The time course and regional variations of lipid peroxidation after diffuse brain injury in rats.

Free radicals are generated after head injury. These radicals rapidly react with polyunsaturated fatty acids in the cell membrane and cause membrane destruction. This process is called lipid peroxidation. Malondialdehyde (MDA) is one of the end products of lipid peroxidation, and it is a frequently used indicator of lipid peroxidation in biological tissues. Using a diffuse head injury animal model, we studied the time course of lipid peroxidation in different regions of injured rat brains. In the present study, the MDA levels were 36.7%, 41.8%, and 35.1% greater than sham at one hour after inj

Superoxide dismutase activity and lipid peroxidation in the liver of guinea pig infected with Leptospira interrogans.

Superoxide dismutase (SOD) activity and the degree of lipid peroxidation were studied over a two week period in guinea pigs infected with Leptospira interrogans derived from wild mice. The total SOD activity in infected host liver increased by four-fold two days after infection; this was followed by a 20% decrease resulting in levels comparable to normal, uninfected liver. During the period of decreasing SOD activity after day two, the levels of TBA-reactive material (TBARS) are increased by three-fold in infected guinea pig, liver, compared to uninfected liver. The results indicate that SOD a

Influence of microwaves on different types of receptors and the role of peroxidation of lipids on receptor-protein shedding.

The effects of a continuous wave or pulse-modulated, 900 MHz microwave field were studied by in vitro assays of rat chemoreceptors. The pulsed field was modulated as rectangular waves at rates of 1, 6, 16, 32, 75, or 100 pps. The pulse-period to pulse-duration ratio was 5 in all cases, and specific absorption rates (SARs) ranged from 0.5 to 18 W/kg. Binding of ligands to cell membranes was differentially affected by exposure to microwaves. For example, binding of H3-glutamic acid to hippocampal cells was not altered by a 15 min exposure to a continuous wave field at 1 W/kg, but binding of H3-d

Liver lipid peroxidation and glutathione-related defence enzyme systems in mice treated with paracetamol.

Glutathione levels were found to be decreased while lipid peroxide levels were increased in total liver homogenates 6 h following paracetamol treatment (500 mg kg-1 i.p.). Furthermore, it has been determined that cytosolic glutathione S-transferase (GST) activity was decreased and glutathione peroxidase (GSH-Px) activity remained unchanged. On the other hand, a decrease in liver microsomal lipid peroxide levels and an increase in GST and GSH-Px activity has been observed. We concluded that decreased lipid peroxide levels in microsomes could be a consequence of increased GSH-Px and GST enzyme a

Suppressive effect of vitamin E on lipid peroxidation in halothane-administered guinea pig liver.

The effect of vitamin E on halothane-induced liver damage was studied in guinea pig halothane hepatitis. Twenty animals were divided into 3 groups, consisting of a control group, a halothane group and a vitamin E + halothane (H) group. The animals in the control group (n = 6) were allowed to inhale air only. The animals in the halothane group (n = 6) and the vitamin E + H group (n = 8) were allowed to inhale 1% halothane with air. Animals in the vitamin E + H group were additionally injected with 30 mg kg-1 of vitamin E 30 minutes prior to inhalation of halothane. Blood was aspirated from the

Effects of an extract of Gingko biloba on bromethalin-induced cerebral lipid peroxidation and edema in rats.

The effects of administration of a commercially available extract of Gingko biloba (EGB) on bromethalin-induced brain lipid peroxidation and cerebral edema in adult male Sprague-Dawley rats was determined. Gingko biloba extract was given (100 mg/kg) by gavage immediately after bromethalin (1.0 mg/kg) administration. Rats were euthanatized at 24 hours after dosing. Brain lipid peroxidation was determined by measurement of brain malonaldehyde-thiobarbituric acid chromophore (MDA-TBA) concentration, brain sodium concentration, and brain water content. Treatment of bromethalin-dosed rats (10/group

The effects of aluminum ingestion on intestinal cadmium absorption, lipid peroxidation and alkaline phosphatase activity.

Consumption of dietary aluminum (A1, 2000 or 4000 ppm) for 1 month did not influence mouse body weight. In order to estimate an absorption of cadmium (Cd) from the gastrointestinal tract, mice were intubated twice every 24 h with Cd (CdCl2). In the A1-supplemented mice, Cd absorption increased rather than decreased. Intestinal Cd and metallothionein concentrations were increased by the A1 supplement. Intestinal alkaline phosphatase activity and lipoperoxide concentrations were also enhanced by A1 ingestion. Of course, intestinal A1 concentration was at a high level in the A1-supplemented group

[Lipid peroxidation and antioxidative activity in the tumor tissue and blood of patients with neuro-oncological diseases].

Study of the content of products of lipid peroxidation, total lipid antioxidation activity, and superoxide dismutase activity in the tissue and blood of 31 patients (23 with gliomas and 8 with meningiomas) showed the level of malonic dialdehyde (MDA) in the tumor tissue to be significantly lower statistically than in the cerebral tissue. The MDA content was much lower in meningiomas than in gliomas. No essential differences were revealed in the MDA level in benign and malignant gliomas. This value correlated well with the total lipid antiperoxidation activity which was twice higher in tumor ti

Early measurement of systemic lipid peroxidation products in the plasma of major blunt trauma patients.

