The present review has extended the general theory of motion sickness proposed by Wood and Graybiel [135, 136] by identifying specific neurophysiological mechanisms that are involved in motion sickness and by interpreting the actions of both scopolamine and amphetamine as effective anti-motion sickness drugs within this neurophysiological context. The neurochemical and neurophysiological effects of scopolamine have been reviewed in relationship to central cholinergic pathways. Cholinergic pathways have been associated with both the perception and expression of normal and excessive levels of mo…

The incidence of motion sickness in a large (N = 3,618) college population was determined by means of a questionnaire. The test-retest reliability coefficient (0.84) suggested that the motion sickness questionnaire (MSQ) for this particular population yields highly consistent information. Significantly greater proportions of men than women had low susceptibility scores and, conversely, significantly greater proportions of women had high susceptibility scores. The relationships between motion sickness susceptibility scores and several behavioral characteristics in men and women were also invest…

A double-blind placebo-controlled study compared the efficacy of the antimotion sickness drug scopolamine when administered by oral or transdermal routes. A secondary purpose was to extend our bioassay involving fixed-dose combinations of the homergic drugs promethazine and ephedrine. After receiving 12 apparently identical drug-placebo treatments, eight normal male students were exposed to a slow rotation room to stressful accelerations generated by their execution of 40 head movements out of the plane of the room's rotation of 1 rpm and at 1-rpm increments until either symptoms were experien…

BACKGROUND: Sensory input mismatches among the vestibular system, autonomic control, and visual perception cause motion sickness. Anticholinergics and antihistamines are commonly used but have limited efficacy and cause significant side effects. Orexin-A, a hypothalamic neuropeptide, has recently garnered attention for its potential role in controlling motion sickness.
OBJECTIVE: To summarize current knowledge on the effects and mechanisms of orexin-A in reducing motion sickness, identify gaps, and propose future research directions.
METHODS: Five qualified animal experiments were identified…

BACKGROUND: Individuals seem to be differently susceptible to motion-related sickness (motion sickness, visually induced sickness etc.). Investigations of the reasons for these different susceptibilities have revealed many potential factors that could predict individual susceptibility to motion-related sickness.
OBJECTIVE: This paper attempts to conduct a comprehensive literature review on inter-individual predictors of susceptibility to motion-related sickness using systematic approaches.
METHODS: After a systematic literature research, titles and abstracts of 1778 publications were screene…

The postural instability theory of motion sickness predicts that subjective symptoms of motion sickness will be preceded by unstable control of posture. In previous studies, this prediction has been confirmed with measures of the spatial magnitude and the temporal dynamics of postural activity. In the present study, we examine whether precursors of visually induced motion sickness might exist in postural time-to-contact, a measure of postural activity that is related to the risk of falling. Standing participants were exposed to oscillating visual motion stimuli in a standard laboratory protoco…

This randomized cross-over study tested the hypothesis that exposure to short-wavelength light induces symptoms of motion sickness (MS). The study participants were 28 healthy adults (14 women; mean age±SD, 25.96±3.11 years). Two stimuli oscillating within a range of 0.4-0.6 Hz were used to induce MS: a blue wave stimulus with short-wavelength light (460 nm) and a green wave stimulus with middle-wavelength light (555 nm). All participants were exposed to both stimuli throughout two separate periods. After a baseline period, participants were exposed to each stimulus three times for 4 min. The …

It has been theorized that motion sickness evolved as a negative reinforcement system which terminates motion involving postural instability and/or sensory conflict. A hypothetical example is provided by a "thought experiment" whereby protohominids are in a tree looking for food. Selection pressure results when the organisms that have an aversion to motion-producing sensory conflict do not venture out too far on the tree limbs and therefore tend to survive. In order to support an evolutionary model for motion sickness there must be evidence for genetic and/or heritable predisposition. The pres…

BACKGROUND: Exposure to unfamiliar motion patterns commonly results in motion sickness and a false perception of motion, termed mal de debarquement, on the return to stable conditions.
OBJECTIVE: To investigate whether motion sickness severity is correlated with the duration and severity of mal de debarquement; to study the possible preventive effect of projecting earth-referenced scenes (an artificial horizon) during exposure to motion on the development of mal de debarquement.
METHODS: Thirty subjects were exposed to the recorded motion profile of a boat in a 3-degrees-of-freedom ship moti…

