Investigation of an alkylamine modified and pH-controlled mobile phase for separation of pyrazolone derivatives by high-performance liquid chromatography.

The separation of pyrazolone derivatives with different acid-base properties and polarities has been investigated in view of recent concepts about the retention mechanisms in reversed-phase liquid chromatography. It is shown that both the protonated (BH+) and uncharged (B) bases undergo a dual mechanism of retention due to solvophobic and silanophilic interactions. The latter are easily suppressed by the addition of an alkylamine modifier (dibutylamine) to the mobile phase. An ion-pairing mechanism is found to be valid in the retention of negatively charged (A-) pyrazolone derivatives, since t

Overcoming challenges in LC separation of oxysterols through guided design space modelling: A comparison of three stationary phases.

Liquid chromatography-mass spectrometry (LC-MS) is an attractive alternative to GC-MS for oxysterol (OS) analysis, as it eliminates the need for chemical derivatization. However, in-source fragmentation during electrospray ionization (ESI) or atmospheric pressure chemical ionization (APCI), usually adopted for LC-MS analysis of OS, may lead to the generation of isomeric ions from non-isomeric precursors. This prevents a complete unambiguous identification along the MS dimension and makes chromatographic separation a fundamental step. In this work, analytical design space modelling empowered by

Grass carp reovirus VP35 hijacks DHX15 into phase-separated inclusion bodies to evade host antiviral immunity.

Many viral proteins undergo liquid-liquid phase separation (LLPS) to form biomolecular condensates known as viral inclusion bodies (VIBs), which are utilized for genome replication and virion assembly, thus serving as potential targets for antiviral drugs. However, the role of VIBs in viral immune evasion has rarely been explored. In this study, we demonstrated that VP35 protein of type II grass carp reovirus (GCRV-II) formed VIBs through LLPS in cells and in vitro. Moreover, we identified a host interaction partner of GCRV-II VP35, DEAH (Asp-Glu-Ala-His)-box helicase 15 (DHX15), which promote…

Ionic Liquid-Mediated Phase Separated Hydrogels With Bicontinous Conductive Networks for Long-Term Underwater Strain Sensing.

Ionic conductive hydrogels have attracted significant attention as wearable sensors due to their intrinsic flexibility, biocompatibility, an … rategy utilizes a DMSO-IL binary solvent that exploits differential polymer-water affinity to drive phase separation. Notably, hydrophobic IL enhances phase separation through multiple interactions with hydrophilic groups while constructing a bicontinuous conductive network within the hydrogel. The strategic incorporation of poly(vinyl alcohol) (PVA) increases the …

Green synthesis of biomass derived monodisperse microspheres used as chromatographic stationary phases for separation of dye molecules in ethanol.

Green liquid chromatography (LC) has garnered increasing attention; however, its adoption as a replacement for conventional methods is limited by inferior separation performance. Herein, we present a fully integrated design for green LC, encompassing biomass-derived starting materials, the green synthesis of a solid stationary phase, and environmentally benign separation using ethanol as the mobile phase. Via a green precipitation polymerization technique (room temperature quiescent redox-initiated precipitation polymerization, RTQRPP), we synthesized a series of monodisperse microspheres deri

N6-methyladenosine-mediated feedback regulation of abscisic acid perception via phase-separated ECT8 condensates in Arabidopsis.

N6-methyladenosine (m6A) is the most abundant internal modification in eukaryotic mRNAs, yet how plants recognize this chemical modification to swiftly adjust developmental plasticity under environmental stresses remains unclear. Here we show that m6A mRNA modification and its reader protein EVOLUTIONARILY CONSERVED C-TERMINAL REGION 8 (ECT8) act together as a key checkpoint for negative feedback regulation of abscisic acid (ABA) signalling by sequestering the m6A-modified ABA receptor gene PYRABACTIN RESISTANCE 1-LIKE 7 (PYL7) via phase-separated ECT8 condensates in stress granules in respons

Are Highly Stable Covalent Organic Frameworks the Key to Universal Chiral Stationary Phases for Liquid and Gas Chromatographic Separations?

