The aggressive nature of metastatic human cancer has been shown to be related to numerous abnormalities in growth factors and their receptors. These perturbations confer a tremendous growth advantage to the malignant cells. Interleukin-8 (IL-8), originally discovered as a chemotactic factor for leukocytes, has recently been shown to contribute to human cancer progression through its potential functions as a mitogenic, angiogenic, and motogenic factor. While it is constitutively detected in human cancer tissues and established cell lines, IL-8 expression is regulated by various tumor microenvir

Established genetic causes of familial Alzheimer disease (AD) involve genes for beta-amyloid precursor protein (betaAPP), presenilin-1, and presenilin-2. For the more common sporadic forms of AD, increased risk has been associated with a number of genes; the most important of which is the epsilon4 allele of apolipoprotein E. Two recent studies, one clinical and one using postmortem material, now show increased risk for AD associated with certain polymorphisms in the genes encoding the alpha and beta isoforms of interleukin-1 (IL-1). IL-1 levels are elevated in Alzheimer brain, and overexpressi

Interleukin-8 (IL-8), a cytokine produced by a host of cells including monocytes, macrophages, Kupffer cells and hepatocytes, can activate neutrophils. Peripheral neutrophilia and liver neutrophil infiltration are frequently noted in patients with alcoholic liver disease (ALD). Serum IL-8 levels are markedly elevated in patients with alcoholic hepatitis compared with those in patients with alcoholic cirrhosis, alcoholic fatty liver and non-alcoholic liver disease. The levels are also elevated in patients who die of hepatic failure and correlate with biochemical and histologic parameters and se

The phenotype of cultured fibroblasts from patients affected by Apert's syndrome, a rare connective disorder, differs from that of normal cells in its extracellular matrix macromolecule composition (glycosaminoglycans, collagens and fibronectin) and is further modulated by treatment with interleukins (ILs). As the mechanisms responsible for the changes are unknown, we used our recently described model system for Apert periosteal fibroblasts to ascertain whether the pattern of ILs they secrete into the medium is comparable to that of normal fibroblasts. The results obtained by enzyme-linked imm…

Interleukin-6 (IL-6) is a multifunctional cytokine that plays a central role in host defense due to its wide range of immune and hematopoietic activities and its potent ability to induce the acute phase response. Overexpression of IL-6 has been implicated in the pathology of a number of diseases including multiple myeloma, rheumatoid arthritis, Castleman's disease, psoriasis, and post-menopausal osteoporosis. Hence, selective antagonists of IL-6 action may offer therapeutic benefits. IL-6 is a member of the family of cytokines that includes interleukin-11, leukemia inhibitory factor, oncostati

In recent years, it has become apparent that IL-7, originally characterized as a growth factor for pre-B lymphocytes, also has important implications for the skin. Keratinocytes have been shown to produce IL-7. which in turn can elicit a variety of biological responses on several cell types residing in the skin. IL-7 has been demonstrated to augment the cytolytic activity of cytotoxic T cells (CTL) and natural killer (NK) cells against various neoplastic targets including melanoma cells. Proliferation and long-term survival of murine dendritic epidermal T lymphocytes (DETC) in vitro is support

OBJECTIVE: To examine the role of cytokines in cervical dilation.
METHODS: In 55 patients undergoing cesarean delivery, we took samples from the lower uterine segment, the decidua, and the membranes. We determined the concentrations of interleukin (IL)-2, IL-8, tumor necrosis factor-alpha, matrix metalloproteinase (MMP)-8, and MMP-9 in the different tissues.
RESULTS: Depending on the state of labor, we observed a significant increase (P < .001) in IL-8 concentrations in the lower uterine segment. The leukocyte enzymes MMP-8 and MMP-9 were highly significantly correlated with the IL-8 concent

Involvement of the cytokines in male infertility were considered from in vitro studies, however the nature and the role of the mediator susceptible of arising in vivo is not completely elucidated. The purpose of this study is to measure the concentrations of interleukin-2 (IL-2) in normal and pathologic seminal fluid and to analyse their relationship with semen parameters. Our results demonstrate the presence of IL-2 in seminal fluid. Comparatively to normal seminal fluid, significant lower IL-2 concentrations were observed in infected semen samples or with sperm antibodies. No correlation was

