Ammonium removal through anaerobic ammonium oxidation coupled to iron(III) reduction along the Yangtze river-estuary continuum.

Anaerobic ammonium oxidation coupled to iron(III) reduction (Feammox) process has recently been recognized as an important pathway for removing ammonium in various natural habitats. However, our understanding for Feammox in river-estuary continuum is limited. In this study, stable isotope tracers and high-throughput amplicon sequencing were employed to determine Feammox rates and identify associated microbial communities in sediments along the Yangtze river-estuary continuum. Feammox rates averaged 0.0058 ± 0.0069 mg N/(kg·d) and accounted for approximately 22.3% of the ammonium removed from t

Impact of pesticides on marine coral reef foraminifera.

Our laboratory study looked into how pesticides affect the foraminifera species Heterostegina depressa and their obligatory algal endosymbionts. We incubated the foraminifera separately with different types of pesticides at varying concentrations (1 %, 0.01 % and 0.0001 %); we included the insecticide Confidor© (active substance: imidacloprid), the fungicide Pronto©Plus (tebuconazole), and the herbicide Roundup© (glyphosate). Our evaluation focused on the symbiont's photosynthetically active area (PA), and the uptake of dissolved inorganic carbon (DIC) and nitrogen (nitrate) to determine the v

The glutamate/aspartate transporter EAAT1 is crucial for T-cell acute lymphoblastic leukemia proliferation and survival.

T-cell acute lymphoblastic leukemia (T-ALL) is a cancer of the immune system. Approximately 20% of pediatric and 50% of adult T-ALL patients have refractory disease or relapse and die from the disease. To improve patient outcome new therapeutics are needed. With the aim to identify new therapeutic targets, we combined the analysis of T-ALL gene expression and metabolism to identify the metabolic adaptations that T-ALL cells exhibit. We found that glutamine uptake is essential for T-ALL proliferation. Isotope tracing experiments showed that glutamine fuels aspartate synthesis through the tricar

Possible evidence for care and treatment in the Tyrolean Iceman.

The Tyrolean Iceman is the world's oldest glacier mummy. He was found in September 1991 in the Italian part of the Ötztal Alps. Since his discovery a variety of morphological, radiological and molecular analyses have been performed that revealed detailed insights into his state of health. Despite the various pathological conditions found in the Iceman, little is known about possible forms of care and treatment during the Copper Age in Northern Italy. A possible approach to this topic is the presence of tattoos on the mummified body. In previous work, it was already believed that the tattoos we

Niche partitioning in sympatric Gorilla and Pan from Cameroon: implications for life history strategies and for reconstructing the evolution of hominin life history.

Factors influencing the hominoid life histories are poorly understood, and little is known about how ecological conditions modulate the pace of their development. Yet our limited understanding of these interactions underpins life history interpretations in extinct hominins. Here we determined the synchronisation of dental mineralization/eruption with brain size in a 20th century museum collection of sympatric Gorilla gorilla and Pan troglodytes from Central Cameroon. Using δ13C and δ15N of individuals' hair, we assessed whether and how differences in diet and habitat use may have impacted on a

Assignment of the orphan nuclear receptor Nurr1 by NMR.

The orphan nuclear receptor Nurr1 has been implicated in a number of conditions including Parkinson's disease and Schizophrenia. As such, it is of interest to study its interactions with other proteins, possibly mediated by small molecules, considering possible use as a drug target. We produced (2)H, (15)N, (13)C labelled-Nurr1 to generate the backbone amide NH, carbonyl C', C(alpha) and C(beta) assignments. About 84.0% of residues could be assigned. Most of the 37 missing assignments fall in 3 regions of the protein. Two of these surround a putative ligand-binding region of Nurr1, suggesting

Control of dimorphism in Mucor rouxii.

Haidle, C. W. (The University of Texas, Austin), and R. Storck. Control of dimorphism in Mucor rouxii. J. Bacteriol. 92:1236-1244. 1966.-Yeastlike cells of Mucor rouxii NRRL 1894 were converted to filaments in a medium containing glucose, mineral salts, casein hydrolysate, nicotinic acid, and thiamine when the gas phase was changed from CO(2)-N(2) or N(2) alone to air. Germ tubes began to appear 3 to 4 hr after exposure to air. Ribonucleic acid (RNA) precursors were incorporated into RNA in a discontinuous fashion during this conversion, but the incorporation was continuous during the anaerobi

Mass Spectrometric Analysis of Lewy Body-Enriched α-Synuclein in Parkinson's Disease.