High-performance liquid chromatography (HPLC) and gas chromatography (GC) over chiral stationary phases (CSPs) represent the most popular and highly applicable technology in the field of chiral separation, but there are currently no CSPs that can be used for both liquid and gas chromatography simultaneously. We demonstrate here that two olefin-linked covalent organic frameworks (COFs) featuring chiral crown ether groups can be general CSPs for extensive separation not only in GC but also in normal-phase and reversed-phase HPLC. Both COFs have the same 2D layered porous structure but channels o

Analysis of the solvent strength parameter (linear solvent strength model) for isocratic separations in reversed-phase liquid chromatography.

The solvent strength parameter (slope) and intercept (log kw) of the linear solvent strength model are determined for methanol-water (17 columns), acetonitrile-water (15 columns), acetone-water (7 columns), tetrahydrofuran-water (6 columns), and 2-propanol-water (4 columns) for varied compounds. It is shown that the linear region of the plots of the retention factor (log k) against the volume fraction of organic solvent (ϕ) is largely a system property independent of solute type for compounds with accessible retention factors. Solvent strength parameters for methanol (3.12 ± 0.12), acetonitril

Porous graphite as stationary phase for the chromatographic separation of polymer additives - determination of adsorption capability by Raman spectroscopy and physisorption.

Additives are added to polymers in small concentration to achieve desired application properties widely used to tailor the properties. The rapid diversification of their molecular structures, with often only minute differences, necessitates the development of adequate chromatographic techniques. While modified silica so far is the workhorse as stationary phase we have probed the potential of porous graphitic carbon (HypercarbTM) for this purpose. The results show that the multitude of physicochemical interactions between analyte molecules and the graphitic surface enables separations of polyol

Phase separation in phosphatidylcholine membrane caused by the presence of a pyrimidine analogue of fluphenazine with high anti-multidrug-resistance activity.

Phenothiazine compounds are known as effective inhibitors of a multidrug resistance (MDR) of tumor cells to chemotherapeutic agents. This group consists of many important substances used in human medicine such as antipsychotic drugs in the case of fluphenazine (FPh) or chlorpromazine (CPZ). Fluphenazine was on the World Health Organization (WHO) list of Essential Medicines of 2009, and its new pyrimidine analog (FPh-prm) presented in this work has been documented to have a high anti-MDR activity. In order to discover the character of alterations of the lipid bilayer structure caused by the pre

Changes in amplitude and phase of distortion-product otoacoustic emission fine-structure and separated components during efferent activation.

Medial olivocochlear (MOC) efferent fibers synapse directly on the outer hair cells (OHCs). Efferent activation evoked by contralateral acoustic stimulation (CAS) will affect OHC amplification and subsequent measures of distortion-product otoacoustic emissions (DPOAEs). The aim of this study was to investigate measures of total and separated DPOAEs during efferent activation. Efferent activation produces both suppression and enhancement of the total DPOAE level. Level enhancements occurred near fine-structure minima and were associated with consistent MOC evoked upward shifts in DPOAE fine-str

Enhanced extraction yields and mobile phase separations by solvent mixtures for the analysis of metabolites in Annona muricata L. leaves.

The effects of five extraction solvents and their mixtures on the yield of metabolites in crude and fractionated extracts of Annona muricata L. leaves were investigated by direct comparison. Extraction media were prepared using simplex centroid mixtures of ethanol, ethyl acetate, dichloromethane, acetone, and chloroform. The effects of the mobile phase solvent strength and the analysis wavelength on the chromatographic separation were also investigated. Solvent mixtures rather than pure solvents were found to be the most efficient extractors for the different fractions. The results indicated t

[Comparative study of the immunogenicity of neurotoxin preparations and their separate fractions obtained from Shigella sonnei, phases I and II].