This review discusses the effects of interleukin-1 (IL-1) on bone metabolism and the evidence implicating IL-1 in periodontal disease. IL-1 is a potent cytokine with significant effects on the metabolism of many tissues, including bone. Several in vitro and in vivo studies have established the potent bone-resorbing effect of IL-1. Studies have indicated that IL-1 inhibits bone formation, although it can stimulate formation under certain conditions. Osteoblastic cells are the bone cells expressing IL-1 receptors and responding to IL-1. IL-1 effects on osteoblastic cells include stimulation of p

Interleukin-2 (IL-2) is a lymphokine produced by activated T cells and its receptor (IL-2R) is expressed on T cells; one of the IL-2R components can be measured as a soluble serum protein (sIL-2R). Levels of sIL-2R were measured as a sign of T cell activation in serum of 12 patients suffering from alopecia areata (totalis or universalis) and in a group of healthy control subjects. An enzyme-linked immunosorbent assay was used to determine the levels of sIL-2R in blood samples drawn during both the active and resting phase of the disease. In patients with alopecia areata in active phase the sIL

We have developed a bioassay for human IL-4 based upon its ability to upregulate CD23 (low affinity IgE receptor) expression. Ramos, a B lymphocyte line derived from a Burkitt lymphoma, was repetitively subcloned yielding a clone, Ramos.G6.C10, which is several fold more sensitive to this effect of IL-4. In microtiter plates cells were cultured for 48 h in the presence of dilutions of recombinant human IL-4 or samples, and then stained with murine anti-human CD23 and goat anti-mouse IgG-FITC. IL-4 induced an eight-fold increase (60 channel shift) in fluorescence intensity as measured by flow c

Forty-seven patients with renal carcinoma were included in first line or rescue protocols of immunotherapy including IL2 alone or in association with LAK cells, INF alpha or TNF. The toxicity was mild and the mortality was 2% (1 patient). The response rate was 26%. Nineteen children with neuroblastoma received IL2 either alone or in combination with LAK cells. The morbidity and mortality were higher in patients with end stage disease who had previously received high dose and prolonged chemotherapy. In contrast, the toxicity was mild and transient in patients treated in the months following aut

The majority of peripheral blood mononuclear cells (PBMC) from acute-phase infectious mononucleosis (IM) patients express high levels of T10 (CD38) and HLA-DR surface antigens, which are markers characteristic of activated T cells. However, Tac antigen (p55) expression on these cells was not detectable by flow cytometric immunofluorescence, and only a low level of specific interleukin-2 (IL-2) binding was found by Scatchard analysis. These results suggest that IL-2 receptors (IL-2R) are lost or down-regulated on activated T cells in acute IM. Since a large proportion of T cells die during the

IL-1 induces PMN and monocyte infiltration when injected into rabbit knee joints (1). This transient synovitis is associated with depletion of proteoglycan from the articular cartilage matrix. However, it is not clear whether the cartilage proteoglycan depletion is dependent on the infiltrating leucocytes. Consequently, we have investigated the effect of intra-articular injection of interleukin-1 in rabbits depleted of circulating PMN and monocytes with nitrogen mustard.

Augmentation of mitogen-induced release of the T cell lymphokine interleukin-2 (IL-2) occurred using several cytochalasins in coculture with a T cell lymphoma line (JM) or with purified T cells. When added concurrently with mitogen, cytochalasins had no apparent negative effect either on the ability of mitogen to signal IL-2 production or on the total amount produced. Use of cytochalasins that affect submembranous actin in peripheral lymphocytes established a link between cytoskeletal alterations and lymphokine release, although direct membrane perturbation cannot be excluded. These data indic

The synthesis of interleukin-2 (IL-2) has been determined in 5 patients suffering from pemphigus vulgaris. It is well-known that these lymphokine play an important role with regard to the stimulation of lymphocytes. All 5 patients were treated with auric sodium thiomalate (Tauredon) according to Penney. Therapy with corticosteroids had to be reduced or even interrupted because of their numerous side effects. In comparison to the control group, all 5 patients showed decreased synthesis of interleukin-2. Total T-, T-helper, and T-suppressor lymphocytes were measured by immunofluorescence with mo
