis from Burkina Faso. One-third of the genes were differentially expressed between populations, often involving insecticide resistance-related genes in a sample type-specific …

Resistance of Spodoptera frugiperda (Lepidoptera: Noctuidae) to spinetoram: inheritance and cross-resistance to spinosad.

BACKGROUND: The use of spinosyn insecticides is one of the major control strategies of the fall armyworm, Spodoptera frugiperda (J. E. Smith) in Brazil. In this study, we selected a spinetoram-resistant strain from a field-population of S. frugiperda to characterize the inheritance of the resistance and cross-resistance relationship between spinosyn insecticides. RESULTS: The values of LC50 (95% CI) obtained from concentration-response bioassays were 0.63 (0.55-0.73) μg spinetoram mL-1 for the susceptible strain (SUS), and 1170.96 (1041.61-1323.89) μg spinetoram mL-1 for the strain resistant

Rhamnolipids: solution against Aedes aegypti?

Aedes aegypti mosquitoes are the primary transmitters of dengue fever, urban yellow fever, and chikungunya viruses. This mosquito has developed resistance to the insecticides currently used to control their populations. These chemical insecticides are harmful to the environment and can have negative effects on human health. Rhamnolipids are environmentally compatible biological surfactants, but their insecticidal activity has not been extensively studied. The present study evaluated the potential larvicidal, insecticidal, and repellent activities of rhamnolipids against A. aegypti. At concentr

Diversity of culturable gut bacteria and their role in conferring resistance to alpha-cypermethrin in field populations of Stegomyia aegypti.

INTRODUCTION: Control of Stegomyia aegypti (St. aegypti), a major vector of arboviruses, has been increasingly challenged by insecticide resistance, driven by genetic, metabolic, behavioral, and environmental factors. While extensively studied in other contexts, the role of gut microbiota in insecticide resistance remains largely unexplored. Therefore, this study assessed the α-cypermethrin susceptibility status of St. aegypti from Puducherry by characterizing the culturable gut bacteria from resistant and susceptible mosquito populations and evaluating their role in mediating resistance. MAT…

Structural basis for diamide modulation of ryanodine receptor.

The diamide insecticide class is one of the top-selling insecticides globally. They are used to control a wide range of pests by targeting their ryanodine receptors (RyRs). Here, we report the highest-resolution cryo-electron microscopy (cryo-EM) structure of RyR1 in the open state, in complex with the anthranilic diamide chlorantraniliprole (CHL). The 3.2-Å local resolution map facilitates unambiguous assignment of the CHL binding site. The molecule induces a conformational change by affecting the S4-S5 linker, triggering channel opening. The binding site is further corroborated by mutagenesi

Analysis of the Knockdown Resistance Locus (kdr) in Anopheles stephensi, An. arabiensis, and Culex pipiens s.l. for Insight Into the Evolution of Target-site Pyrethroid Resistance in Eastern Ethiopia.

The malaria vector, Anopheles stephensi, which is typically restricted to South Asia and the Middle East, was recently detected in the Horn of Africa. Addressing the spread of this vector could involve integrated vector control that considers the status of insecticide resistance of multiple vector species in the region. Previous reports indicate that the knockdown resistance mutations (kdr) in the voltage-gated sodium channel (vgsc) are absent in both pyrethroid-resistant and pyrethroid-sensitive An. stephensi in eastern Ethiopia; however, similar information about other vector species in the …

Reversion of CYP450 monooxygenase-mediated acetamiprid larval resistance in dengue fever mosquito, Aedes aegypti L.

Aedes-borne diseases are on the rampant rise despite continued application of chemical insecticide-based interventions. The appearance of high degree of insecticide resistance in Aedes species and noxious effects on environment and non-targets have raised further concerns. Among new chemical interventions, neonicotinoids are considered a safe and effective approach. The present study investigated the control potency of acetamiprid and development of resistance in Aedes aegypti larvae; and the involvement of CYP450 monooxygenases in inducing resistance. The early fourth instars of Ae. aegypti p…

Identification of QTLs Conferring Resistance to Deltamethrin in Culex pipiens pallens.

Culex pipiens pallens is the most abundant Culex mosquito species in northern China and is an important vector of bancroftian filariasis and West Nile virus. Deltamethrin is an insecticide that is widely used for mosquito control, however resistance to this and other insecticides has become a major challenge in the control of vector-borne diseases that appear to be inherited quantitatively. Furthermore, the genetic basis of insecticide resistance remains poorly understood. In this study, quantitative trait loci (QTL) mapping of resistance to deltamethrin was conducted in F2 intercross segregat…