The immunogenicity of the preparations of phase I S. sonnei neurotoxin, determined by the keratoconjunctival test on guinea pigs, is linked mainly with the presence of a high-molecular component other than endotoxin in these preparations. The preparations of phase II S. sonnei neurotoxin do not contain the endotoxin and high-molecular antigens of S. sonnei, phase I, which protect guinea pigs from the development of experimental keratoconjunctivitis; these preparations are toxic for mice and possess low immunogenicity.

Evaluation of a glassy-carbon electrode for amperometric detection of selected methylxanthines in serum after their separation by reversed-phase chromatography.

We examined the relative merits of direct-current, pulse, and differential pulse amperometry at a flow-through, glassy-carbon electrode used in monitoring column chromatography. We give a rapid, simple procedure for determining theophylline (1,3-dimethylxanthine) in 20--100 microliter of blood serum. Interference by 1,7-dimethylxanthine, a metabolite of caffeine, is compared for photometric and amperometric detection, and some advantages of combined photometric and amperometric detection are discussed.

Engineering thiol-ene click chemistry for the preparation of a chiral stationary phase based on a [4+6]-type homochiral porous organic cage for enantiomeric separation in normal-phase and reversed-phase high performance liquid chromatography.

In this study, a new chiral stationary phase (CSP) was fabricated by covalent bonding of a [4+6]-type homochiral porous organic cage (POC) CC19-R onto thiolated silica via a thiol-ene click reaction. The CC19-R was synthesized via Schiff-base reaction between 2-hydroxybenzene-1,3,5-tricarbaldehyde and (1R, 2R)-(-)-1,2-diaminocyclohexane. The enantioseparation capability of the resulting CC19-R-based CSP was systematically evaluated upon separating various chiral compounds or chiral pharmaceuticals in normal phase HPLC (NP-HPLC) and reversed phase HPLC (RP-HPLC), including alcohols, organic aci

Orthogonal separation and identification of long-chain peptides from scorpion Buthus martensi Karsch venom by using two-dimensional mixed-mode reversed phase-reversed phase chromatography coupled to tandem mass spectrometry.

Peptide components of scorpion venom have been employed as useful pharmacological tools in the study of ion channel function. The isolation of individual components is necessary for determination of their biological significance. Here, we have described a novel reversed phase (RP)/ion exchange stationary phase, Click oligo ethylene glycol (Click OEG), and the chromatographic efficiency of its mixed-mode sorbent in peptide separation experiments. The Click OEG presents a mixed-mode RP/weak anion-exchange type stationary phase at pH 3.5 and mixed-mode RP/weak cation-exchange type stationary phas

Post-modification of Gemini cross-linked poly(ionic liquid) stationary phase and the effect of phenyl/pentafluorophenyl groups on separation selectivity.

Gemini cross-linked poly(ionic liquid) possess excellent stability and tunable structures, demonstrating significant application potential in the field of chromatographic separation. By introducing functional groups with different properties onto their backbones via post-modification, the separation selectivity of the stationary phases can be further precisely modulated. In this study, using a Gemini cross-linked poly(ionic liquid) as the fundamental backbone, two functionalized chromatographic stationary phases were prepared by post-modification with phenyl and pentafluorophenyl functional li

Application of solid phase extractants TEVA and TK200 for the separation of medically relevant 199Au from thermal neutron irradiated Pt target.

This study describes the separation of no-carrier-added (n.c.a.) 199Au from thermal neutron irradiated natural platinum targets using two extraction chromatographic resins: TEVA containing a quaternary amine (Aliquat 336) and TK200 impregnated with trioctylphosphine oxide (TOPO, Cyanex 921). The behavior of Au, Pt, and Ir in HCl media (0.1-10 mol·L-1 HCl) is examined through batch distribution coefficient (Kd) measurements and dynamic column experiments. For TEVA, the Kd of Au remains high across the entire HCl range (2-3·104 mL·g-1), whereas Pt shows a maximum Kd of 3.3·104 mL·g-1 at 1 mol·L-
