Spontaneous confabulation, reality monitoring, and the limbic system--a review.

Patients with anterior limbic damage may present a distinct syndrome, spontaneous confabulation: they fail in common memory tests, act on the basis of previous habits rather than currently relevant memories, produce confabulations composed of elements of past true events, are disorientated, and are absolutely convinced about the veracity of their perceived reality. Spontaneous confabulation is independent of other false memories, such as, provoked confabulations or illusory recognition. Studies showed that spontaneous confabulators fail to suppress (inactivate) evoked memories that do not pert

Exploring Hippocampal Neural Network Dynamics in Rodents Using Virtual Reality: A Narrative Review on Systems, Spatial Coding, and Applications.

Virtual reality (VR) provides a highly controllable, immersive, and interactive platform for investigating hippocampal mechanisms underlying spatial learning and memory. In rodent models, VR-based paradigms have elucidated how hippocampal subregions including CA1, CA3, and the dentate gyrus support spatial representation, pattern separation, and memory consolidation. Within VR environments, place cells, grid cells, head-direction cells, and boundary vector cells exhibit characteristic remapping dynamics, reflecting both the experimental advantages and sensory constraints inherent to VR. Althou…

Converging diencephalic and hippocampal supports for episodic memory.

To understand the neural basis of episodic memory it is necessary to appreciate the significance of the fornix. This pathway creates a direct link between those temporal lobe and medial diencephalic sites responsible for anterograde amnesia. A collaboration with Andrew Mayes made it possible to recruit and scan 38 patients with colloid cysts in the third ventricle, a condition associated with variable fornix damage. Complete fornix loss was seen in three patients, who suffered chronic long-term memory problems. Volumetric analyses involving all 38 patients then revealed a highly consistent rel

A serine protease KLK8 emerges as a regulator of regulators in memory: Microtubule protein dependent neuronal morphology and PKA-CREB signaling.

The multitude of molecular pathways underlying memory impairment in neurological disorders and aging-related disorders has been a major hurd … osphorylated CREB (pCREB), leading to downregulation of memory-linked genes and consequent impaired memory consolidation. These findings revealed a protease associated novel pathway of memory impairment in which KLK8 may act as a "regulator of regulators", suggesting its exploration as an important therapeutic target of memory disorders.

Toxin-Induced Experimental Models of Learning and Memory Impairment.

Animal models for learning and memory have significantly contributed to novel strategies for drug development and hence are an imperative part in the assessment of therapeutics. Learning and memory involve different stages including acquisition, consolidation, and retrieval and each stage can be characterized using specific toxin. Recent studies have postulated the molecular basis of these processes and have also demonstrated many signaling molecules that are involved in several stages of memory. Most insights into learning and memory impairment and to develop a novel compound stems from the i

Dissociation of long-term verbal memory and fronto-executive impairment in first-episode psychosis.

BACKGROUND: Verbal memory is frequently and severely affected in schizophrenia and has been implicated as a mediator of poor clinical outcome. Whereas encoding deficits are well demonstrated, it is unclear whether retention is impaired. This distinction is important because accelerated forgetting implies impaired consolidation attributable to medial temporal lobe (MTL) dysfunction whereas impaired encoding and retrieval implicates involvement of prefrontal cortex. METHOD: We assessed a group of healthy volunteers (n=97) and pre-morbid IQ- and sex-matched first-episode psychosis patients (n=97

Interim testing prospectively and retrospectively boosts precision and accuracy but not bias of continuous visual memory.

Testing studied information not only facilitates long-term consolidation of tested content but also enhances subsequent acquisition of new knowledge. These dual aspects of test-enhanced learning have predominantly been examined using verbal materials. In the present study, we extended this investigation to low-level visual features, in particular visual salience (luminance and color saturation), to determine whether interim testing can retrospectively consolidate and prospectively boost learning of continuous visual information. Across two experiments, participants studied five lists of scene

Melatonin modulates daytime-dependent synaptic plasticity and learning efficiency.

Mechanisms of hippocampus-related memory formation are time-of-day-dependent. While the circadian system and clock genes are related to timing of hippocampal mnemonic processes (acquisition, consolidation, and retrieval of long-term memory [LTM]) and long-term potentiation (LTP), little is known about temporal gating mechanisms. Here, the role of the neurohormone melatonin as a circadian time cue for hippocampal signaling and memory formation was investigated in C3H/He wildtype (WT) and melatonin receptor-knockout ( MT 1 / 2 - / - ) mice. Immunohistochemical and immunoblot analyses revealed

Effects of single oral doses of clobazam, diazepam and lorazepam on performance tasks and memory.

The effects on memory and psychomotor performance and the subjective effects of three anxiolytic benzodiazepines (lorazepam 2 mg, diazepam 10 mg and clobazam 20 mg p.o.) have been evaluated in a double-blind, placebo-controlled, cross-over study in 10 healthy volunteers. At each session, measurements were made prior to and +3.5 h after drug administration, except in the case of REY's test, which was presented at H + 1 h (learning) and was evaluated at H + 8 h and at H + 24 h (delayed recall). Single clinical doses of diazepam and lorazepam caused anterograde amnesia by disturbing acquisition,

Cholecystokinin (CCK): a neuromodulator with therapeutic potential in Alzheimer's and Parkinson's disease.

Cholecystokinin (CCK) is a neuropeptide modulating digestion, glucose levels, neurotransmitters and memory. Recent studies suggest that CCK exhibits neuroprotective effects in Alzheimer's disease (AD) and Parkinson's disease (PD). Thus, we review the physiological function and therapeutic potential of CCK. The neuropeptide facilitates hippocampal glutamate release and gates GABAergic basket cell activity, which improves declarative memory acquisition, but inhibits consolidation. Cortical CCK alters recognition memory and enhances audio-visual processing. By stimulating CCK-1 receptors (CCK-1Rs

Handling, task complexity, time-of-day, and sleep deprivation as dynamic modulators of recognition memory in mice.

Sleep is essential for optimal cognitive functioning. Although we lack a complete understanding of the role of sleep in memory consolidation, we know that various factors that disturb sleep or sleep quality have consequences for cognitive performance. Such factors can be unintended components of behavioral experiments on rodents and other experimental animals that generate differing results from different labs. These experimental variables include habituation to handling, intended or unintended sleep deprivation, task complexity, time of testing, and environmental features. We have examined ho…

Activity-Dependent Neuroplasticity Induced by an Enriched Environment Reverses Cognitive Deficits in Scribble Deficient Mouse.

Planar cell polarity (PCP) signaling is well known to play a critical role during prenatal brain development; whether it plays specific roles at postnatal stages remains rather unknown. Here, we investigated the role of a key PCP-associated gene scrib in CA1 hippocampal structure and function at postnatal stages. We found that Scrib is required for learning and memory consolidation in the Morris water maze as well as synaptic maturation and NMDAR-dependent bidirectional plasticity. Furthermore, we unveiled a direct molecular interaction between Scrib and PP1/PP2A phosphatases whose levels were…
