Low-frequency tone pips elicit exaggerated startle reflexes in C57BL/6J mice with hearing loss.

The strength of the acoustic startle reflex (ASR) as a function of age was studied in adult C57BL/6J and CBA/CaJ mice, because altered ASR levels are a potential behavioral consequence of the neural reorganization that accompanies the early-onset hearing loss of the C57BL, in contrast to the normal-hearing CBA. For C57BL mice at 14-36 weeks of age, compared with 7-week-old mice, high-frequency thresholds measured with the auditory brainstem response (ABR) were less sensitive by about 25-30 dB while the hearing loss at low frequencies was 10-15 dB, but by 60 weeks losses of 45-50 dB were presen

The functional age of hearing loss in a mouse model of presbycusis. I. Behavioral assessments.

Presbycusis is a common form of hearing loss that progresses from high to low frequencies with advancing age. C57BL/6J mice experience a rapid progression of presbycusis-like hearing deficits and thus provide a convenient animal model for evaluating behavioral, physiological and anatomical correlates of the disorder. Previous studies of C57BL/6J mice have relied on short-term observations of age-matched subject groups to reconstruct a time course for auditory pathologies. Such statistical approaches are weakened by the variability of hearing thresholds in young mice and the inconsistent timing

Th1-polarizing immunization with egg antigens correlates with severe exacerbation of immunopathology and death in schistosome infection.

In schistosomiasis mansoni, parasite eggs precipitate an intrahepatic granulomatous and fibrosing inflammatory process, which is mediated by, and dependent on, MHC class II-restricted CD4 T helper (Th) lymphocytes specific for schistosome egg antigens (SEA). In the mouse model of the disease, CBA mice develop large granulomas, whereas in C57BL/6 (BL/6) mice these granulomas are significantly smaller. To further investigate how the prevailing cytokine environment influences the development of the egg-induced immunopathology, we immunized the low-pathology BL/6 mice with SEA in complete Freund's

Acquired, but not innate, immune responses to Streptococcus pneumoniae are compromised by neutralization of CD40L.

Streptococcus pneumoniae is a significant pathogen of young children and the elderly. Systemic infection by pneumococci is a complex process involving several bacterial and host factors. We have investigated the role of CD40L in host defense against pneumococcal infection. Treatment of mice with MR-1 antibody (anti-CD154/CD40L) markedly reduced antibody responses to the pneumococcal protein PspA, elicited by immunization of purified protein or whole bacteria. In mice immunized with whole bacteria, MR-1 treatment reduced antibody responses to capsular polysaccharides but not cell wall polysacch

Influence of embryos on PGF2 alpha production by uterine tissue in pregnant mice.

The aims of the present study are to find out if preimplantation mice embryos can synthesize prostaglandins and if the presence of the embryos in the uteri modifies the uterine prostaglandin synthesis. Both uteri of adult pregnant mice and embryos were collected at day two, three and four of pregnancy and the synthesis and release of PGE and PGF2 alpha was measured by radioimmunoassay. At day five of pregnancy, embryos are tightly adherent to epithelium, so tied uterine horns (without embryos) were compared to control uteri (with embryos). We found that embryos synthesize PGE and PGF2 alpha an

Limb and other postcranial skeletal defects induced by amniotic sac puncture in the mouse.

A mouse model for studying the teratogenic effects of amniotic sac puncture has recently been established (MacIntyre et al. 1995), and the anomalies encountered included cleft palate, limb and tail abnormalities, and postural deformities. In order to investigate the underlying mechanism(s) involved, mouse embryos subjected to amniotic sac puncture on day 13 of gestation were examined externally on day 19, then 'cleared' and bulk stained with alizarin red S and Alcian blue to reveal, respectively, their bony and cartilaginous elements. This procedure allowed a comparison to be made between the

Intensity of Schistosoma mansoni infection determines alterations in hepatic drug metabolism.

Zoxazolamine paralysis times have been used as a probe to measure the activities of cytochrome P450 1A1 in mice in vivo. The results indicate that male and female mice of the BALB/c and CBA/J strain do not show altered paralysis times if infected with less than 4 worm pairs. Alterations were observed only in animals harbouring more than 5 worm pairs irrespective of the sex or strain of mouse being used. The study has extended the findings of other workers showing that mice infected with fewer than 4-5 worm pairs of S. mansoni do not show any alteration in the metabolism of pentobarbital in viv

Development of aging-associated monoclonal gammapathies with antibody activity to the antigen used for immunization of young mice.

The influence of immunization with dinitrophenylated human serum albumin (DNP-HSA) at a young age on the development of age-related monoclonal gammapathies (MG) was investigated in a longitudinal study in intact and neonatally thymectomized (NTx) C57BL/KaLwRij and CBA/BrARij mice. Three-month-old mice were immunized four times in monthly intervals with DNP-HSA. Control mice received saline and adjuvant only. Mice immunized with DNP-HSA responded with heterogeneous antibodies, occasionally with some clonal dominance. The antibody levels further declined and were hardly detectable when the mice

Participation of cyclophosphamide-resistant T cells in murine lymphocytic choriomeningitis.

A severe inflammatory process can be induced by transferring lymphocytic choriomeningitis virus (LCMV)-immune spleen cell populations into cyclophosphamide (Cy)-suppressed, LCMV-infected recipients. The extent of cellular invasion can be quantified accurately by counting cells obtained from the cisterna magna. The underlying lymphocyte-target cell interactions are apparently very specific, with massive cellular invasion being dependent on the donor and recipient sharing class I major histocompatibility complex glycoproteins. Unlike the situation found by others for delayed-type hypersensitivit

Exacerbation of murine cutaneous leishmaniasis by adoptive transfer of parasite-specific helper T cell populations capable of mediating Leishmania major-specific delayed-type hypersensitivity.

