A 37-year-old woman with scapulohumeral muscular atrophy, rigid spine and cardiopathy is reported. Muscle weakness, advanced atrioventricular block and contractures at the neck, elbows and ankles had occurred during her childhood. An autosomal dominant mode of inheritance was suggested because her mother, sister and brother had the same disorder. Pleomorphic mitochondria had accumulated in the subsarcolemmal space of the skeletal muscle. There was no evident enzyme defect in the mitochondrial electron transport system. Although the clinical features had some similarity with those of Emery-Drei

A 7-day-old girl with congenital hypotonia and unexplained episodes of bradycardia had a broad spectrum of similar skeletal muscle and myocardial degenerative ultrastructural abnormalities. Ultrastructural studies showed obliteration of cross striations, myofilament disorganization, streaming, smearing, clumping, and zigzag Z-band deformities. A decrease in glycogen, mitochondria, and T-tubular system occurred in the regions showing Z-band abnormalities of both skeletal muscle and myocardium. Concurrent structural cardiomyopathy should be considered in patients with congenital myopathies, part

An 8-year-old boy underwent general anesthesia and experienced an episode of malignant hyperthemia, characterized by elevated temperature, cardiac arrhythmias, markedly elevated serum enzymes (SGOT, LDH, and CPK), proteinuria, and hemoglobinuria. Sixty-six days after anesthesia a skeletal muscle biopsy was obtained for examination by electron and light microscopy, which showed skeletal muscle cells with abnormally numerous mitochondria, enlarged, and variable in shape. Some contained abnormal cristae. There were more lysosomes than normal, and lipofuscin was increased in quantity. Myelin-like

Primary and secondary overacting inferior oblique muscles were examined by light and electron microscopy and compared to normal inferior oblique muscles. The most conspicuous light microscopic difference between primary overacting and normal muscles was the presence of granular fibers in overacting muscles that contained sarcoplasmic masses. Some primary and secondary overacting muscles also contained fibers displaying increased vacuolization and atrophy. Other secondary overacting muscles showed only numerous fibers at various stages of atrophy. The affected fibers were distributed throughout

The skeletal muscle, one of the largest tissues in mammals, plays a crucial role in maintaining body movement and energy metabolism. Dysfunction or damage to the skeletal muscle can lead to various muscle diseases, such as muscular dystrophy, myasthenia, and others. The myofiber presents the fundamental structural unit of the skeletal muscle, and research on its structure and biological function is of great significance. Here, we review the latest progress in the structural and functional aspects of the myofiber, focusing on myofibril, the sarcoplasmic reticulum, mitochondria, and the cytoskel

The ultrastructural findings in a muscle biopsy of a patient with Cushing's disease associated with severe myopathy are described. They included pronounced mitochondrial damage, thickening and deep invaginations of the sarcolemmal basement membrane, and thickening of the basement membrane of the capillaries. In addition, the muscle fibers showed a marked degree of disarray and wide interfibrillar spaces, containing large numbers of vacuoles, which presented degenerated mitochondria. The similarity between these findings and the ultrastructural features of corticosteroid-induced myopathy is dis

In this study we report on the effect of halothane exposure on the skeletal muscle membrane and calcium localization in 9 patients with confirmed susceptibility to malignant hyperthermia and compare them with 11 non-susceptible cases of malignant hyperthermia. Two normal muscle specimens with physiologically induced muscle contractures were also included for comparison. Halothane exposure in malignant hyperthermia-susceptible cases showed severe muscle contractures, definite breaks in the plasma membrane with peroxidase penetration, and large number of calcium granules within the mitochondria

Obesity involves positive energy balance and mitochondrial dysfunction. Mitochondria play a vital role in reshaping tissues by oxidizing substrates; however, tissue-specific organellar plasticity during energy deprivation remains poorly characterized in obesity. We investigated the effects of 24-h fasting on mitochondrial dynamics in adipose tissue, liver, and muscle of lean and diet-induced obese (DIO) male C57BL/6 mice. Fasting reduced body weight by 13% and subcutaneous adipose tissue (SAT) by 40% in lean mice, but only 6.4% in DIO mice, which maintained SAT mass. Indirect calorimetry revea

Mass-specific metabolic rate negatively co-varies with body mass from the whole-animal to the mitochondrial levels. Mitochondria are the mainly consumers of oxygen inspired by mammals to generate ATP or compensate for energetic losses dissipated as the form of heat (proton leak) during oxidative phosphorylation. Consequently, ATP synthesis and proton leak compete for the same electrochemical gradient. Because proton leak co-varies negatively with body mass, it is unknown whether extremely small mammals further decouple their mitochondria to maintain their body temperature or whether they imple

Ullrich congenital muscular dystrophy and Bethlem myopathy are caused by mutations in collagen VI (ColVI) genes, which encode an extracellular matrix protein; yet, mitochondria play a major role in disease pathogenesis through a short circuit caused by inappropriate opening of the permeability transition pore, a high-conductance channel, which causes a shortage in ATP production. We find that melanocytes do not produce ColVI yet they bind it at the cell surface, suggesting that this protein may play a trophic role and that its absence may cause lesions similar to those seen in skeletal muscle.

