Metabolic basis of quality deterioration in soybean variety Heihe 43 driven by storage time: Based on non-targeted metabolomics.

Soybeans are a high-quality plant-based food with both nutritional value and physiological activity, widely used in various fields such as food and feed. The quality evolution during soybean storage is a complex biological process regulated by multiple factors, but the mechanism of quality deterioration during long-term soybean storage is not yet fully understood. To reveal the underlying mechanism of soybean deterioration, this study constructed an accelerated storage simulation system under normal temperature and humidity conditions (20 °C, 65% RH) for 0-3 years, and explored the changes in

Comparison of the skin sensitizing potential of unsaturated compounds as assessed by the murine local lymph node assay (LLNA) and the guinea pig maximization test (GPMT).

The skin sensitization potential of eight unsaturated and one saturated lipid (bio)chemicals was tested in both the LLNA and the GPMT to address the hypothesis that chemicals with unsaturated carbon-carbon double bonds may result in a higher number of unspecific (false positive) results in the LLNA compared to the GPMT. Seven substances (oleic acid, linoleic acid, linolenic acid, undecylenic acid, maleic acid, squalene and octinol) gave clear positive results in the LLNA (stimulation index (SI)> or = 3) and thus would require labelling as skin sensitizer. Fumaric acid and succinic acid gave cl

Dynamic changes of serum metabolites associated with infection and severity of patients with acute hepatitis E infection.

Dynamic changes in metabolites may affect liver disease progression, and provide new methods for predicting liver damage. We used ultra-performance liquid chromatography-mass spectroscopy to assess serum metabolites in healthy controls (HC), and patients with acute hepatitis E (AHE) or hepatitis E virus acute liver failure (HEV-ALF). The principal component analysis, partial least squares discriminant analysis, and discriminant analysis of orthogonal projections to latent structures models illustrated significant differences in the metabolite components between AHE patients and HCs, or between

Natural CLA-Enriched Lamb Meat Fat Modifies Tissue Fatty Acid Profile and Increases n-3 HUFA Score in Obese Zucker Rats.

Ruminant fats are characterized by different levels of conjugated linoleic acid (CLA) and α-linolenic acid (18:3n-3, ALA), according to animal diet. Tissue fatty acids and their N-acylethanolamides were analyzed in male obese Zucker rats fed diets containing lamb meat fat with different fatty acid profiles: (A) enriched in CLA; (B) enriched in ALA and low in CLA; (C) low in ALA and CLA; and one containing a mixture of olive and corn oils: (D) high in linoleic acid (18:2n-6, LA) and ALA, in order to evaluate early lipid metabolism markers. No changes in body and liver weights were observed. CLA

Effect of dietary columbinic acid on the fatty acid composition and physical membrane properties of different tissues of EFA-deficient rats.

The effect of columbinic acid (5 trans, 9 cis, 12 cis, octadeca-trienoic acid) supplemented to a fat-free diet on the fatty acid composition and its correlation to the physical properties of several tissues of rats, was studied. The absence of lipids in the diet produced the typical changes in the fatty acid composition characteristic of essential fatty acid (EFA) deficiency, namely a significant increase in the relative percentage of monoenoic fatty acids with a concomitant decrease in linoleic and arachidonic acids and a rise in eicosa-5,8,11-trienoic acid in liver, kidney, lung and spleen h

Fatty acid composition of serum lipids of mothers and their babies after normal and hypertensive pregnancies.

The biochemical essential fatty acid (EFA) status of neonates born after normal and hypertensive pregnancies (PIH) and that of their mothers was assessed by measuring the fatty acid composition of phospholipids (PL), triglycerides (TG) and cholesterol esters (CE) of umbilical cord serum and maternal serum, respectively. Relative contents of linoleic acid of serum PL and CE were significantly lower in mothers with PIH compared to normal pregnancies. Most other (n-6) polyenes in PL tended to be higher under hypertensive conditions. Total maternal (n-3) polyenes of serum PL were significantly hig

A combined transcriptomics and lipidomics analysis of subcutaneous, epididymal and mesenteric adipose tissue reveals marked functional differences.

Depot-dependent differences in adipose tissue physiology may reflect specialized functions and local interactions between adipocytes and surrounding tissues. We combined time-resolved microarray analyses of mesenteric- (MWAT), subcutaneous- (SWAT) and epididymal adipose tissue (EWAT) during high-fat feeding of male transgenic ApoE3Leiden mice with histology, targeted lipidomics and biochemical analyses of metabolic pathways to identify differentially regulated processes and site-specific functions. EWAT was found to exhibit physiological zonation. De novo lipogenesis in fat proximal to epididy

Dynamics of fatty-acid composition of neutral acylglycerols in maturing euonymus fruits.

