Properties of composite time to first event versus joint marginal analyses of multiple outcomes.

Many clinical studies (eg, cardiovascular outcome trials) investigate the effect of an intervention on multiple event time outcomes. The most common method of analysis is a so-called "composite" analysis of a composite outcome defined as the time to the first component event. Other approaches have been proposed, including the win ratio (or win difference) for ordered outcomes and the application of the Wei-Lachin test. Herein, we assess the influence of the marginal and joint distributions of the component events, and their correlation structures, on the operating characteristics of these meth

Recurrence Quantification for the Analysis of Coupled Processes in Aging.

OBJECTIVES: Aging is a complex phenomenon, with numerous simultaneous processes that interact with each other on a moment-to-moment basis. One way to quantify the interactions of these processes is by measuring how much a process is similar to its own past states or the past states of another system through the analysis of recurrence. This paper presents an introduction to recurrence quantification analysis (RQA) and cross-recurrence quantification analysis (CRQA), two dynamical systems analysis techniques that provide ways to characterize the self-similar nature of each process and the proper

Multiply robust inference of average treatment effects by high-dimensional empirical likelihood.

In this paper, we develop a multiply robust inference procedure for the average treatment effect (ATE) for data with high-dimensional covariates. We consider the case where it is difficult to correctly specify a single parametric model for the propensity scores (PS). We propose a novel high-dimensional empirical likelihood weighting method under soft covariate balancing constraints to combine multiple working PS models. An extended set of calibration functions is used, and a regularized augmented outcome regression (OR) is developed to correct the bias due to non-exact covariate balancing. Tho

One-at-a-time knockoffs.

We propose one-at-a-time knockoffs (OATK), a new methodology for detecting important explanatory variables in linear regression problems (including ridge and lasso) while controlling the false discovery rate (FDR). For each explanatory variable, OATK generates a knockoff design matrix that preserves the Gram matrix by replacing one-at-a-time only the single corresponding column of the original design matrix. OATK is a substantial relaxation and simplification of the knockoff filter, which simultaneously generates all columns of the knockoff design matrix to satisfy a much larger set of constra

Interpretation of cellular proliferation data: avoid the panglossian.

There are several statistics that may be calculated to characterize a cellular proliferation experiment. By far, the most commonly-reported statistic is the percent of cells in the final culture that have divided; however, this statistic has significant limitations. Other statistics provided by software modeling provide a much richer characterization of the biological response; however, their use also comes with caveats. Here, I discuss the practical application of these statistics, including their limitations and interdependencies, using hypothetical data. The goal of this perspective is to p

A marginalized conditional linear model for longitudinal binary data when informative dropout occurs in continuous time.

Within the pattern-mixture modeling framework for informative dropout, conditional linear models (CLMs) are a useful approach to deal with dropout that can occur at any point in continuous time (not just at observation times). However, in contrast with selection models, inferences about marginal covariate effects in CLMs are not readily available if nonidentity links are used in the mean structures. In this article, we propose a CLM for long series of longitudinal binary data with marginal covariate effects directly specified. The association between the binary responses and the dropout time i

Marginally Interpretable Spatial Logistic Regression With Bridge Processes.

In including random effects to account for dependent observations, the odds ratio interpretation of logistic regression coefficients is changed from population-averaged to subject-specific. This is unappealing in many applications, motivating a rich literature on methods that maintain the marginal logistic regression structure without random effects, such as generalized estimating equations. However, for spatial data, random effect approaches are appealing in providing a full probabilistic characterization of the data that can be used for prediction. We propose a new class of spatial logistic

Statistical analysis of Likert-based ordinal scales: a guide for clinical trialists.

BACKGROUND: Likert-based scales are a popular tool in clinical trials for assessing patient-reported outcomes. A key analytical decision involves whether to treat these data as binary, continuous, or ordinal. Each approach has implications for statistical power, bias, and interpretation of the results. In this report, we examine methods for evaluating Likert scales, with a particular focus on ordinal approaches, including win probability methods and proportional odds models. METHODS: We examined the use of proportional odds logistic regression, win probability estimation, dichotomisation with

A Primer on Inference and Prediction With Epidemic Renewal Models and Sequential Monte Carlo.

Renewal models are widely used in statistical epidemiology as semi-mechanistic models of disease transmission. While primarily used for estimating the instantaneous reproduction number, they can also be used for generating projections, estimating elimination probabilities, modeling the effect of interventions, and more. We demonstrate how simple sequential Monte Carlo methods (also known as particle filters) can be used to perform inference on these models. Our goal is to acquaint a reader who has a working knowledge of statistical inference with these methods and models and to provide a pract

Transportable inference using target population summary statistics under covariate shift.

