Gene expression profiling at early organogenesis reveals both common and diverse mechanisms in foregut patterning.

The thyroid and lungs originate as neighboring bud shaped outgrowths from the midline of the embryonic foregut. When and how organ specific programs regulate development into structures of distinct shapes, positions and functions is incompletely understood. To characterize, at least in part, the genetic basis of these events, we have employed laser capture microdissection and microarray analysis to define gene expression in the mouse thyroid and lung primordia at E10.5. By comparing the transcriptome of each bud to that of the whole embryo as well as to each other, we broadly describe the gene

Dividing the tubular gut: generation of organ boundaries at the pylorus.

The discrete organs that comprise the gastrointestinal tract (esophagus, stomach, small intestine, and large intestine) arise embryonically by regional differentiation of a single tube that is initially morphologically similar along its length. Regional organ differentiation programs, for example, for stomach or intestine, involve signaling cross-talk between epithelium and mesenchyme and result in the formation of precise boundaries between organs, across which dramatic differences in both morphology and gene expression are seen. The pylorus is a unique area of the gut tube because it not onl

Developmental toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in the most TCDD-resistant and -susceptible rat strains.

To gain insight into the relationship between TCDD acute toxicity and teratogenicity, developmental effects of TCDD were assessed in the most TCDD-resistant and -sensitive rat strains, Han/Wistar (Kuopio [H/W]; LD50 > 7200 micrograms/kg) and Long-Evans (Turku AB [L-E]; LD50 about 10 micrograms/kg), respectively. A single oral dose of 0, 1, or 10 micrograms/kg TCDD was given to mated female H/W rats on Gestation Day 8 or 12. In L-E rats, the doses were 0, 1, or 5 micrograms/kg TCDD and the exposure took place on Gestation Day 8. Fetuses were examined on Gestational Day 20 for external and soft

Acquisition of neural crest promoted thyroid evolution from chordate endostyle.

The endostyle is an endodermal organ unique to nonvertebrate chordates except for lamprey larvae, where it serves as forerunner to the adult thyroid. Here, we examine whether the acquisition of neural crest in the vertebrate lineage played a role in the elaboration of the endostyle. CM-DiI lineage tracing reveals a neural crest contribution to the endostyle, and CRISPR-Cas9 mutagenesis of key neural crest genes causes endostyle defects including formation of a single rather than bilobed structure. RNA sequencing reveals gene profiles characteristic of embryonic neural crest cells and Schwann c

The genetic landscape of cardiovascular left-right patterning defects.

Heterotaxy is a disorder with complex congenital heart defects and diverse left-right (LR) patterning defects in other organ systems. Despite evidence suggesting a strong genetic component in heterotaxy, the majority of molecular causes remain unknown. Established genes often involve a ciliated, embryonic structure known as the left-right organizer (LRO). Herein, we focus on genetic discoveries in heterotaxy in the past two years. These include complex genetic architecture, novel mechanisms regulating cilia formation, and evidence for conservation of LR patterning between distant species. We f

Inactivation of Lats1 and Lats2 highlights the role of hippo pathway effector YAP in larynx and vocal fold epithelium morphogenesis.

Proliferation and differentiation of vocal fold epithelial cells during embryonic development is poorly understood. We examined the role of Hippo signaling, a vital pathway known for regulating organ size, in murine laryngeal development. Conditional inactivation of the Hippo kinase genes Lats1 and Lats2, specifically in vocal fold epithelial cells, resulted in severe morphogenetic defects. Deletion of Lats1 and Lats2 caused abnormalities in epithelial differentiation, epithelial lamina separation, cellular adhesion, basement membrane organization with secondary failed cartilage, and laryngeal

A centrosomal view of CNS growth.

Embryonic development of the central nervous system (CNS) requires the proliferation of neural progenitor cells to be tightly regulated, allowing the formation of an organ with the right size and shape. This includes regulation of both the spatial distribution of mitosis and the mode of cell division. The centrosome, which is the main microtubule-organizing centre of animal cells, contributes to both of these processes. Here, we discuss the impact that centrosome-mediated control of cell division has on the shape of the overall growing CNS. We also review the intrinsic properties of the centro

Abdominal wall defects: autopsy findings of distinct groups suggest different pathogenetic mechanisms.

Central and lateral abdominal wall defects are probably distinct and likely arise from different pathogenetic mechanisms. An autopsy study was done using data from a total of 45 central and lateral abdominal wall defect cases to evaluate if they are indeed separate entities and to suggest possible mechanisms involved in their formation. Central defects were found to be statistically different from lateral defects; malformations that co-existed with central defects were mainly bilateral and internal and also involved "inferior" organs in relation to fetal-embryonal blood flow. Patients with lat

Stem cell plasticity: the debate begins to clarify.

The stem cell story begins with the recognition of the regenerative powers of the head of the Lernean Hydra and the human liver (Prometheus) by the ancient Greeks. In modern times, the adult human stem cell has been epitomized by the hematopoietic stem cell in the bone marrow. More recently, bone marrow derived cells were reported to contribute to nonhematopoietic organs, suggesting a level of plasticity not previously expected. However, other reports failed to repeat some of these results, resulting in a heated debate on the plasticity of adult stem cells that has crossed over into the public

Transactivation of Igf2 in a mouse model of Beckwith-Wiedemann syndrome.

The gene IGF2, which encodes a fetal insulin-like growth factor, is imprinted, so only one of two parental copies of the gene is expressed. The altered expression of IGF2 has been implicated in Beckwith-Wiedemann syndrome, a human fetal overgrowth syndrome, which is characterized by overgrowth of several organs and an increased risk of developing childhood tumours. We have introduced Igf2 transgenes into the mouse genome by using embryonic stem cells, which leads to transactivation of the endogenous Igf2 gene. The consequent overexpression of Igf2 results in most of the symptoms of Beckwith-Wi

Anomalies of heart, spleen, kidneys, gut, and limbs may result from an overdistended neural tube: a hypothesis.

