The mouse fetal-placental arterial connection: A paradigm involving the primitive streak and visceral endoderm with implications for human development.

In Placentalia, the fetus depends upon an organized vascular connection with its mother for survival and development. Yet, this connection w … ed visceral endoderm undergoes an epithelial-to-mesenchymal transition, contributing extraembryonic mesoderm to the placental arterial vasculature, and to the allantois, or pre-umbilical tissue. In addition, visceral endoderm bifurcates into the allantois where, with the primitive streak, it organizes the nascent umbilical artery and promotes allantoic elo …

Chiari malformation type I: a case-control association study of 58 developmental genes.

Chiari malformation type I (CMI) is a disorder characterized by hindbrain overcrowding into an underdeveloped posterior cranial fossa (PCF), often causing progressive neurological symptoms. The etiology of CMI remains unclear and is most likely multifactorial. A putative genetic contribution to CMI is suggested by familial aggregation and twin studies. Experimental models and human morphometric studies have suggested an underlying paraxial mesoderm insufficiency. We performed a case-control association study of 303 tag single nucleotide polymorphisms (SNP) across 58 candidate genes involved in…

Repression of Wasp by JAK/STAT signalling inhibits medial actomyosin network assembly and apical cell constriction in intercalating epithelial cells.

Tissue morphogenesis requires stereotyped cell shape changes, such as apical cell constriction in the mesoderm and cell intercalation in the ventrolateral ectoderm of Drosophila. Both processes require force generation by an actomyosin network. The subcellular localization of Myosin-II (Myo-II) dictates these different morphogenetic processes. In the intercalating ectoderm Myo-II is mostly cortical, but in the mesoderm Myo-II is concentrated in a medial meshwork. We report that apical constriction is repressed by JAK/STAT signalling in the lateral ectoderm independently of Twist. Inactivation …

Giant Occipital Meningoencephalocele in a Neonate: A Therapeutic Challenge.

Encephalocele is a rare lesion, being an embryological mesodermal anomaly which results in a defect in the cranium and dura, associated with herniation of meninges, cerebrospinal fluid, or brain tissues through a defect usually covered by scalp. Surgical management of children with giant occipital encephalocele requires careful attention to pediatric anesthetic and surgical principles. We present a case of a giant occipital encephalocele highlighting the problems encountered in its management.

A case of Ollier disease with non-small cell lung cancer and review of the literature.

Ollier disease is a rare, non-hereditary mesodermal dysphasia, characterized by multiple enchondromas, which demonstrate asymmetric involvement of the metaphyses of the long bones. Many malignancies, especially chondrosarcomas, may be observed in association with this disease. The clinical and radiologic characteristics of a case involving a 44-year-old male patient with non-small cell lung cancer are presented together with the clinical characteristics of other cases reported in the literature.

[Stomatofacial morphogenesis. Some specifications].

The initial phases of the embryogenetic process are evaluated in order to define its mesenchymal matrix. The influence of cephalic neural crests on mesenchymal derivatives is also underlined. Finally paraxial and lateral mesodermal function on cranial development and brachial arches composition are described. A correlation is established between head and neck, neural crests, mixed and sensorial cranial nerves. Tongue and orbital cavity muscles are identified as paraxial matrix derivative organs.

Müllerian dysgenesis.

The multifaceted syndrome of müllerian dysgenesis appears to represent a defect in mesodermal organization and is often associated with renal and skeletal anomalies. This report discusses the embryology, multifactorial etiology, clinical presentation and management of the syndrome from a historical standpoint and details our experience with 35 affected patients. Both Frank dilatation and McIndoe vaginoplasty appear to be acceptable means of therapy. The therapeutic success depends to a large extent on the psychosocial adaptation of the patient.

The pedicle in neurofibromatosis.

Three cases of von Recklinghausen's disease (neurofibromatosis) are described, one with agenesis of a pedicle, another with multiple hypoplastic pedicles, and a third with bilateral hypoplastic pedicles producing a spondylolistesis. These peidcle deficiencies are manifestations of mesodermal dysplasia. Agenesis of the pedicle most likely represents a defect in chondrification. Hypoplasia probably represents a partial lack of development of the pedicle and the ipsilateral arch. Hypoplasia or agenesis of a pedicle should suggest the possibility of underlying neurofibromatosis.

[Libernan-Cole-Golts syndrome. Focal dermic hypoplasia. Report of a case].

A case is reported corresponding to the so-called focal dermic hypoplasia and that we have named Liberman-Cole-Goltz syndrome. This is possibly the fourth case published in Mexican literature and is an additional report to those described in the international literature. This syndrome is characterized by variable ectodermal and mesodermal defects with congenital lesions most frequently found in skin and in the skeleton. The clinical, radiological and histopathological finds in the case studied are pointed out.

A pluripotent stem cell model of Emberger syndrome reveals reduced lymphatic endothelial differentiation.

Emberger syndrome (ES), an autosomal dominant disorder characterized by congenital deafness, primary lymphedema, and predisposition to myeloid malignancies, is caused by mutations in the GATA2 gene. Although primary lymphedema is an important hallmark of ES, the pathophysiology remains unclear due to the lack of a suitable experimental model. In this study, we isolated induced pluripotent stem cells (iPSCs) from two patients with ES (i.e., ES-iPSCs) and analyzed their in vitro lymphatic differentiation potential via the mesodermal progenitor stage. KDR+ CD34+ early mesodermal progenitors gener…

Novel biphasic mechanism of the canonical Wnt signalling component PYGO2 promotes cardiomyocyte differentiation from hUC-MSCs.

