Prenatal diagnosis and genetic counseling of a paternally inherited chromosome 8q24.22q24.23 microdeletion in a Chinese family.

BACKGROUND: Copy number variants (CNVs) are an important source of normal and pathogenic genome variations. Especially CNVs identified in prenatal cases need careful considerations and correct interpretation if those are harmless or harmful variants from the norm. The literature on 8q24.22q24.23 microdeletion is rare, which is a challenge for genetic counselling. CASE PRESENTATION: We have performed prenatal diagnosis and genetic counseling of a paternally inherited 8q24.22q24.23 microdeletion. In this family, father with normal phenotype and fetus with abnormal phenotype have the same microd

Identification of hub genes associated with decreased fertility in male mice of advanced paternal age.

INTRODUCTION: Aging and delayed parenthood are major social concerns. Men older than 35 years, which is an advanced paternal age, experience reduced sperm quality and fertility. METHODS: In this study, 12-month-old mice served as a model for males of advanced paternal age. RNA sequencing (RNA-seq) of epididymides from 2- and 12-month-old mice was performed. RESULTS: Spermatogonia and sperm counts were significantly lower in these mice. We identified 449 differentially expressed genes by RNA-seq. Altered pathways were enriched using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes (K

Maternal and paternal smoking and offspring antral follicle count in women from an infertility clinic.

BACKGROUND: The ovarian reserve is established in utero, and therefore may be influenced by parental characteristics such as tobacco smoking. However, the epidemiologic literature on parental smoking and adult antral follicle count (AFC) remains conflicted. RESEARCH DESIGN AND METHODS: Our study included 631 women enrolled in the Environment and Reproductive Health (EARTH) study, a prospective cohort at an academic fertility center between 2005 and 2019. Exposure was assessed by women self-reporting their mother's and father's overall and pregnancy specific smoking status. Outcome was assesse

AAV-dCas9 vector unsilences paternal Ube3a in neurons by impeding Ube3a-ATS transcription.

Angelman syndrome (AS) is a debilitating neurodevelopmental disorder caused by loss of maternally-inherited UBE3A. In neurons, paternally-inherited UBE3A is silenced in cis by a long non-coding RNA called Ube3a-ATS. Here, we found that Neisseria meningitidis Cas9 with two mutations (D15A and H587A) in the nuclease domains (dNmCas9) can unsilence the dormant paternal Ube3a allele in mouse and human neurons when targeted to Snord115 snoRNA genes located in introns of Ube3a-ATS. Importantly, dNmCas9 disrupted Ube3a-ATS with a non-template bias and in the absence of a chromatin modifying domain, s

Association of Paternal BMI and Diet During Pregnancy with Offspring Birth Measures: The STEPS Study.

BACKGROUND/OBJECTIVES: Maternal Body Mass Index (BMI), diet quality, and their associated effects on offspring birth measures are well-established. Emerging evidence, largely from animal studies, has indicated paternal factors can influence offspring birth outcomes. However, this effect is poorly understood in humans. Our aim was to examine the association between paternal BMI and diet quality score and offspring birth measures. METHODS: Participants from the STEPS (Steps to the healthy development) Study in Southwest Finland were recruited during the first trimester of pregnancy or after del

Foetal cardiac rhabdomyoma due to paternal TSC1 Mutation: a case report and literature review.

Rhabdomyomas are the most common prenatal cardiac tumours, and are often associated with tuberous sclerosis complex (TSC). They have been shown to grow during foetal development, but may often regress or shrink in early childhood. In the present case, ultrasonography at 20+2 gestational weeks identified two echogenic masses suspicious of rhabdomyomas in the foetal heart. Neither of these tumours caused significant haemodynamic instability. Genetic testing of DNA extracted from amniocytes revealed a pathogenic variant of the TSC1 gene, supporting the diagnosis of tuberous sclerosis. The pregnan

Does paternal alcohol consumption affect the severity of traits of fetal alcohol spectrum disorders?

BACKGROUND: Animal models suggest that paternal alcohol consumption may influence offspring traits, yet few human studies exist. METHODS: Data from population-based studies of fetal alcohol spectrum disorders (FASD) among first-grade students provided case-control data to explore traits of fathers of children with: FASD, alcohol exposure, and unexposed controls. RESULTS: Most males in this population drank, but more fathers of children with FASD drank during pregnancy ( x ¯ = 73.2%) than fathers of controls ( x ¯ = 63.4%). Among drinkers, fathers of children with FASD: (a) consumed

PEG10 loss of function causes Silver-Russell syndrome: a familial case with paternal deletion.

Silver-Russell syndrome (SRS, MIM#180860) is an imprinting disorder characterized by prenatal and postnatal growth retardation, relative macrocephaly at birth, prominent forehead, feeding difficulties, and body asymmetry. Clinical diagnosis is based on at least 4 out of 6 clinical signs (Netchine-Harbison clinical scoring system). The main molecular mechanisms are loss of methylation at the paternal H19/IGF2:IG-DMR (30-60%) at the 11p15 chromosomal region and maternal uniparental disomy of chromosome 7 (5-10%). While it is well known that deregulation of 11p15 imprinted genes (IGF2, H19, and C

Adult bi-paternal offspring generated through direct modification of imprinted genes in mammals.