Macrocyclic nanocompounds, particularly calix[4]arenes, are attracting growing interest in biomedical and molecular biotechnology research. Calix[4]arene C-715, functionalized with trifluoroacetamide groups on the upper rim, does not affect Ca2+ transport systems of the plasma membrane or sarcoplasmic reticulum, nor the ATPase activity of the myosin head, suggesting a selective action on mitochondrial Ca2+ transport. This study investigated the effects of C-715 on electron transport chain activity, Ca2+ handling, and nitric oxide production in isolated mitochondria of uterine smooth muscle, al

Intrauterine growth restriction (IUGR) of the fetus, resulting from placental insufficiency (PI), is characterized by low fetal oxygen and nutrient concentrations that stunt growth rates of metabolic organs. Numerous animal models of IUGR recapitulate pathophysiological conditions found in human fetuses with IUGR. These models provide insight into metabolic dysfunction in skeletal muscle and liver. For example, cellular energy production and metabolic rate are decreased in the skeletal muscle and liver of IUGR fetuses. These metabolic adaptations demonstrate that fundamental processes in mitoc

Utilizing human-induced pluripotent stem-cell-derived vascular smooth muscle cells (hiPSC-VSMCs) to address seed cell source in tissue-engineered vascular grafts necessitates promoting cellular maturation. Morphological changes and functional remodeling of mitochondria are crucial to cellular differentiation and maturation. However, our current understanding of these changes during the transition from hiPSCs to VSMCs is limited. Our findings revealed that after hiPSC-VSMC differentiation, mitochondria transitioned from perinuclear to cytoplasmic distribution, increased in number, and changed s

A six-month-old boy with mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes (MELAS) syndrome developed repeated crops of purpuric macules on his soles and palms, which were histologically identified as hemorrhage into the dermis without inflammatory infiltrates. Transmission electron microscopy of the skin eruptions revealed various stages of endothelial degeneration in the dermal capillaries associated with consequent extravasation of erythrocytes. The degenerative change was characterized by swollen and vacuolated mitochondria which showed disintegration of the

Duchenne muscular dystrophy (DMD) is an X-linked recessive disorder characterized by progressive muscle degeneration and weakness due to mutations in the dystrophin gene. The symptoms of DMD share similarities with those of accelerated aging. Recently, hydrogen sulfide (H2S) supplementation has been suggested to modulate the effects of age-related decline in muscle function, and metabolic H2S deficiencies have been implicated in affecting muscle mass in conditions such as phenylketonuria. We therefore evaluated the use of sodium GYY4137 (NaGYY), a H2S-releasing molecule, as a possible approach

The mitochondrial function in skeletal muscle biopsies of three patients with chronic progressive external ophthalmoplegia, having deletions of the mitochondrial DNA, was studied by laser-excited fluorescence measurements of NAD(P)H and flavoproteins in saponin-skinned fibers. We detected substantially elevated steady state redox states of the mitochondrial NAD-system in the muscle fibers of these patients. Moreover, the respiratory chain-linked autofluorescence changes in the muscle fibers of these patients were larger in comparison to controls indicating substantial alterations of the mitoch

Of 185 patients with myopathy, 22 showed abnormal muscle mitochondria. In 12 of the 22 patients, all of whom had ocular myopathy or the ophthalmoplegia-plus syndrome, muscle biopsies contained 5%-25% "ragged red" fibers. In 4 patients with a facioscapulohumeral distribution of weakness, ragged red fibers were less numerous (3%-8%). In both groups, routine histology showed almost normal muscle. The remaining 6 patients were clinically heterogeneous, all without ptosis or ophthalmoplegia. The biopsies of three of these patients showed severely affected muscle. It is possible that mitochondrial c

Ocular myopathies are manifested by primary and progressive involvement of extraocular muscles. In most cases of involvement of extra-ocular muscles a biopsy from somatic muscles studied by histochemistry and electron microscopy permits to make the diagnosis of the underlying condition. The two main clinico-pathological types of ocular myopathies are the oculocraniosomatic syndrome (Kearns-Sayre syndrome) and oculopharyngeal muscular dystrophy. The oculocraniosomatic syndrome is a multisystemic disorder and its histopathological hallmark is the presence of ragged-red muscle fibres which contai

A male with mitochondrial myopathy, encephalopathy, lactic acidemia, and strokelike episodes is reported. He had also recurrent episodes of ileus. Muscle biopsy revealed ragged-red fibres. The cytochemistry of cytochrome c oxidase (CCO) showed scattered nonstained fibres, while all muscle fibres were heavily stained by immunocytochemistry using CCO antibody. These findings suggest that partical CCO deficiency may be present in the skeletal muscles of the patient. NADH cytochrome c reductase in the patient's muscle mitochondria was low compared with normal controls (about 26%), although succina

A 3-month-old female infant had profound generalized weakness, de Toni-Fanconi-Debre syndrome, and lactic acidosis. She required assisted ventilation and died at the age of 8 months. Muscle biopsy showed accumulation of mitochondria, glycogen, and lipid droplets. Histochemical reaction and immunocytochemical stain for cytochrome c oxidase showed very weak results, but both reactions were normal in intrafusal fibers of the muscle spindle. In crude extracts of the patient's muscle, cytochrome c oxidase activity was undetectable and enzyme-linked immunosorbent assay showed decreased reaction at a
