PRBS Gas Challenges Reveal Impaired Chemoreflex and Cholinergic Dynamics in MCI.

PURPOSE: This study investigates the dynamic regulation of cerebral oxygenation in individuals with mild cognitive impairment (MCI) compared to healthy controls, using a novel pseudorandom binary sequence (PRBS) gas challenge. Traditional assessments often overlook frequency-dependent features of cerebrovascular control; this work aims to uncover latent deficits through broadband perturbations and nonparametric dynamic modeling. METHODS: Seventeen ApoE4-negative participants (9 controls, 8 MCI) underwent a three-session supine protocol involving PRBS-modulated inhalation of hypoxic and hyperc

Gasotransmitters in the regulation of connexins in central and peripheral nervous system injuries.

Gasotransmitters, such as nitric oxide, carbon monoxide, and hydrogen sulfide can freely diffuse across biological membranes, bind to receptors and molecular targets, and modulate multiple intracellular signaling pathways. However, no systematic review exists regarding how gasotransmitters influence central and peripheral nervous system injury by regulating connexins. This review study presents, for the first time, a systematic analysis of the effects of these three gasotransmitters on connexin regulation, with a focus on their potential roles in injuries to the central and peripheral nervous

A programmable spatiotemporal bioelectrical conduit promotes peripheral nerve regeneration by suppressing ferroptosis through modulation of ER-mitochondria coupling in Schwann cells.

Following peripheral nerve injury, abnormal endoplasmic reticulum-mitochondria coupling and activation of ferroptosis in Schwann cells are important causes of impaired myelination and regenerative failure. Based on this, we developed a multiscale anisotropic electroactive nerve conduit. The outer layer consists of a BaTiO3-loaded GelMA piezoelectric hydrogel for ultrasound-triggered acoustic-electric conversion, while the inner layer is formed by rolling an aligned gelatin/PLA/G conductive fiber membrane into a tubular structure to provide directional conductive support and biomimetic topograp

Endothelial-to-mesenchymal Transition at the Blood-brain Barrier: Molecular Mechanisms and Pathological Roles Across Brain Diseases.

Endothelial-to-Mesenchymal Transition (EndMT) is a significant contributor to Blood- Brain Barrier (BBB) dysfunction in various brain diseases. The majority of current therapies, aimed at reducing BBB dysfunction, focus on preventing inflammation or stabilizing tight junctions. In most cases, these therapies do not provide adequate or long-lasting vascular protection. Endothelial cells undergo phenotypic programming, losing their barrier-forming capacity and developing features of mesenchymal and extracellular matrix-producing cells as the disease progresses. The change leads to chronic vascul

Modeling nitric oxide diffusion and plasticity modulation in cerebellar learning.

Nitric oxide (NO) is a versatile signaling molecule with significant roles in various physiological processes, including synaptic plasticity and memory formation. In the cerebellum, NO is produced by neural NO synthase and diffuses to influence synaptic changes, particularly at parallel fiber-Purkinje cell synapses. This study aims to investigate NO's role in cerebellar learning mechanisms using a biologically realistic simulation-based approach. We developed the NO Diffusion Simulator (NODS), a Python module designed to model NO production and diffusion within a cerebellar spiking neural netw

Uncertainty in population receptive field estimates revealed by variational qPRF.

BACKGROUND: Population receptive field (pRF) modeling is a cornerstone of retinotopic mapping in visual neuroscience, enabling precise mapping of visual stimulus processing in the human brain. However, pRF estimates are influenced by multiple sources of variability, including scanner properties, neurovascular coupling, physiological noise, and task-related factors. Traditionally, these estimates are treated as definitive because quantifying variance has been computationally infeasible. NEW METHOD: We introduce qPRF-v, an innovative extension of the qPRF software which computes pRF point estim

The life cycle of a capillary: Mechanisms of angiogenesis and rarefaction in microvascular physiology and pathologies.