We sought evidence of oxidant-induced biological membrane damage in 43 resuscitated blunt trauma patients (average ISS, 36.9) within 2-6 hours of injury and before anaesthesia and surgery. The plasma levels of the lipid peroxidation products (conjugated dienes, CDs A 233 nm) and malondialdehyde (MDA, nMol/ml) and the oxidant-inducing effect of the trauma plasma on normal FMLP-stimulated neutrophils were compared to those of control subjects. No differences were observed in the plasma levels of MDA (1.73 +/- 2.15 vs. 1.45 +/- 0.70 nMol/ml) and CDs (2.07 +/- 2.16 vs. 1.28 +/- 0.60 A 233nm), or o

Oxygen radicals, lipid peroxidation, and neutrophil infiltration after small-intestinal ischemia and reperfusion.

In this study the role of free radicals, lipid peroxidation, and neutrophil infiltration as mediators of ischemia and reperfusion-induced intestinal mucosal damage were investigated. We used a rat experimental model in which a ligated loop of the distal ileum was subjected to ischemia and reperfusion and the ensuing mucosal damage was assessed by means of lysosomal enzyme release and intestinal permeability measurements. We also determined the mucosal content of malondialdehyde, a lipid peroxidation product, and the mucosal activity of myeloperoxidase, a neutrophil granulocyte marker. Ischemia

[Experimental studies on the role of retinal lipid peroxidation induced by intraocular iron foreign body].

Experimental studies were made to evaluate the role of lipid peroxidation in retinal damage by intraocular iron foreign bodies. It was found that after iron particles were implanted into the vitreous bodies (1) iron concentration in the aqueous humour rose rapidly and significantly (n = 24, P less than 0.01), (2) malondialdehyde (MDA) a degradation metabolite of lipid peroxidation, in the retinas increased significantly (n = 24, P less than 0.05), and (3) superoxide dismutase (SOD) activity decreased (n = 11, P less than 0.05). These findings suggest that lipid peroxidation plays a key role in

[Pathogenetic significance of lipid peroxidation in damage of the proximal segment of the nephron in acute Masugi's nephritis].

During the acute experimental nephritis a decrease of reabsorption by proximal tubules is combined with the activation of the lipid peroxidation into the renal cortex without changes in the activation of lysosomal enzyme of acid phosphatase and cathepsin D. Ionol, an inhibitor of oxygen products, exerts a protective action on the renal function and reabsorption by proximal tubules, decreasing concentration of the malondialdehyde into the renal cortex.

Correlation among lipid peroxidation, brain energy metabolism and brain oedema in cerebral ischaemia.

In order to study the involvement of lipid peroxidation in the generation of ischaemic cell damage, sequential measurements were made on the chemiluminescence, energy metabolism, water content and the volume of Na+ and K+ in the cerebral cortex using a rat model for severe cerebral ischaemia. It was found that the amount of chemiluminescence increased due to ischaemia and increased further following recirculation. In chemiluminescence spectral analysis, peaks were found at 480, 520-530, 570, 620-640, 680-700 nm. It is thought that these wavelengths reflect the release of energy due to the redu

Nonheme iron in sickle erythrocyte membranes: association with phospholipids and potential role in lipid peroxidation.

Previous studies documented the abnormal association of heme and heme proteins with the sickle RBC membrane. We have now examined RBC ghosts and inside-out membranes (IOM) for the presence of nonheme iron as detected by its formation of a colored complex with ferrozine. Sickle ghosts have 33.8 +/- 18.2 nmol nonheme iron/mg membrane protein, and sickle IOM have 4.3 +/- 3.0 nmol/mg. In contrast, normal RBC ghosts and IOM have no detectable nonheme iron. The combination of heme and nonheme iron in sickle IOM averages nine times the amount of membrane-associated iron in normal IOM. Kinetics of the

[Role of bilirubin as a natural antioxidant in regulating lipid peroxidation intensity in acute viral hepatitis].

Elevation of the content of lipid peroxidation (LPO) products in blood serum of patients with acute virus hepatitis (VH) is caused by an increase in the patients' blood serum lipids rather than by the intensity of peroxide reactions in lipids. There is a reverse correlation between the content of LPO products and bilirubin level and a direct correlation between lipid antioxidant activity (AOA) and bilirubin level. Marked antioxidant action of bilirubin that compares very favourably with the action of ionol (4-methyl-2,6-ditretbutylphenol) was demonstrated in the model of oxidation of methyl ol

Glutathione depletion and in vitro lipid peroxidation in mercury or maleate induced acute renal failure.

Nephrotoxic acute renal failure was experimentally induced in male rats by s.c. application of mercuric chloride and i.p. administration of maleate, respectively. Mercuric chloride and maleate are known to enhance the formation of free radicals and peroxides, which presumably overload the cell's natural elimination mechanisms for these highly reactive intermediates. In addition, a reduction in activities of superoxide dismutase, catalase and glutathione-peroxidase, enzymes responsible for the protection of cells against peroxidative action of superoxide anions and hyperperoxides was found.