Human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) are used to regenerate the myocardium during cardiac repair after myocardial infarction. However, the regulatory mechanism underlying their ability to form mesodermal cells and differentiate into cardiomyocytes remains unclear. Here, we established a human-derived MSCs line isolated from healthy umbilical cords and established a cell model of the natural state to examine the differentiation of hUC-MSCs into cardiomyocytes. Quantitative RT-PCR, western blotting, immunofluorescence, flow cytometry, RNA Seq, and inhibitors of canonica…

Vitamin A administration in lizards during tail regeneration determines epithelial mucogenesis and delays muscle and cartilage differentiation.

Regenerating epidermis and spinal cord is essential to maintain tail regeneration in lizards. The effects of vitamin A, an inhibitor of epit … e formation of a corneous layer are affected as well. The effects of vitamin A are dramatic also on mesoderm cells since the treatment stimulates an invasion of blood cells likely due to the disruption of the wall of blood vessels, mesenchymal cell death (pycnosis), and diffuse phagocytosis by immune cells. A delay of cartilage differentiation and cartilag …

Type 3 inositol 1,4,5-trisphosphate receptor negatively regulates apoptosis during mouse embryonic stem cell differentiation.

Ca(2+) signals generated by inositol 1,4,5-trisphosphate receptors (IP(3)Rs) are crucial for cellular processes such as apoptosis and differ … increased in control ones. Furthermore, IP(3)R3 knockdown led to a suppression of the expression of mesodermal and mesoendodermal but not ectodermal markers. The differentiation suppressions were further confirmed by the impaired differentiation of mesodermal and some of the endodermal but not ectodermal derivatives. Such defects were partially because of …

Retinoic acid selectively regulates Fgf10 expression and maintains cell identity in the prospective lung field of the developing foregut.

Although respiratory tract defects that result from disruption of retinoic acid (RA) signaling have been widely reported, the mechanism by which endogenous RA regulates early lung morphogenesis is unknown. Here, we provide novel evidence that a major role for RA is to selectively maintain mesodermal proliferation and induce fibroblast growth factor 10 (Fgf10) expression in the foregut region where the lung forms. By using a pan-RAR antagonist (BMS493) in foregut explant cultures, we show that bud initiation is selectively blocked in the prospective respiratory region by failure to induce Fgf10…

RNA interference demonstrates a role for nautilus in the myogenic conversion of Schneider cells by daughterless.

Schneider SL2 cells activate the myogenic program in response to the ectopic expression of daughterless alone, as indicated by exit from the cell cycle, syncytia formation, and the presence of muscle myosin fibrils. Myogenic conversion can be potentiated by the coexpression of DMEF2 and nautilus with daughterless. In RT-PCR assays Schneider cells express two mesodermal markers, nautilus and DMEF2 mRNAs, as well as very low levels of daughterless mRNA but no twist. Full-length RT-PCR products for nautilus and DMEF2 encode immunoprecipitable proteins. We used RNA-i to demonstrate that both endog…

Muscle and nerve-specific regulation of a novel NK-2 class homeodomain factor in Caenorhabditis elegans.

We have identified a new Caenorhabditis elegans NK-2 class homeobox gene, designated ceh-24. Distinct cis-acting elements generate a complex neuronal and mesodermal expression pattern. A promoter-proximal enhancer mediates expression in a single pharyngeal muscle, the donut-shaped m8 cell at the posterior end of the pharynx. A second mesodermal enhancer is active in a set of eight nonstriated vulval muscles used in egg laying. Activation in the egg laying muscles requires an 'NdE-box' consensus motif (CATATG) which is related to, but distinct from, the standard E-box motif bound by the MyoD fa…

H19 and Igf2 are expressed and differentially imprinted in neuroectoderm-derived cells in the mouse brain.

Igf2 and H19 are reciprocally imprinted genes that are closely linked and coexpressed in tissues of mesodermal and endodermal origin. Here we report that coexpression of these genes is also found in specific fetal tissues of neuroectodermal origin, that is in the ventral midline region of both the hindbrain and spinal cord. For cells of neuroectodermal origin, complete absence of Igf2 and H19 transcription was previously described. Analysis of allele-specific expression of both Igf2 and H19 in the ventral midline region of the hindbrain shows that H19 is expressed monoallelically, with the pat…

Differentiation of beating cardiac muscle cells from a derivative of P19 embryonal carcinoma cells.

A clonal derivative named CL6 has been isolated from pluripotent P19 embryonal carcinoma (EC) cells, after long term culture under conditions for mesodermal differentiation. The CL6 subline has a morphology similar to P19 cells and grows exponentially in standard medium, but unlike P19 cells, it efficiently differentiates into beating cardiac muscle in adherent culture with 1% dimethyl sulfoxide (DMSO). During differentiation, CL6 cells displayed the following general cardiac muscle properties: (1) alpha- and beta-cardiac myosin heavy chain (MHC) transcripts were first detected on day 10 after…