A hypothesis is advanced that many congenital anomalies of non-neural organs may be produced by damage to their mesodermal or entodermal anlagen caused by overdistention of the embryonic neural tube. Evidence to support the hypothesis derives from: (1) seldom appreciated but unequivocal embryologic facts about prechoroid plexus neural tube morphogenesis; (2) an understanding of the role of neural tube overdistention in the production of various dysraphic conditions; (3) the frequency of association of non-neural anomalies with dysraphic conditions; and 4) an analysis of the anatomic features o

[Congenital german measles encephalitis (author's transl)].

German measles is an infections exanthematous disease, long regarded as completely harmless. The clinical symptoms are banal and often go unnoticed. In complete contrast is the severe malformation of the embryo after infection early on in pregnancy, first discovered by Gregg in 1941. The virus infects the embryo via the placenta and either causes death or impairs the differentiation of embryonic tissues, causing malformation. What is understood today under the congenital German measles syndrome is considerably more extensive and includes, in addition to malformation, growth retardation, premat

Studies on reproduction in the goat. 3. The functional activity of the ovaries of the goat.

1. A study of the functional activity in the ovaries was undertaken on 366 organs from pregnant goats and 171 organs from nonpregnant goats. The ovulations were recorded by the presence of corpora lutea in the ovaries. 2. The right ovary appears to be the most active in the goat, since 57 % of the ovulations occurred in the right ovary. In cases of multiple ovulations there appears to be a greater tendency for the first of these ovulations to occur in the right ovary (62.34 %). 3. In almost 50 % of twin pregnancies both ova will be shed from the same ovary. 4. In six cases, five twin pregnanci

The role of vertebrate nonmuscle Myosin II in development and human disease.

Three different genes each located on a different chromosome encode the heavy chains of nonmuscle myosin II in humans and mice. This review explores the functional consequences of the presence of three isoforms during embryonic development and beyond. The roles of the various isoforms in cell division, cell-cell adhesion, blood vessel formation and neuronal cell migration are addressed in animal models and at the cellular level. Particular emphasis is placed on the role of nonmuscle myosin II during cardiac and brain development, and during closure of the neural tube and body wall. Questions a

[Cilia and renal cysts].

Advances in genomics, bioinformatics and the creation of model organisms have identified many genes associated with polycystic kidney diseases. Historically, these genes were not necessarily associated with ciliopathies, but it appeared that many connections can be made between the cystic kidney disease and function of the primary cilium. Indeed, the proteins encoded by these genes are localized to the cilium itself, to the basal body or are known to regulate the expression and localization of ciliary proteins. The goal of this article is to describe the multiple cellular processes that may le

Anterior pituitary cell networks.

Both endocrine and non-endocrine cells of the pituitary gland are organized into structural and functional networks which are formed during embryonic development but which may be modified throughout life. Structural mapping of the various endocrine cell types has highlighted the existence of distinct network motifs and relationships with the vasculature which may relate to temporal differences in their output. Functional characterization of the network activity of growth hormone and prolactin cells has revealed a role for cell organization in gene regulation, the plasticity of pituitary hormon

Mice lacking p27(Kip1) display increased body size, multiple organ hyperplasia, retinal dysplasia, and pituitary tumors.

Mice lacking p27(Kip1) have been created by gene targeting in embryonic stem cells. These mice are larger than the control animals, with thymus, pituitary, and adrenal glands and gonadal organs exhibiting striking enlargement. CDK2 activity is elevated about 10-fold in p27(-/-) thymocytes. Development of ovarian follicles seems to be impaired, resulting in female sterility. Similar to mice with the Rb mutation, the p27(-/-) mice often develop pituitary tumors spontaneously. The retinas of the mutant mice show a disturbed organization of the normal cellular layer pattern. These findings indicat

Surgical implications of pediatric endocrine disorders.

The adrenal gland and the gonads both originate in the embryonic posterior mesenchyme and secrete steroids. Malfunction in either of them results in certain typical developmental problems. The adrenogenital syndrome resulting from excess androgen production occurs as masculinization in the female and precocious maturation in the male. Adrenal tumor usually has the same presentation; feminizing tumors are rare and usually are malignant. Gonadal problems include enlargement of sex organs, ambiguous genitalia, missing testes, and tumor causing precocious puberty. Endocrine problems must be assess

Speed vs completeness: a comparative study of solitary and colonial tunicate embryogenesis.

Solitary ascidians, such as Ciona robusta, have been used for over a century as model systems for embryological studies. These species are oviparous, producing many relatively small and transparent eggs, which are released and fertilized outside the parent body. Embryos develop rapidly in a stereotyped manner and reach the larva stage in less than 1 day (at 20°C). The larvae then settle and metamorphose into sessile juveniles in approximately 2 days. On the other hand, colonial ascidians are ovoviviparous, with heavily yolked eggs that develop inside the parent body. In the colonial Botryllus

Developmental evolution and developmental plasticity of the olfactory epithelium and olfactory skills in Mexican cavefish.

The fish Astyanax mexicanus comes in two forms: the normal surface-dwelling (SF) and the blind depigmented cave-adapted (CF) morphs. Among many phenotypic differences, cavefish show enhanced olfactory sensitivity to detect amino-acid odors and they possess large olfactory sensory organs. Here, we questioned the relationship between the size of the olfactory organ and olfactory capacities. Comparing olfactory detection abilities of CF, SF and F1 hybrids with various olfactory epithelium (OE) sizes in behavioral tests, we concluded that OE size is not the only factor involved.