Capillaries are the smallest blood vessels (<10 μm in diameter) in the body and their walls are lined by endothelial cells. These microvessels play a crucial role in nutrient and gas exchange between blood and tissues. Capillary endothelial cells also produce vasoactive molecules and initiate the electrical signals that underlie functional hyperemia and neurovascular coupling. Accordingly, capillary function and density are critical for all cell types to match blood flow to cellular activity. This begins with the process of angiogenesis, when new capillary blood vessels emerge from pre-existin

What's new on septic encephalopathy? Ten things you need to know.

Sepsis associated encephalopathy (SAE) is a frequent complication of sepsis and is associated with a higher risk of short-term mortality and long-term cognitive impairment. The EEG is a sensitive complement of the clinical examination that can also detect and quantify encephalopathy and identify features with prognostic value, such as lack of reactivity. Moreover, despite their effect on outcome is still debated, the EEG is the only tool to detect non-convulsive seizures which can occur in a septic setting. Understanding the pathophysiology of SAE is fundamental to define potential therapeutic

Vasoactive neuropeptides and Alzheimer's disease: a systematic review focusing on calcitonin gene-related peptide.

Vasoactive peptides constitute a heterogenous family of mediators exerting various physiological functions, mostly studied for their vasotropic effects and role as peripheral neurotransmitters/neuromodulators, mainly involved in nociceptive transmission modulation. They have been divided into vasodilatory or vasoconstrictive peptides, according to their predominant effects on vascular tone. Recent research has shown in the Central Nervous System effects as transmitters and "growth factor-like" signals. Therefore, deregulation of their signaling systems has been thought to play a role in neural

Spatial and temporal patterns of nitric oxide diffusion and degradation drive emergent cerebrovascular dynamics.

Nitric oxide (NO) is a gaseous signaling molecule that plays an important role in neurovascular coupling. NO produced by neurons diffuses into the smooth muscle surrounding cerebral arterioles, driving vasodilation. However, the rate of NO degradation in hemoglobin is orders of magnitude higher than in brain tissue, though how this might impact NO signaling dynamics is not completely understood. We used simulations to investigate how the spatial and temporal patterns of NO generation and degradation impacted dilation of a penetrating arteriole in cortex. We found that the spatial location of N

Regional impairment of cerebrovascular reactivity and BOLD signal in adults after stroke.

BACKGROUND AND PURPOSE: Comparative studies across populations using functional magnetic resonance imaging (fMRI) rely on a similar relationship between blood oxygen level-dependent (BOLD) signal and neural activity. However, in elderly and patients with cerebrovascular disease, impaired cerebrovascular dynamics and neurovascular coupling may explain differences in BOLD contrast across populations and brain regions. The purpose of the study was to determine whether poststroke patients have regional heterogeneities of cerebrovascular reactivity (CVR) and their potential influence on voxel-wise

Astrocytic Redox Homeostasis as a Metabolic Modulator of DNA Damage and Repair in the Ischemic Penumbra.

Ischemic stroke triggers a severe redox disequilibrium that critically shapes cell survival within the penumbra. Although oxidative DNA damage arises from excessive ROS production, the capacity to repair such lesions is tightly constrained by cellular metabolic status. Growing evidence indicates that astrocytes, key metabolic regulators of the neurovascular unit, modulate neuronal susceptibility to genomic injury through redox buffering, NAD+ maintenance, and metabolic support. In the metabolically impaired yet structurally preserved penumbra, astrocytic control of glutathione turnover, mitoch

Dietary salt promotes neurovascular and cognitive dysfunction through a gut-initiated TH17 response.

A diet rich in salt is linked to an increased risk of cerebrovascular diseases and dementia, but it remains unclear how dietary salt harms the brain. We report that, in mice, excess dietary salt suppresses resting cerebral blood flow and endothelial function, leading to cognitive impairment. The effect depends on expansion of TH17 cells in the small intestine, resulting in a marked increase in plasma interleukin-17 (IL-17). Circulating IL-17, in turn, promotes endothelial dysfunction and cognitive impairment by the Rho kinase-dependent inhibitory phosphorylation of endothelial nitric oxide syn

Neurovascular mechanisms underlying augmented cold-induced reflex cutaneous vasoconstriction in human hypertension.

