OBJECTIVE: Occupational exposure to ionizing radiation is a known risk factor for genomic instability. This study evaluated whether physicians occupationally exposed to ionizing radiation exhibit higher frequencies of chromosomal alterations compared to unexposed hospital workers, utilizing micronuclei and sister chromatid exchanges as biomarkers.
METHODS: Peripheral blood samples were analyzed from 50 participants: 25 physicians exposed to ionizing radiation (radiologists) and a control group of 25 sex- and age-matched hospital staff with no history of occupational exposure. The cytokinesis-

The reported relative biological effectiveness (RBE) for thermal neutrons has a large range (5-51, for cytogenetic endpoints), which can confound radiation protection decision-making. To determine whether thermal neutron spectra can influence RBE, the RBE of reactor-derived thermal neutrons of average energy 31 meV was evaluated in human peripheral blood lymphocytes using two classical DNA double-strand break endpoints: the dicentric chromosome assay (DCA) and the cytokinesis-block micronucleus assay. Dose-response curves for 41 to 408 mGy revealed a preference for linear regression. Maximum R

BACKGROUND: Micronuclei (MN), which are chromosome fragments, are formed after exposure to ionizing radiation. Radiation-induced MN is currently used as a quantitative indicator of the chromosomal aberrations detectable at a relatively early phase (e.g., within one cell-cycle progression). Meanwhile, the MN formation assay is also used to evaluate radiosensitivity (e.g., cell-killing). As such, the technique to assay the MN formation has been followed with increasing interest. However, the meaning of MN and the corresponding cellular responses remains uncertain.
PURPOSE: This study presents a

With the development and applications of ionizing radiation in medicine, the radiation effects on human health get more and more attention. Ionizing radiation can lead to various forms of cytogenetic damage, including increased frequencies of micronuclei (MNi) and chromosome abnormalities. The cytokinesis block micronucleus (CBMN) assay is widely used method for measuring MNi to determine chromosome mutations or genome instability in cultured human lymphocytes. The visual scoring of MNi is time-consuming and scorer fatigue can lead to inconsistency. In this work, we designed software for the s

The Santiago River is one of the most contaminated rivers in Mexico, with heavy metal levels above the allowed limits. Scientific evidence indicates that chronic heavy metal exposure leads to cytogenotoxic effects. The aims of this study were to evaluate the genotoxic and cytotoxic effects of such exposure in buccal mucosa cells by micronucleus (MN) assay and to identify other nuclear abnormalities (NAs), such as nuclear buds (NBUDs), binucleated cells (BNs), pyknotic nuclei (PNs), karyorrhexis (KX), karyolysis (KL), and abnormally condensed chromatin (CC). Assays were performed on samples fro

The dicentric chromosome assay (DCA) is the gold-standard assay for accurately estimating unknown radiological doses to individuals following radiological or nuclear accidents. However in a mass-casualty scenario, this assay is not well suited for providing timely dose estimates due to its time- and expertise-intensive nature. In Canada, two approaches are being developed in an attempt to increase triage-quality biological dosimetry throughput. These are 1) increasing the number of trained personnel capable of conducting the DCA, and 2) evaluating alternative biodosimetry approaches or DCA var

In recent years, a number of events have occurred that highlight the necessity of being prepared for a possible large-scale radiological event. An important question is how well are European Union (EU) Member States prepared to cope with mass radiological casualties. A survey to establish the current status of biological dosimetry across the EU was carried out with the aim of assessing capacity to perform biodosimetric triage of accident victims. Information was sought from the radiation protection authorities of the 27 Member States plus Switzerland and Norway. Biology dosimetry is establishe

Crystalline silica has been classified as a human carcinogen, but there is still considerable controversy regarding its fibrogenic and carcinogenic potential. In the present study, we investigated the genotoxic potential of bentonite particles (diameter < 10 microm) with an a-quartz content of up to 6% and different chemical modifications (alkaline, acidic, organic). Human lung fibroblasts (IMR90) were incubated for 36 h, 48 h, or 72 h with bentonite particles in concentrations ranging from 1 to 15 microg/cm2. Genotoxicity was assessed using the micronucleus (MN) assay and kinetochore analysis

Higenamine is a catecholic benzyltetrahydroisoquinoline alkaloid occurring in plant-derived foods and food supplements, the latter potentially resulting in substantially higher exposure levels than conventional foods. Because of its catechol and alkylphenol structures, oxidative conversion to electrophilic quinone or quinone methide intermediates is plausible, raising concern about a potential genotoxic hazard. However, experimental data on the genotoxic potential of higenamine are lacking. The present study, therefore, investigated its potential to induce gene mutations and chromosomal damage

The development of industrial operations has led to a significant rise in human contact with environmental toxins, especially for individuals who live close to steel industries and petroleum refineries. Public health is seriously threatened by the potential for genetic impairment from continued exposure to these emissions. Chromosomal aberration (CA) and micronucleus (MN) assays were used in this study as reliable biological markers to assess DNA damage in people who lived close to a steel factory and an oil refinery in Iraq's Erbil Province. Blood and buccal samples were collected from 50 exp

This study aimed to investigate the impact of chronic low-level exposure to chemical carcinogens with different modes of action on the cellular response to ionising radiation. Human lymphoblastoid GM1899A cells were cultured in the presence of 4-nitroquinoline N-oxide (4NQO), N-nitroso-N-methylurea (MNU) and hydrogen peroxide (H2O2) for up to 6 months at the highest non-(geno)toxic concentration identified in pilot experiments. Acute challenge doses of 1 Gy X-rays were given and chromosome damage (dicentrics, acentric fragments, micronuclei, chromatid gaps/breaks) was scored. Chronic exposure
