Septins are a conserved family of GTPases that regulate important cellular processes such as cell wall integrity, and septation in fungi. The requirement of septins for virulence has been demonstrated in the human pathogenic yeasts Candida albicans and Cryptococcus neoformans, as well as the plant pathogen Magnaporthe oryzae. Aspergillus spp. contains five genes encoding for septins (aspA-E). While the importance of septins AspA, AspB, AspC, and AspE for growth and conidiation has been elucidated in the filamentous fungal model Aspergillus nidulans, nothing is known on the role of septins in g

Arundina graminifolia is an early successional plant on Iriomote Island, the Ryukyus, Japan, where it is endangered. Populations flower for more than half a year, and many inflorescences bloom for one to several months. The nectarless gullet flowers, which open for up to six days, are self-compatible but cannot self-pollinate spontaneously; thus they rely on pollinating agents for capsule production. Field observations at two habitats identified at least six species of bees and wasps, primarily mate-seeking males of Megachile yaeyamaensis and Thyreus takaonis, as legitimate pollinators. Thus,

Outer membrane vesicles (OMVs) have been identified in a wide range of bacteria, yet little is known of their biogenesis. It has been proposed that OMVs can act as long-range toxin delivery vectors and as a novel stress response. We have found that the formation of OMVs in the gram-negative opportunistic pathogen Serratia marcescens is thermoregulated, with a significant amount of OMVs produced at 22 or 30°C and negligible quantities formed at 37°C under laboratory conditions. Inactivation of the synthesis of the enterobacterial common antigen (ECA) resulted in a hypervesiculation phenotype, s

The authors describe an accident leading to penetration of the eye of an 11-year-old boy by an unusual foreign body, and the subsequent diagnosis and treatment. Persistent ocular irritation following a corneal perforation wound, in spite of appropriate treatment, led to the presence of an intraocular foreign body being suspected. The results of radiologic examination, including CT (Siemens Somatom 2), were negative. Only A-scan sonography (Kretztechnik 7002) called attention to the possibility of a foreign body in the vitreous behind the traumatic cataract. Extracapsular extraction was immedia

The gut microbiota forms a competitive biological barrier against enteric pathogens and may also modulate antiviral immunity and disease prevention. This review presents recent advances demonstrating the tripartite model of the gut microbiome-immunity-virus axis in lepidopteran insects. Emerging evidence indicates that their gut microbiota regulates antiviral immunity through context-dependent mechanisms shaped by host species identity, microbial community composition, and strain-specific differences in viral resistance. Rather than acting as a uniformly protective factor, gut microbes fine-tu

Understanding the biochemical impact of nontarget clinical drugs on insect models provides critical insights into xenobiotic toxicity. In this study, we investigated the biochemical effects of dietary AZR on the hemolymph of seventh-instar Galleria mellonella (Lepidoptera: Pyralidae) larvae. First-instar larvae of the insect were reared under laboratory conditions until the seventh-instar larvae. The larvae were fed an artificial diet supplemented with azithromycin (AZR) at different concentrations: 0.0012, 0.0072, 0.0432, 0.2592, 0.7776, and 1.0368%. Our results demonstrated that dietary AZR

Although herbs can contain several secondary metabolites potentially harmful to insects, dried herbs, like all stored foods, can be damaged by various insect pests. These pests have the potential to diminish both the quantity and the quality of food, and the consequences of infestation often only become noticeable once the pest has already become established. This study investigates the ability of two Lepidoptera pests, one polyphagous, Plodia interpunctella, and one selective, Idaea inquinata, to complete the postembryonic development on eleven dried herbs commonly used in cooking: dill (Anet

Insects' body coloration may be indirectly influenced by their host plants. Perina nuda (Lepidoptera: Lymantriidae), commonly known as the Banyan Tussock Moth and a serious pest of banyan trees (Ficus spp.) in southern China, exhibits light body coloration during its first- to third-instar stages, with its coloration progressively darkening as it matures, but little is known of the relationship between larval body coloration and host plants. To address this gap, we examined the R (red), G (green), B (blue), and L (lightness) values of the head, dorsal thorax and abdomen, stripe, dorsal mid-lin

The Homeotic complex (Hox) genes play a crucial role in determining segment identity and appendage morphology in bilaterian animals along the antero-posterior axis. Recent studies have expanded to agricultural pests such as fall armyworm (FAW), scientifically known as Spodoptera frugiperda J. E. Smith (Lepidoptera: Noctuidae), which significantly threatens global agricultural productivity. However, the specific role of the hox gene Sfabd-B in FAW remains unexplored. This research investigates the spatial and temporal expression patterns of Sfabd-B in various tissues at different developmental

The specificity of woody plant defense responses to different attacking herbivores is poorly known. We investigated the responses of black poplar (Populus nigra) to leaf feeding by three lepidopteran species (Lymantria dispar, Laothoe populi and Amata mogadorensis) and two leaf beetle species (Phratora vulgatissima and Chrysomela populi). Of the direct defenses monitored, increases in trypsin protease inhibitor activity and the salicinoid salicin were triggered by herbivore damage, but this was not herbivore-specific. Moreover, the majority of leaf salicinoid content was present constitutively