KEY POINTS: In hypertensive adults (HTN), cardiovascular risk increases disproportionately during environmental cold exposure. Despite ample evidence of dysregulated sympathetic control of the peripheral vasculature in hypertension, no studies have examined integrated neurovascular function during cold stress in HTN. The findings of the present study show that whole-body cold stress elicits greater increases in sympathetic outflow directed to the cutaneous vasculature and, correspondingly, greater reductions in skin blood flow in HTN. We further demonstrate an important role for non-adrenergic

Advanced regenerative biomaterials for ischaemic stroke: a comprehensive therapeutic strategy.

Ischaemic stroke is a major global public health problem, accounting for 62.4% of all strokes, with far-reaching consequences in terms of health burden in all regions. Nonetheless, a therapeutic window that is excessively narrow, coupled with restricted therapy modalities, has hindered the majority of patients from receiving effective treatment. With the rapid advancement of biomaterials technology, stroke treatment is being propelled towards a regenerative medicine era characterized by the tripartite collaboration of "materials, drugs, and cells". This review systematically reviews the cuttin

Bessel beam optical coherence microscopy enables multiscale assessment of cerebrovascular network morphology and function.

Understanding the morphology and function of large-scale cerebrovascular networks is crucial for studying brain health and disease. However, reconciling the demands for imaging on a broad scale with the precision of high-resolution volumetric microscopy has been a persistent challenge. In this study, we introduce Bessel beam optical coherence microscopy with an extended focus to capture the full cortical vascular hierarchy in mice over 1000 × 1000 × 360 μm3 field-of-view at capillary level resolution. The post-processing pipeline leverages a supervised deep learning approach for precise 3D seg

Large-FOV, dual-region, two-photon fluorescence lifetime imaging microscopy (LD-2P-FLIM) for multiparameter and quantitative brain function imaging.

Large-scale imaging of multiple dynamic behaviors and quantitative neurochemical concentrations with high spatiotemporal resolution is essential for understanding complex brain functions. Two-photon microscopy (TPM) is ideally suited for in vivo brain function imaging because of its high resolution and deep tissue penetration. However, conventional TPM is limited by a restricted field-of-view (FOV), an inherent trade-off between the imaging area and temporal resolution, and an insufficient amount of information obtained using only intensity recording. Here, we propose large-FOV, dual-region, t

Integrative analysis of spontaneous brain activity in Parkinson's disease: associations with gene expression, cell types, and receptor density.

BACKGROUND: Parkinson's disease (PD) shows widespread alterations in intrinsic brain activity, yet the molecular and cellular bases of these disruptions remain unclear. Resting-state fMRI metrics-amplitude of low-frequency fluctuations (ALFF) and regional homogeneity (ReHo)-offer complementary views of spontaneous neural activity but have rarely been examined within an integrated biological framework. METHODS: We studied 31 PD patients and 28 healthy controls using voxel-wise ALFF and ReHo analyses combined with cortical transcriptomic data from the Allen Human Brain Atlas, cell type-specific

Cognitive Impairment and Cerebral Haemodynamics in Individuals with Symptomatic Peripheral Artery Disease.

Peripheral artery disease (PAD) is highly prevalent in older adults. PAD is associated with cognitive impairment; however the underlying mechanisms are unclear. The brain's metabolic needs rapidly change in response to neuronal activity requiring adjustment of regional cerebral blood flow, termed neurovascular coupling (NVC). The microvascular endothelium contributes to NVC responses. Patients with PAD have endothelial dysfunction that may extend to the cerebral circulation, impairing NVC and cognitive function. The present study examined NVC responses to cognitive stimulation using Transcrani

Subarachnoid hemorrhage: epidemiology, risk factors, pathogenesis, and clinical therapies.

Subarachnoid hemorrhage (SAH) is an extremely lethal cerebrovascular emergency that can lead to severe long term neurological deficits. Modern research evidence indicates that the incidence of SAH varies significantly across different regions and populations, and this variation is determined by the interaction between non-modifiable factors (such as age, gender, family history, and genetic susceptibility) and modifiable factors (such as hypertension, smoking, drug use, and metabolic disorders). Advancements in vascular biology and genomics further suggest that extracellular matrix instability,