The dynamics of the fatty-acid (FA) composition of neutral acylglycerols (NAGs) composed of 1,2,3-triacyl-sn-glycerols (TAGs) and 3-acetyl-1,2-diacyl-sn-glycerols (acDAGs) was determined in the fruit seeds and arils of three Euonymus L. species at three stages of their maturity. The NAG composition comprised 29 FAs, linoleic, oleic, palmitic, and α-linolenic acids being predominant. Noticeable amounts of other FAs, such as lauric, myristic, hexadec-9-enoic, stearic, (Z)-vaccenic, and arachidic acid, etc., could also be present. In the course of maturation, the qualitative composition of major

Plasma concentrations of estradiol 17beta and PGF2alpha metabolite and placental fatty acid composition and antioxidant enzyme activity in cows with and without retained fetal membranes.

Fetal membrane retention is one of the most common problems in Holstein cattle after parturition. To investigate mechanisms involved, the following parameters were studied in the peri-parturition period: plasmatic concentrations of estradiol-17beta (E2) and PGFM (PGF2alpha metabolite), activity of antioxidant enzymes (superoxide dismutase-SOD, catalase-CAT and glutathione peroxidase-GSH-Px), thiobarbituric acid reagent substances (TBAR) concentrations and fatty acid composition of the placentae. E2 at parturition in the NPR group (control cows, n = 10) was higher than in PR cows (placental ret

Surface Modification of Gold Nanoparticle Impacts Distinct Lipid Metabolism.

Gold nanomaterials have garnered significant attention in biomedicine owing to their tunable size and morphology, facile surface modification capabilities, and distinctive optical properties. The surface functionalization of these nanoparticles can enhance their safety and efficacy in nanomedical applications. In this study, we examined the biological effects of gold nanoparticles (GNPs) with three distinct surface modifications (polyethylene glycol, chitosan, and polyethylenimine) in murine models, elucidating their mechanisms of action on hepatic tissue at both the transcriptomic and metabol

Emergence of omega-3 fatty acids in biomedical research.

Shortly after the discovery that linoleic acid was an essential fatty acid in 1930, α-linolenic acid also was reported to prevent the fatty acid deficiency syndrome in animals. However, several prominent laboratories could not confirm the findings with α-linolenic acid, and as a result there was a loss of interest in omega-3 fatty acids in lipid research. Even the findings that a prostaglandin can be synthesized from eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) is necessary for optimum retinal function generated only limited interest in omega-3 fatty acids. The breakthrough came

High heritability of metabolomic profiles in families burdened with premature cardiovascular disease.

Integration of genetic and metabolic profiling holds promise for providing insight into human disease. Coronary artery disease (CAD) is strongly heritable, but the heritability of metabolomic profiles has not been evaluated in humans. We performed quantitative mass spectrometry-based metabolic profiling in 117 individuals within eight multiplex families from the GENECARD study of premature CAD. Heritabilities were calculated using variance components. We found high heritabilities for amino acids (arginine, ornithine, alanine, proline, leucine/isoleucine, valine, glutamate/glutamine, phenylalan

Biochemical and scanning electron microscope study of lipids chloroform-methanol extracted from unerupted and erupted human tooth enamel.

Almost twice as much fatty acid was extracted without demineralization from the mature enamel of erupted teeth treated with 2:1 chloroform-methanol as from the enamel of unerupted teeth (32.64 micrograms/g versus 15.60 micrograms/g). Palmitic C16:0, stearic C18:0, oleic C18:1 omega 9 and linoleic acids C18:2 omega 6 were the chief fatty acids detected by gas-liquid chromatography. The percentage of free fatty acids and diglycerides was larger in unerupted than erupted teeth, whereas that of triglycerides, phosphatidylethanolamine and phosphatidylcholine was larger in erupted teeth. High concentrations of arachidonic acid induce platelet aggregation and serotonin release independent of prostaglandin endoperoxides and thromboxane A2.

We examined platelet aggregation and serotonin release, induced by less than 60 micro M arachidonic acid, using washed platelet suspensions in the absence of albumin. The concentration of arachidonic acid used did not cause platelet lysis. Platelet responses induced by less than 20 micro M arachidonic acid were inhibited by aspirin, whereas those induced by above 30 micro M arachidonic acid were not inhibited, even by both aspirin and 5,8,11,14-eicosatetraynoic acid. Although phosphatidic acid and 1,2-diacylglycerol increased after the addition of arachidonic acid in aspirin-treated platelets,