Transporting findings from a study population to a target population is central to evidence-based decision-making in real-world settings. Most existing methods require individual-level data from both populations to account for covariate shift. However, privacy regulations and data-sharing constraints often preclude access to such data from the target population, leaving only covariate summaries available for analysis. In this paper, we develop transportability methods that enable valid inference using source individual-level data and target covariate summaries. First, we apply entropy balancin

The many reliabilities of psychological dynamics: An overview of statistical approaches to estimate the internal consistency reliability of intensive longitudinal data.

[Correction Notice: An Erratum for this article was reported in Vol 31(2) of Psychological Methods (see record 2027-65824-001). The authors discovered an error in the computation of the reliability coefficients according to the 2RDM. Upon correcting this, several results required minor updates. Specifically, the corrections affect a couple of sentences in the Results and Discussion sections, Figure 3, and the corresponding parts of the Supplementary Material. While the overall conclusions of the paper remain unchanged, the article has been updated to accurately reflect the corrected computatio

Best practices for your confirmatory factor analysis: A JASP and lavaan tutorial.

Confirmatory factor analysis (CFA) is a fundamental method for evaluating the internal structural validity of measurement instruments. In most CFA applications, the measurement model serves as a means to an end rather than an end in itself. To select the appropriate model, prior validity evidence is crucial, and items are typically assessed on an ordinal scale, which has been used in the applied social sciences. However, textbooks on structural equation modeling (SEM) often overlook this specific case, focusing on applications estimable using maximum likelihood (ML) instead. Unfortunately, sev

Monitoring mortality at interim analyses while testing a composite endpoint at the final analysis.

Mortality is often used as the clinical endpoint in clinical trials for acute diseases and takes precedence over any other outcome. A composite outcome such as death plus disease occurrence (or recurrence) or death plus hospitalization may also be considered, sometimes even as the primary outcome due to practical sample size issues. That is, a composite endpoint should have a higher event rate and thus a smaller sample size than for mortality alone to reach the same power. Two different scenarios are considered: in Scenario 1, the composite outcome is the primary endpoint and the mortality out

Targeted maximum likelihood estimation for psychological research: From causal identification to statistical inference.

Causal questions have long been central to psychological research, particularly in randomized experiments, while formal causal-inference methods are increasingly being applied to observational and quasi-experimental data. Common outcome-regression and propensity-score approaches can be sensitive to nuisance-model misspecification, whereas flexible machine learning alone does not ensure valid target-parameter estimation or inference. Targeted maximum likelihood estimation (TMLE) addresses this estimation problem by combining outcome and treatment information through an efficient-influence-funct

A Bayesian transfer learning model for mixed-effects longitudinal data analysis.

The analysis of contemporary longitudinal data problems often involves high-dimensional measurements of time-course data collected on a small number of observations. The estimation of such models with limited sample sizes can pose substantial challenges and lead to highly variable estimates, unstable predictions, and low power. In such instances, it is natural to borrow information from additional datasets with similar, although not necessarily identical, covariate-outcome relations to improve inference in the target population. We develop a novel Bayesian transfer learning model for longitudi

Regression Discontinuity Analysis with Latent Variables.

A regression discontinuity (RD) design is often employed to provide causal evidence when the randomization of the treatment assignment is infeasible. When variables of interest are latent constructs measured by observed indicators, the conventional RD analysis using observed variable scores does not allow researchers to examine heterogeneity in the estimated local average treatment effect (ATE) and to generalize the ATE to participants away from the cutoff. We propose a novel methodological augmentation to the conventional RD analysis, which assumes the availability of multiple indicator varia

Mixture-based gatekeeping procedures for multiplicity problems with multiple sequences of hypotheses.

Complex multiplicity problems arise in drug development programs with several sets of clinical objectives. This article considers a common setting with two sources of multiplicity induced by the analysis of multiple dose levels based on ordered endpoints. This results in multiplicity problems with multiple sequences of null hypotheses of no effect. Type I error rate inflation in problems of this type is typically addressed by using gatekeeping procedures that account for the hierarchical structure of the trial objectives. A general method for building gatekeeping procedures, known as the mixtu

Enhancing the Jacquez k nearest neighbor test for space-time interaction.

The Jacquez k nearest neighbor test, originally developed to improve upon shortcomings of existing tests for space-time interaction, has been shown to be a robust and powerful method of detecting interaction. Despite its flexibility and power, however, the test has three main shortcomings: (i) it discards important information regarding the spatial and temporal scales at which the detected interaction takes place; (ii) the results of the test have not been visualized; and (iii) recent research demonstrates the test to be susceptible to population shift bias. This study presents enhancements to

A Bayesian Parametric and Nonparametric Approach for the Imputation of Multivariate Left-Censored Data Due to Limit of Detection.

Left-censored observations due to limits of detection and/or quantification are common in clinical and epidemiologic research when continuous predictors are assessed from human specimens. In these settings, values below a certain threshold are not detectable in laboratory analysis and are reported as missing in the dataset. Classical imputation approaches have mostly relied on imputing the same number for all non-detected samples, thus compromising the continuous nature of the censored variables and affecting their variability and potential inclusion in regression modeling. Continuous imputati