We analyzed the antifungal susceptibility profiles, genotypes, and virulence of clinical Aspergillus terreus isolates from six university hospitals in South Korea. Thirty one isolates of A. terreus, comprising 15 respiratory and 16 ear isolates were assessed. Microsatellite genotyping was performed, and genetic similarity was assessed by calculating the Jaccard index. Virulence was evaluated by Galleria mellonella survival assay. All 31 isolates were susceptible to itraconazole, posaconazole, and voriconazole, while 23 (74.2%) and 6 (19.4%) showed amphotericin B (AMB) minimum inhibitory concen

OBJECTIVES: Over the last 30 years, the number of invasive fungal infections among immunosuppressed patients has increased significantly, while the number of effective systemic antifungal drugs remains low. The aim of this study was to identify and characterize antifungal compounds that inhibit fungus-specific metabolic pathways not conserved in humans.
METHODS: We screened a diverse compound library for antifungal activity in the pathogenic mould Aspergillus fumigatus . We determined the in vitro activity of bromoquinol by MIC determination against a panel of fungi, bacteria and cell lines.

The study of the host-pathogen interaction is essential to understand the mechanisms underlying adhesion, colonization and tissue damage by pathogens. This is usually achieved by performing in vivo studies using small mammals, such as rats, mice and guinea pigs. Nowadays, the mouse models of systemic or subcutaneous infection are the gold standard assays to analyze the virulence of members of the Sporothrix schenckii complex. There are, however, invertebrates that have been recently used as alternative hosts to assess the virulence of both bacteria and fungi, and among them, larvae of Galleria

PURPOSE: Hairs from the caterpillar of the pine processionary moth, Thaumetopoea pityocampa, located in the pine forests of southern Europe and elsewhere, can become embedded in the cornea and conjunctiva. Disorders produced by the urticating hairs can be classified into a broad spectrum of severity from mild keratoconjunctivitis to anaphylactic shock. This report presents 3 cases that illustrate the range of corneal responses to embedded hairs of the processionary moth caterpillar.
METHODS: Case series and review of the literature.
RESULTS: A 51-year-old man (case 1), a 46-year-old woman (c

We measured structural and chemical traits of the leaves of native, broad-leaved trees in two temperate localities [southern Ontario, Canada (34 species), and Missouri (36 species)] and one tropical locality [central Panama (samples of 21 and 23 species)] to test the hypothesis that the greater diversity of tree species and herbivore species in the tropics is associated with greater resource niche space for herbivores. Variables were leaf toughness, water content, dry mass per unit area, several structural and nutritional carbohydrates, common mineral elements, including nitrogen and phosphoru

BACKGROUND: Candida can cause mucocutaneous and/or systemic infections in hospitalized and immunosuppressed patients. Most individuals are colonized by Candida spp. as part of the oral flora and the intestinal tract. We compared oral and systemic isolates for the capacity to form biofilm in an in vitro biofilm model and pathogenicity in the Galleria mellonella infection model. The oral Candida strains were isolated from the HIV patients and included species of C. albicans, C. glabrata, C. tropicalis, C. parapsilosis, C. krusei, C. norvegensis, and C. dubliniensis. The systemic strains were iso

Pandoraea species have emerged as opportunistic pathogens among cystic fibrosis (CF) and non-CF patients. Pandoraea pulmonicola is the predominant Pandoraea species among Irish CF patients. The objective of this study was to investigate the pathogenicity and potential mechanisms of virulence of Irish P. pulmonicola isolates and strains from other Pandoraea species. Three patients from whom the P. pulmonicola isolates were isolated have since died. The in vivo virulence of these and other Pandoraea strains was examined by determining the ability to kill Galleria mellonella larvae. The P. pulmon

Essential aspects of the innate immune response to microbial infection are conserved between insects and mammals. This has generated interest in using insects as model organisms to study host-microbe interactions. We used the greater wax moth Galleria mellonella, which can be reared at 37 degrees C, as a model host for examining the virulence potential of Listeria spp. Here we report that Galleria is an excellent surrogate model of listerial septic infection, capable of clearly distinguishing between pathogenic and nonpathogenic Listeria strains and even between virulent and attenuated Listeri

Acetylcholine modulates the virulence of Candidaalbicans and regulates an appropriate immune response to infection in a Galleria mellonella infection model. Indeed, the evidence suggests that C. albicans possesses a functional cholinergic receptor that can regulate filamentous growth and biofilm formation. Furthermore, G. mellonella immune cell subsets possess repertories of cholinergic receptors which regulate an effective and appropriate cellular immune response to C. albicans infection. This study aimed to investigate the cholinergic receptor subtype involved in regulation of filamentous gr

Myocilin (MYOC, TIGR) variations are associated with juvenile and primary open angle glaucoma (POAG). To investigate consequences of MYOC wildtype overexpression and selected mutations, we established a heterologous insect cell system (High Five). Wildtype, Pro370Leu, Gln368X and Lys423Glu were cloned into a modified pIB/V5-His (pEXIV) vector with and without downstream GFP in frame fusion. Mutations were introduced by in vitro mutagenesis. Heterologous expression was shown and analysed by RT-PCR, Western blotting, immunocytochemistry and fluorescence microscopy. Extended cultivation (>14 days