Parkinson's disease (PD) is characterized by intraneuronal inclusions of aggregated α-synuclein protein (so-called Lewy bodies) in distinct brain regions. Multiple posttranslational modifications may affect the structure and function of α-synuclein. Mass spectrometry-based analysis may be useful for the characterization and quantitation of α-synuclein forms, but has proven challenging, mainly due to the insolubility of Lewy bodies in aqueous buffer. In the present study, we developed a novel method by combining differential solubilization with immunoprecipitation and targeted proteomics using

Polylactic acid microplastics increase N2O emissions from mainstream biological nitrogen removal systems: overlooked concern.

Despite being marketed as environmentally benign alternatives for conventional synthetic polymers, biodegradable plastics inevitably generate large amounts of microplastics, posing currently overlooked yet potentially significant environmental risks. This study selected polylactic acid microplastics (PLA-MPs) as model biodegradable microplastics to investigate their effects on nitrous oxide (N2O) emissions from a mainstream biological nitrogen removal system. Long-term exposure to PLA-MPs at environmentally relevant level (0.5 mg/L) increased the N2O emission factor by 28.9%, and inhibited max

Nitrate leaching losses and the fate of 15N fertilizer in perennial intermediate wheatgrass and annual wheat - A field study.

Perennial grains, such as the intermediate wheatgrass (Thinopyrum intermedium) (IWG), may reduce negative environmental effects compared to annual grain crops. Their permanent, and generally larger, root systems are likely to retain nitrogen (N) better, decreasing harmful losses of N and improving fertilizer N use efficiency, but there have been no comprehensive N fertilizer recovery studies in IWG to date. We measured fertilizer N recovery with stable isotope tracers in crop biomass and soil, soil N mineralization and nitrification, and nitrate leaching in IWG and annual wheat in a replicated

Thermal stratification-mediated DOM transformation shapes the vertical coupling-to-decoupling of carbon and nitrogen in an oxic alpine reservoir.

Owing to depth and distinctive geography, alpine reservoirs develop persistent seasonal thermal stratification that reshapes habitats and shapes carbon (C) and nitrogen (N) cycling, yet the fine-scale vertical mechanisms of C-N coupling remain unresolved. Combining FT-ICR MS, dual N-O isotopes, and multi-omic microbial analyses, we resolved biogeochemical dynamics across a full stratification cycle, targeting its stable phase to decipher C-N coupling and decoupling. In the epilimnion, intense radiation and full oxygenation sustained active dissolved organic matter (DOM) transformation; its hig

Drivers of variability in mercury and methylmercury bioaccumulation and biomagnification in temperate freshwater lakes.

The four largest freshwater lakes in southwestern France are of both ecological and economic importance. However, some of them are subjected to mercury (Hg) contamination, resulting in the ban of human consumption of piscivorous fish. Moreover, beyond predatory fish, little information exist regarding Hg levels in other species of these ecosystems. In this context, we used a food web analytical approach to investigate Hg bioaccumulation and biomagnification in relation to the trophic structure of these four lakes. More specifically, various organisms (macrophytes, epiphyton, invertebrates and

Backbone and side-chain (1)H, (15)N, (13)C assignment and secondary structure of BPSL1445 from Burkholderia pseudomallei.

BPSL1445 is a lipoprotein produced by the Gram-negative bacterium Burkholderia pseudomallei (B. pseudomallei), the etiological agent of melioidosis. Immunodetection assays against sera patients using protein microarray suggest BPSL1445 involvement in melioidosis. Herein we report backbone, side chain NMR assignment and secondary structure for the recombinant protein.

Towards Single Biomolecule Imaging via Optical Nanoscale Magnetic Resonance Imaging.

