RETINA: Reconstruction-based pre-trained enhanced TransUNet for electron microscopy segmentation on the CEM500K dataset.

Electron microscopy (EM) has revolutionized our understanding of cellular structures at the nanoscale. Accurate image segmentation is required for analyzing EM images. While manual segmentation is reliable, it is labor-intensive, incentivizing the development of automated segmentation methods. Although deep learning-based segmentation has demonstrated expert-level performance, it lacks generalizable performance across diverse EM datasets. Current approaches usually use either convolutional or transformer-based neural networks for image feature extraction. We developed the RETINA method, which

Predicting genes associated with ossification of the posterior longitudinal ligament using graph attention network.

Ossification of the posterior longitudinal ligament is a degenerative disease that severely impacts the spine, with a complex pathogenesis involving the interplay of multiple genes. This study utilizes a combination of graph neural networks and deep neural networks to systematically analyze genes associated with OPLL, leveraging genomics and bioinformatics techniques. By integrating gene data from the DisGeNET and HumanNetV2 databases, we constructed a GNN model to identify potential pathogenic genes for OPLL and validated the expression characteristics and mechanisms of these genes in differe

Adult Mouse Leptomeninges Exhibit Regional and Age-related Cellular Heterogeneity Implicating Mental Disorders.

The leptomeninges envelop the central nervous system (CNS) and contribute to cerebrospinal fluid (CSF) production and homeostasis. We analyzed the meninges overlying the anterior or posterior forebrain in the adult mouse by single nuclear RNA-sequencing (snucRNA-seq). This revealed regional differences in fibroblast and endothelial cell composition and gene expression. Surprisingly, these non-neuronal cells co-expressed genes implicated in neural functions. The regional differences changed with aging, from 3 to 18 months. Cytokine analysis revealed specific soluble factor production from anter

Applications of Neural Network-Based Plan-Cancer Method for Primary Diagnosis of Mesothelioma Cancer.

"Malignant mesothelioma (MM)" is an uncommon although fatal form of cancer. The proper MM diagnosis is crucial for efficient therapy and has significant medicolegal implications. Asbestos is a carcinogenic material that poses a health risk to humans. One of the most severe types of cancer induced by asbestos is "malignant mesothelioma." Prolonged shortness of breath and continuous pain are the most typical symptoms of the condition. The importance of early treatment and diagnosis cannot be overstated. The combination "epithelial/mesenchymal appearance of MM," however, makes a definite diagnosi

Pupal behavior emerges from unstructured muscle activity in response to neuromodulation in Drosophila.

Identifying neural substrates of behavior requires defining actions in terms that map onto brain activity. Brain and muscle activity naturally correlate via the output of motor neurons, but apart from simple movements it has been difficult to define behavior in terms of muscle contractions. By mapping the musculature of the pupal fruit fly and comprehensively imaging muscle activation at single-cell resolution, we here describe a multiphasic behavioral sequence in Drosophila. Our characterization identifies a previously undescribed behavioral phase and permits extraction of major movements by

Fusiform Activity Distinguishes Between Subjects With Low and High Xenophobic Attitudes Toward Refugees.

This study analyzes how people's attitudes to the European refugee crisis (ERC) correspond to selected psychological state and trait measures and impact the neural processing of media images of refugees. From a large pool of respondents, who filled in an online xenophobia questionnaire, we selected two groups (total N = 38) with the same socio-demographic background, but with opposite attitudes toward refugees. We found that a negative attitude toward refugees (high xenophobia - HX) was associated with a significantly higher conscientiousness score and with a higher trait aggression and hostil

Thalamic Volume Is Related to Increased Anterior Thalamic Radiations in Children with Reactive Attachment Disorder.

Reactive attachment disorder (RAD) is associated with childhood maltreatment and affects approximately 1% of the general population. Recent data suggest that childhood maltreatment is associated with brain alterations in white and gray matter. However, the neural mechanisms of RAD-related brain alterations remain unknown. Herein, we evaluated the white matter pathways and gray matter volumes in 31 and 41 age-matched children with RAD and typical development (TD), respectively, by analyzing T1- and diffusion-weighted images. An increased fractional anisotropy (FA) and axial diffusivity in the a

Cyclothymic temperament and glucose metabolism in the right superior parietal lobule.

Cyclothymic temperament possesses a central dimension that includes rapid fluctuations in mood and emotional instability, and it is regarded as a prodromal state of bipolar disorder. The aim of the present study is to explore the neural correlates of cyclothymic temperament. We used the data of 55 healthy participants in our previous study and analyzed the association between cyclothymic temperament scores rated by the Temperament Evaluation of Memphis, Pisa, Paris and San Diego-Autoquestionnaire (TEMPS-A) and the uptake of [18F]-FDG measured by positron emission tomography (PET). A previously undescribed branch of the pterygopalatine ganglion.

BACKGROUND: Endonasal and infrazygomatic pterygopalatine ganglion (PPG) block for facial pain provides pain relief in a broader area than expected on anatomic grounds. The aim of this study was to search for neural structures in the pterygopalatine fossa (PPF) that could explain unexpected pain relief after PPG blockage. METHODS: The neural PPF content was explored through human cadaver study and nerve-specific staining. Five human PPF specimens were dissected as whole-mount preparations with the aid of an operation microscope and stained for acetylcholinesterase. One of these specimens was p

[Increased expression of HOXA9 gene in Hirschsprung disease].