INTRODUCTION: Severe motion sickness is easily identifiable with sufferers showing obvious behavioral signs, including emesis (vomiting). Mild motion sickness and sopite syndrome lack such clear and objective behavioral markers. We postulate that yawning may have the potential to be used in operational settings as such a marker. This study assesses the utility of yawning as a behavioral marker for the identification of soporific effects by investigating the association between yawning and mild motion sickness/sopite syndrome in a controlled environment.
METHODS: Using a randomized motion-coun…

Auditory cues can create the illusion of self-motion (vection) in the absence of visual or physical stimulation. The present study aimed to determine whether auditory cues alone can also elicit motion sickness and how auditory cues contribute to motion sickness when added to visual motion stimuli. Twenty participants were seated in front of a curved projection display and were exposed to a virtual scene that constantly rotated around the participant's vertical axis. The virtual scene contained either visual-only, auditory-only, or a combination of corresponding visual and auditory cues. All pa…

Motion sickness (MS) has been identified as a non-thermal factor that can moderate autonomic thermoregulatory responses. It has been shown that MS exaggerates core cooling during immersion in cold (15 °C) and luke-warm (28 °C) water by attenuating cold-induced vasoconstriction. The aim of the present study was to investigate whether MS affects thermal balance in a thermoneutral air environment. Eleven subjects were exposed to rotation in two conditions, control (CN) and MS. In the CN condition subjects refrained from head movements, whereas in the MS condition they performed a sequence of maxi…

BACKGROUND: Limited evidence suggests that rizatriptan given before vestibular stimulation reduces motion sickness in persons with migraine-related dizziness. The present study was designed to test whether rizatriptan is also effective in protecting against visually-induced motion sickness and to test whether rizatriptan blocks the augmentation of motion sickness by head pain.
MATERIAL AND METHODS: Using randomized double-blind, placebo-controlled methodology, 10 females, 6 with migrainous vertigo (V+) and four without vertigo (V-) received 10 mg rizatriptan or placebo two hours prior to bein…

Maropitant is a neurokinin-1 inhibitor that acts to prevent and treat vomiting by blocking stimuli to the final common pathway in the emetic centre of the brain. The field efficacy and safety of a single oral dose of maropitant were investigated for the prevention of vomiting in dogs with a history of motion sickness resulting from transportation by car in two blinded, placebo-controlled studies. In an exploratory study designed as a two-way crossover trial with 17 dogs, 10 of the dogs given the placebo vomited during a car journey but only three of the dogs vomited under maropitant treatment.…

Tilting the head in roll to or from the upright while rotating at a constant velocity (roll while rotating, RWR) alters the position of the semicircular canals relative to the axis of rotation. This produces vertical and horizontal nystagmus, disorientation, vertigo, and nausea. With recurrent exposure, subjects habituate and can make more head movements before experiencing overpowering motion sickness. We questioned whether promethazine lessened the vertigo or delayed the habituation, whether habituation of the vertigo was related to the central vestibular time constant, i.e., to the time con…

The aim of the present study was to investigate an opioid receptor involvement in the adaptation response to motion sickness in Suncus murinus. Different groups of animals were treated intraperitoneally with either saline, morphine (0.1 and 1.0 mg/kg), naloxone (1.0, 10.0 and 5.0 mg/kg) or a combination of naloxone plus morphine in the absence or 30 min prior to a horizontal motion stimulus of 1 Hz and 40 mm amplitude. For the study of adaptation, different groups received saline on the first trial, and in subsequent trials (every 2 days) they received either saline, naloxone (1.0 and 10.0 mg/…

Model of motion sickness (MS) was induced in rats by accelerating and decelerating vertical rotation for 60 min. The contents of Ach, NA, A and 5-HT and the activity of AchE in brain and blood of rats were examined after 60 min of rotation. The results showed: NA contents and AchE activity decreased significantly while A and 5-HT increased in rat's brain. Levels of Ach, NA and A in blood were increased. The results suggested that MS may be related to the imbalance of transmitters in the central nervous system resulting in the reduction of central arousal, and the imbalance between sympathetic …
