Establishment of an in silico phospholipidosis prediction method using descriptors related to molecular interactions causing phospholipid-compound complex formation.

Although phospholipidosis (PLD) often affects drug development, there is no convenient in vitro or in vivo test system for PLD detection. In this study, we developed an in silico PLD prediction method based on the PLD-inducing mechanism. We focused on phospholipid (PL)-compound complex formation, which inhibits PL degradation by phospholipase. Thus, we used some molecular interactions, such as electrostatic interactions, hydrophobic interactions, and intermolecular forces, between PL and compounds as descriptors. First, we performed descriptor screening for intermolecular force and then develo

Renal lipidosis in patients enrolled in a methadone substitution program.

Kidney biopsies often show accumulation of lipids or lipidlike material. Evidence has been provided that lipids can directly initiate and contribute to the progression of glomerular and tubulointerstitial lesions. In this study we describe a renal lipidosis occurring in patients with a positive history of narcotic abuse who were enrolled in a methadone substitution program. All 3 patients presented with proteinuria (2.5-20 g/d) and impaired renal function. Renal biopsy revealed a pronounced extracellular and intracellular deposition of lipidlike material in the glomerular, interstitial, and tu

In vitro validation of drug-induced phospholipidosis.

Intracellular accumulation of phospholipids with lamellar bodies is a hallmark of drug-induced phospholipidosis (PLD) which is caused by impaired phospholipid metabolism of the lysosome. Although it remains uncertain whether PLD is associated with the adverse effects, sponsors generally terminate the development of a candidate drug when PLD is observed in an organ. For drugs that are used without serious adverse events, there should be labels indicating that the drug can induce PLD. We conducted LipidTox and NBD-PE assays for detecting PLD to compare and validate the methods. In the case of co

A 96-well flow cytometric screening assay for detecting in vitro phospholipidosis-induction in the drug discovery phase.

Drug-induced phospholipidosis is caused by lysosomal accumulation of the drug, resulting in the disturbance of phospholipid degradation and a consequent excessive phospholipid accumulation. Depending on the type and number of tissues affected, phospholipidosis occurrence in test animals can raise safety issues, which may be critical for the risk assessment. Safety profiling of potential phospholipidosis-inducing drugs in the drug discovery phase can predict these late obstructions of drug development. For this purpose, a flow cytometric assay based on the difference in fluorescent phospholipid

Pulmonary involvement with Erdheim-Chester disease: radiographic and CT findings.

OBJECTIVE: We determined the chest radiographic and CT findings of Erdheim-Chester disease with pulmonary involvement. MATERIALS AND METHODS: We retrospectively reviewed the radiologic images of 15 patients with biopsy-proven Erdheim-Chester disease. Nine patients had chest radiographic images and CT scans that were available for review. Six men and three women were studied (age range, 25-70 years; mean age, 56 years). Two radiologists interpreted all images by consensus. Lung parenchyma was assessed for the type and distribution of disease. Bronchi, pleurae, hila, and mediastinal and extrath

[Hyalinosis cutis and mucosae. Ultrastructural histochemical aspects indicating intracellular accumulation of glycosaminoglycans].

The exact nature of the genetic defect of hyalinosis cutis et mucosae or d'Urbach-Whiete syndrome is still matter of controversy. The present article reports on three new cases in which several different ultrastructural and biochemical investigations add more arguments to support an anomaly of the glycosaminoglycans degradation in the dermal fibroblasts. Cationic dyes as polyethyleneimine and alcian blue show an intense ultrastructural staining of the abnormal basal laminae and the intracellular lysosomal bodies in cultured fibroblasts. These are related to the accumulation of anionic charged

[Chlorphentermine-induced lipidosis of the cochlea and the cochlear nucleus].

The anoretic drug chlorphentermine induces, after long-term feeding, an accumulation of phospholipids mainly stored in lysosomes. Inner hair cells and nerve running to these cells are especially affected, whereas the outer hair cells and their nerves are less changed. Supporting cells and ganglion cells of the spiral ganglion always exhibit inclusions. Changes in the perikarya of the cochlear nucleus are not so outstanding and are in the scope of the alterations found in other parts of the nervous system. The morphological alterations allow some conclusions about the phospholipid metabolism an

Clinical and radiological findings in subjects with unilateral Ménière's disorder.

In order to contribute to the knowledge of the possible causes of Ménière's disorder, the authors have conducted systematic studies based on blood analysis and on petrous bone radiological examinations in patients suffering from unilateral Ménière's disorder and in two control groups, one of patients with unilateral sudden sensorineural deafness and the other one of normally hearing subjects suffering from neurological diseases. Blood tests have revealed systemic disorders (dyslipidosis, hypoglycaemia, hyperglycaemia, hypothyroidism lues) in 49%, 48% and in 50% of the subjects respectively wit

Lysosomal alterations in cultured macrophages exposed to anorexigenic and psychotropic drugs.

Cultured rat macrophages were used for an in vitro study of drug-induced lipidosis. Cells were exposed for 24 hours to equimolar concentrations (5 X 10(-5) and 1 X 10(-4) M) of the following amphiphilic (amphipathic) cationic drugs: chlorphentermine, amitriptyline, 1-chloro-amitriptyline, iprindole, noxiptiline, chlorpromazine. In addition the less amphiphilic drug phentermine was used. Ultrastructurally, the cytologic changes essentially consisted of formation of multilamellated cytoplasmic inclusions, which possessed acid phosphatase activity. The abnormal inclusions are interpreted to resul