OBJECTIVE: Hirschsprung disease is characterized by segmental aganglionosis of the terminal bowel. Neurons of the enteric nervous system arise from neural crest, migrate and colonize intestinal muscle coat where they proliferate and differentiate. The first pathophysiologic hypothesis suggests an absence of neural cell migration. The most recent hypothesis involves disorders of their homing and/or their differentiation due to an altered intestinal microenvironment. PATIENTS AND METHODS: We analyzed the expression of muscle markers and laminin isoforms by immunocytochemistry and of homeotic ge

Neural Stem/Progenitor Cell Therapy in Patients and Animals with Amyotrophic Lateral Sclerosis: A Systematic Review and Meta-analysis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative malady that causes progressive degeneration and loss of motor neuron function in the brain and spinal cord, eventually resulting in muscular atrophy, paralysis, and death. Neural stem/progenitor cell (NSPC) transplantation can improve bodily function in animals and delay disease progression in patients with ALS. This paper summarizes and analyzes the efficacy and safety of neural stem/progenitor cell (NSPC) transplantation as a treatment for ALS, aiming to improve function and delay disease progression in patients. We present a summary

Sports tourism as a pathway to older adults' psychological well-being: a dual-pathway model integrating broaden-and-build and social identity perspectives.

This study explores how sports tourism is associated with older adults' eudaimonic well-being through dual psychological mechanisms. Drawing on Broaden-and-Build Theory and Social Identity Theory, the research examines an emotion pathway in which positive emotions and sense of physical challenge achievement are linked to psychological resources, and an identity pathway in which sports group identification is linked to positive age identity. Survey data from 467 older Chinese sports tourism participants were analyzed using PLS-SEM and artificial neural network analysis. PLS-SEM results provide

Charting the human-specific properties of gene expression networks in the infant prefrontal cortex.

Human infancy is characterized by protracted brain development coinciding with sensitive periods of extensive synaptic remodeling. Whether this is supported by human infant-specific transcriptional programs is unknown as comparative material in closely related primate species was unavailable. Here, we analyze rare newborn chimpanzee and age-matched human and rhesus macaque brain samples using single-cell transcriptomics and epigenomics. We identify a human infant-specific transcriptional program in immature oligodendrocytes that is overrepresented in autism risk genes and patient gene expressi

Single-cell transcriptomic analyses provide insights into TME related with tumor-promoting in AIDS-CNS-DLBCL.

DLBCL is a common fatal disease in AIDS patients, especially involved in CNS, due to immunity dysfunction and unknown pathogenic mechanism. We analyzed scRNA-seq data from seven AIDS patients and two immunocompetent patients. Through subgroup analysis comparing the cellular composition characteristics of different CNS-DLBCL, thereby elucidating the pathogenic drivers in distinct patient populations. In AIDS related CNS-DLBCL samples, we identified a population of tumor cells with neural characteristics, designated as neuro-signature cells. These population demonstrated high-grade malignancy, c

Denoising Diffusion Implicit Model Combined with TransNet for Rolling Bearing Fault Diagnosis Under Imbalanced Data.

Data imbalances present a serious problem for intelligent fault diagnosis. They can lead to reduced diagnostic precision, which can jeopardize equipment reliability and safety. Based on that, this paper proposes a novel fault diagnosis method combining the denoising diffusion implicit model (DDIM) with a new convolutional neural network framework. First, the Gramian angular difference field (GADF) is used to generate 2D images, which are then augmented using DDIM. Next, by utilizing the weight-sharing properties of a convolutional neural network and the self-attention mechanism along with the

Investigating brain structure and tDCS response in obsessive-compulsive disorder.

Obsessive-Compulsive Disorder (OCD) is characterized by intrusive thoughts and repetitive behaviors, with associated brain abnormalities in various regions. This study explores the correlation between neural biomarkers and the response to transcranial Direct Current Stimulation (tDCS) in OCD patients. Using structural MRI data from two tDCS trials involving 55 OCD patients and 28 controls, cortical thickness, and gray matter morphometry was analyzed. Findings revealed thicker precentral and paracentral areas in OCD patients, compared to control (p < 0.001). Correlations between cortical thickn

Correlations enhance the behavioral readout of neural population activity in association cortex.

Noise correlations (that is, trial-to-trial covariations in neural activity for a given stimulus) limit the stimulus information encoded by neural populations, leading to the widely held prediction that they impair perceptual discrimination behaviors. However, this prediction neglects the effects of correlations on information readout. We studied how correlations affect both encoding and readout of sensory information. We analyzed calcium imaging data from mouse posterior parietal cortex during two perceptual discrimination tasks. Correlations reduced the encoded stimulus information, but, see

A fingerprint of amyloid plaques in a bitransgenic animal model of Alzheimer's disease obtained by statistical unmixing analysis of hyperspectral Raman data.

The global prevalence of Alzheimer's disease (AD) points to endemic levels, especially considering the increase of average life expectancy worldwide. AD diagnosis based on early biomarkers and better knowledge of related pathophysiology are both crucial in the search for medical interventions that are able to modify AD progression. In this study we used unsupervised spectral unmixing statistical techniques to identify the vibrational spectral signature of amyloid β aggregation in neural tissues, as early biomarkers of AD in an animal model. We analyzed spectral images composed of a total of 55