The effect of adoptive transfer of in vitro-propagated Leishmania major-specific T cell populations on the course of experimentally induced cutaneous leishmaniasis was studied in mice. The L. major-specific T cells expressed the T helper/inducer phenotype and were able in vitro to a) mount a specific proliferative response, b) provide specific helper activity for antibody responses, c) activate parasitized macrophages resulting in L. major destruction, and d) secrete macrophage-activating factors as tested in a tumoricidal assay. These T cells were also found capable of transferring parasite-s

Immunological regulation of experimental cutaneous leishmaniasis. IV. Prophylactic effect of sublethal irradiation as a result of abrogation of suppressor T cell generation in mice genetically susceptible to Leishmania tropica.

The overwhelming susceptibility of BALB/c mice to infection with Leishmania tropica can be substantially reversed by immediately prior sub-lethal irradiation. This is related to radiation dosage, and at 550 rad, causes 60 percent complete cures and only 19 percent (instead of 100 percent) incidence of progressive disease. Irradiation 10 d before infection is only weakly prophylactic, whereas 10 d after is without effect. Control of lesion development is only apparent after the first 30 d, coincident with the analogous onset previously found in resistant strains and adult thymectomized, x-irrad

Fine-specificity of the immune response to oxazolone. I. Contact sensitivity and early antibodies.

The specificity of the contact sensitivity induced by various chemically related oxazolones was studied. Mice were painted with 1 of them (immunogen). They were divided into several groups and challenged (ear painting on day 5) with various concentrations of either the immunogen or a related oxazolone. The degree of the contact sensitivity was estimated by ear weight or by incorporation of a DNA precursor. Higher concentrations of a challenge compound elicited a stronger DTH reaction than lower concentrations. The efficiency of each compound was defined by the concentration that caused a 30% i

[Effect of abrogating hybrid resistance on the survival of radiation chimeras].

A study was made of the effect of the hybrid resistance abrogation by means of the lymphoid cell administration on the survival of the lethally irradiated mice protected by the transplantation of the semiallogeneic bone marrow. Injection to the C57BLxCBA recipients of the C57BL lymphoid cells one day before the irradiation and the transplantation of the bone marrow of the same genotype (C57BL) increased the chimera survival in comparison with the untreated recipients; such pretreatment 7 days before the irradiation decreased the chimera survival. Vitamin D receptor in osteoblasts is a negative regulator of bone mass control.

The physiological and beneficial actions of vitamin D in bone health have been experimentally and clinically proven in mammals. The active form of vitamin D [1α,25(OH)(2)D(3)] binds and activates its specific nuclear receptor, the vitamin D receptor (VDR). Activated VDR prevents the release of calcium from its storage in bone to serum by stimulating intestinal calcium absorption and renal reabsorption. However, the direct action of VDR in bone tissue is poorly understood because serum Ca(2+) homeostasis is maintained through tightly regulated ion transport by the kidney, intestine, and bone. I

BDNF-enriched small extracellular vesicles protect against noise-induced hearing loss in mice.

Noise-induced hearing loss (NIHL) is one of the most prevalent acquired sensorineural hearing loss etiologies and is characterized by the loss of cochlear hair cells, synapses, and nerve terminals. Currently, there are no agents available for the treatment of NIHL because drug delivery to the inner ear is greatly limited by the blood-labyrinth barrier. In this study, we used mesenchymal stem cell-derived small extracellular vesicles (MSC-sEVs) as nanoscale vehicles to deliver brain-derived neurotrophic factor (BDNF) and evaluated their protective effects in a mouse model of NIHL. Following int

Low Intensity Noise Exposure Enhanced Auditory Loudness and Temporal Processing by Increasing Excitability of DCN.

Sound stimulation is generally used for tinnitus and hyperacusis treatment. Recent studies found that long-term noise exposure can change synaptic and firing properties in the central auditory system, which will be detected by the acoustic startle reflex. However, the perceptual consequences of long-term low-intensity sound exposure are indistinct. This study will detect the effects of moderate-level noise exposure (83 dB SPL) on auditory loudness, and temporal processing was evaluated using CBA/CaJ mice. C-Fos staining was used to detect neural activity changes in the central auditory pathway

Changes in glucosylceramide structure affect virulence and membrane biophysical properties of Cryptococcus neoformans.

Fungal glucosylceramide (GlcCer) is a plasma membrane sphingolipid in which the sphingosine backbone is unsaturated in carbon position 8 (C8) and methylated in carbon position 9 (C9). Studies in the fungal pathogen, Cryptococcus neoformans, have shown that loss of GlcCer synthase activity results in complete loss of virulence in the mouse model. However, whether the loss of virulence is due to the lack of the enzyme or to the loss of the sphingolipid is not known. In this study, we used genetic engineering to alter the chemical structure of fungal GlcCer and studied its effect on fungal growth

The influence of opioids on the humoral and cell-mediated immune responses in mice. The role of macrophages.

BACKGROUND: Our experiments were aimed to test the influence of treatment with different opioids (morphine, fentanyl, methadone) on the humoral and cell-mediated immune responses. METHODS: Mice were treated intraperitoneally (ip) with opioids for several days and next either immunized with sheep red blood cells (SRBC) to test the antibody production or skin-sensitized with hapten picryl chloride (PCL) to induce contact hypersensitivity (CHS). In addition, the effects of opioids on the production of reactive oxygen intermediates (ROIs) and cytokines by peritoneal macrophages (Mf) and on the ex
