Neuroprotective effects of Aucubin against cerebral ischemia-reperfusion injury.

AIMS: To study the role of Aucubin (AU) in cerebral ischemia-reperfusion injury and investigate the potential mechanisms. METHODS: For the in vitro experiment, primary microglia were cultured and stimulated by Lipopolysaccharides (LPS) and treated with AU. Male C57/BL6J mice were used and middle cerebral artery occlusion (MCAO) model was performed to induce cerebral ischemia-reperfusion injury. For the short-term effects, mice administrated with AU (40 mg/kg) for 3 days after MCAO were evaluated for the infarct volume and neurological deficits. The neuroinflammatory factors and microglia acti

Renewable Electroconductive Hydrogels for Accelerated Diabetic Wound Healing and Motion Monitoring.

Diabetic foot ulcers (DFUs), a prevalent complication of diabetes mellitus, may result in an amputation. Natural and renewable hydrogels are desirable materials for DFU dressings due to their outstanding biosafety and degradability. However, most hydrogels are usually only used for wound repair and cannot be employed to monitor motion because of their inherent poor mechanical properties and electrical conductivity. Given that proper wound stretching is beneficial for wound healing, the development of natural hydrogel patches integrated with wound repair properties and motion monitoring was exp

One-touch embeddable microneedles for hair loss treatment.

With increasing clinical demands for painless and easy administration of medications, such as for hair loss, microneedles (MNs) have been widely exploited for facilitating drug permeation in a minimally invasive manner. However, precise dose control and long-term drug delivery without the infection risk through punctured holes have remained unresolved. Herein, we developed swellable microneedles (MNs) with an air-pocket structure, enabling shear-induced implantation inside the skin. The air-pocket MNs (AP-MNs) were prepared by one-step molding process with genipin-crosslinked gelatin solutions

Evaluation of the Antidepressant Activity, Hepatotoxicity and Blood Brain Barrier Permeability of Methyl Genipin.

Geniposide (GE) is the main bioactive component of Gardeniae Fructus. The hepatotoxicity of geniposide limited clinical application. In order to get a new geniposide derivative that has less hepatotoxicity and still possesses the antidepressant activity, a new C-1 hydroxyl methylation derivative named methyl genipin (MG) was synthesized from geniposide. In the present study, we demonstrated that MG did not increase the liver index, alanine aminotransferase (ALT) and aspirate aminotransferase (AST). Histopathological examination suggested that no toxic damages were observed in rats treated oral

Restoration of Opa1-long isoform inhibits retinal injury-induced neurodegeneration.

Optic atrophy 1 (Opa1) is a critical factor that regulates fusion and other important functions of mitochondria. In mitochondrion, the N-terminal mitochondrial targeting sequence of Opa1 precursors is removed to generate Opa1 long isoforms (L-Opa1), which are further cleaved into short isoforms (S-Opa1). In the present study, we found that retinal ischemia-reperfusion (I/R) injury and intravitreal injection of carbonylcyanide m-chlorophenyl hydrazone (CCCP) both dramatically induced Opa1 cleavage and caused loss of L-Opa1. In cultured neuronal cells under hypoxia-reoxygenation (H/R) injury, si

Unveiling the pharmacological landscape of geniposidic acid: mechanisms of action, therapeutic benefits, and scientific challenges.

Geniposidic acid (GPA) is a bioactive iridoid glycoside mainly found in many traditional medicinal plants, including Eucommia ulmoides and Gardenia jasminoides. These plants are also commonly used in traditional diets and herbal teas throughout East Asia, particularly in China and Japan, where they are used to maintain well-being and protect the respiratory, cardiovascular, and renal systems. Despite the growing number of individual studies that highlight GPA's therapeutic potential, to date, there has been a lack of a systematic, critical synthesis of current knowledge. The aim of this review

Deacetylasperulosidic acid methyl ester attenuates liver fibrosis by modulating the SCFAs-Wnt/β-catenin signaling axis.

Liver fibrosis is a common pathological consequence of chronic liver injury, characterized by excessive extracellular matrix deposition, hepatocellular damage and persistent inflammation, which may progress to cirrhosis or hepatocellular carcinoma. Currently, no specific and effective antifibrotic therapies are available. Deacetylasperulosidic acid methyl ester (DAM), a natural iridoid compound derived from Rubiaceae plants, possesses anti-inflammatory and antioxidant properties. However, its antifibrotic effects and underlying mechanisms remain poorly defined. In this study, a CCl4-induced mo