Imprinting abnormalities pose a significant challenge in applications involving embryonic stem cells, induced pluripotent stem cells, and animal cloning, with no universal correction method owing to their complexity and stochastic nature. In this study, we targeted these defects at their source-embryos from same-sex parents-aiming to establish a stable, maintainable imprinting pattern de novo in mammalian cells. Using bi-paternal mouse embryos, which exhibit severe imprinting defects and are typically non-viable, we introduced frameshift mutations, gene deletions, and regulatory edits at 20 ke

Advanced Paternal Age in Focus: Unraveling Its Influence on Assisted Reproductive Technology Outcomes.

As global demographics shift toward increasing paternal age, the realm of assisted reproductive technologies (ARTs), particularly in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI), faces new challenges and opportunities. This study provides a comprehensive exploration of the implications of advanced paternal age on ART outcomes. Background research highlights the social, cultural, and economic factors driving men toward later fatherhood, with a focus on the impact of delayed paternity on reproductive outcomes. Methods involve a thorough review of existing literature, cen

Paternal Incarceration and Adolescent Delinquency: Role of Father Engagement and Early Child Behavior Problems.

Paternal incarceration is an important predictor of teen delinquency, but the factors that may explain this relationship-such as early child problem behaviors and level of father engagement-have not been adequately explored. The current longitudinal study examined paternal history of incarceration as a predictor of teen self-reported delinquency over a 15-year gap, considering early child problem behaviors and father engagement as mediators. Sex differences in these relationships were also evaluated. This four-wave longitudinal study included an analytic sample of 4897 teens who participated i

Paternal age, de novo mutations, and offspring health? New directions for an ageing problem.

This Directions article examines the mechanisms by which a father's age impacts the health and wellbeing of his children. Such impacts are significant and include adverse birth outcomes, dominant genetic conditions, neuropsychiatric disorders, and a variety of congenital developmental defects. As well as age, a wide variety of environmental and lifestyle factors are also known to impact offspring health via changes mediated by the male germ line. This picture of a dynamic germ line responsive to a wide range of intrinsic and extrinsic factors contrasts with the results of trio studies indicati

Exploring paternal postpartum depression and contributing factors in Ethiopia: a systematic review and meta-analysis.

BACKGROUND: Paternal postpartum depression (PPD) is a significant yet often neglected mental health issue affecting fathers during the postpartum period. While maternal postpartum depression is extensively studied, the psychological challenges faced by new fathers, particularly in resource-limited settings like Ethiopia, receive considerably less attention. Paternal postpartum depression not only impacts fathers' well-being but also affects the health and development of their children and their relationships with partners. Understanding the prevalence of paternal postpartum depression and its

Paternal Preconception Hepatitis B Virus Infection and Risk of Congenital Heart Disease in Offspring.

IMPORTANCE: Previous evidence suggests that maternal hepatitis B virus (HBV) infection during prepregnancy or pregnancy is associated with congenital heart diseases (CHDs) in offspring. However, the association of paternal HBV infection with CHDs is not well examined. OBJECTIVE: To explore the association of paternal preconception HBV infection with CHDs in offspring. DESIGN, SETTING, AND PARTICIPANTS: This retrospective cohort study used propensity score matching of data from the Chinese National Free Preconception Checkup Project (NFPCP) from January 1, 2010, to December 31, 2018. Recurrence of CCHS-associated PHOX2B Poly-Alanine expansion variant due to paternal mosaicism.

BACKGROUND: Paired-like Homeobox 2B (PHOX2B) is considered the causative gene of Congenital Central Hypoventilation Syndrome (CCHS), a dominant genetic disorder characterized by impaired central respiratory control and subsequent hypoventilation during sleep. METHODS: Herein, we present a family with recurrent severe CCHS. The potential causative genetic variant was confirmed through Whole-Exome Sequencing (WES), Sanger sequencing, and droplet digital PCR (ddPCR). Furthermore, prenatal diagnosis was performed on the proband's mother at 20 weeks of her fourth pregnancy upon request. RESULTS:

Paternal morphine alters offspring circulating beta-endorphin and corticosterone responses to oxycodone and cocaine.

BACKGROUND: The opioid epidemic is leading to increased opioid use in adolescent populations. A growing body of evidence suggests that taking opioids during adolescence can disrupt normal development and impact future offspring. This study investigates the impact of paternal morphine exposure during adolescence on the hypothalamic-pituitary-adrenal (HPA) axis and release of endorphins in the offspring. METHODS: Male rats were administered morphine once a day from postnatal day (PND)30-39 using an increasing dosing regimen (5-25 mg/kg/day increasing every other day). They were mated during adu

Paternal Age Matters: Association with Sperm Criteria's- Spermatozoa DNA Integrity and Methylation Profile.

Advanced age has been reported to negatively affect sperm parameters and spermatozoa DNA integrity. A decline in sperm criteria was also associated with altered epigenetic marks such as DNA methylation with a potential downstream impact on in vitro fertilization success and clinical outcomes. The aim of the present retrospective study was to clarify the association between advanced paternal age (APA) and sperm parameters, DNA integrity and DNA methylation profile. A total of 671 patients consulting for infertility underwent sperm analysis, sperm DNA integrity assessment and methylation level m