Nuclear magnetic resonance (NMR) spectroscopy is a physical marvel in which electromagnetic radiation is charged and discharged by nuclei in a magnetic field. In conventional NMR, the specific nuclei resonance frequency depends on the strength of the magnetic field and the magnetic properties of the isotope of the atoms. NMR is routinely utilized in clinical tests by converting nuclear spectroscopy in magnetic resonance imaging (MRI) and providing 3D, noninvasive biological imaging. While this technique has revolutionized biomedical science, measuring the magnetic resonance spectrum of single

Nitrite-driven anaerobic methane oxidation by oxygenic bacteria.

Only three biological pathways are known to produce oxygen: photosynthesis, chlorate respiration and the detoxification of reactive oxygen species. Here we present evidence for a fourth pathway, possibly of considerable geochemical and evolutionary importance. The pathway was discovered after metagenomic sequencing of an enrichment culture that couples anaerobic oxidation of methane with the reduction of nitrite to dinitrogen. The complete genome of the dominant bacterium, named 'Candidatus Methylomirabilis oxyfera', was assembled. This apparently anaerobic, denitrifying bacterium encoded, tra

1H, 13C, and 15N backbone resonance assignments of the A2 domain of human von Willebrand factor.

The A2 domain of von Willebrand factor (vWF A2) acts as a mechanosensor, unfolding under shear stress to enable cleavage by ADAMTS13. Dysfunction of this process causes von Willebrand disease (VWD) and thrombotic thrombocytopenic purpura (TTP). Although we previously reported the NMR assignments for mouse vWF A2, the human ortholog shares only 79% sequence identity (38 amino acid differences). Given that VWD and TTP are human pathologies, structural characterization of the human protein is essential. Here, we present the backbone 1H, 13C, and 15N resonance assignments of human vWF A2. Secondar

Measurement of absolute abundance of crystallins in human and αA N101D transgenic mouse lenses using 15N-labeled crystallin standards.

Stable isotope labeled standards of all major human lens crystallins were created to measure the abundance of lens endogenous crystallins from birth to adulthood. All major human crystallins (αA, αB, βA2, βA3/A1, βA4, βB1, βB2, βB3, γA, γB, γC, γD, γS) were cloned with N-terminal 6 x His tagged SUMO for ease of purification and the ability to generate natural N-termini by SUMO protease cleavage when producing crystallins for structure/function studies. They were then expressed in 15N-enriched media, quantified by mass spectrometry, and mixed in proportions found in young human lens to act as a

Mercury bioaccumulation pathways in tusk (Brosme brosme) from Sognefjord, Norway: Insights from C and N isotopes.

Seafood is the main source of methylmercury (MeHg) exposure for humans and elevated total mercury (Hg) concentrations have been reported in marine fish from Norwegian fjords compared with offshore areas. Hg in tusk fillets (n = 201) and liver samples (n = 177) were measured in individuals from different habitats including offshore, coastal area, outer and inner Sognefjord. Specifically, the effects of habitat, energy sources and trophic complexity on Hg bioaccumulation pathways in tusk (Brosme brosme) were investigated using stable isotopes of carbon (δ13C) and nitrogen (δ15N). 1HN, 13C, and 15N resonance assignments of the Clostridioides difficile receptor binding domain 2 (CDTb, residues 757-876).

Clostridioides difficile is a bacterial pathogen responsible for the majority of nosocomial infections in the developed world. C. difficile infection (CDI) is difficult to treat in many cases because hypervirulent strains have evolved that contain a third toxin, termed the C. difficile toxin (CDT), in addition to the two enterotoxins TcdA and TcdB. CDT is a binary toxin comprised of an enzymatic, ADP-ribosyltransferase (ART) toxin component, CDTa, and a pore-forming or delivery subunit, CDTb. In the absence of CDTa, CDTb assembles into two distinct di-heptameric states, a symmetric and an asym

Integrated assessment of wastewater pollution in a karstic coastal lagoon: linking nutrients, caffeine, stable isotopes, and fish physiological responses.

Coastal water pollution is a global problem, especially in karstic areas, where pollutants from point and non-point sources reach coastal systems. The Chelem Lagoon in the Yucatan Peninsula exemplifies such environments, where the presence and effects of pollutants have been previously reported. This study presents and evaluates an integrated assessment framework to characterize pollution sources and ecotoxicological impacts in complex coastal systems. This approach combines nutrient concentrations, caffeine (as an urban wastewater tracer), carbon and nitrogen stable isotopes, and a suite of p
