OBJECTIVE: To ascertain efficacy and tolerability of carbamazepine (CBZ), sodium valproate (VPA), lamotrigine (LTG) and levetiracetam (LEV) using the UKAED register (www.ukaed.info).
METHODS: Patients on CBZ (n=91), VPA (n=61), LTG (n=105), LEV (n=72) and healthy control subjects (CTR) on no medication (n=51) were extracted. All patients had anonymously provided information on seizure type and frequency and completed the Liverpool Adverse Event Profile (LAEP).

BACKGROUND: Numerous anticonvulsant agents are now available for treating status epilepticus (SE). However, a paucity of data is available to guide clinicians in the initial treatment of seizures or SE. This study describes the current strategies being employed to treat SE in the U.S.A.
METHODS: Fifteen American academic medical centers completed a retrospective, multicenter, observational study by reviewing 10-20 of the most recent cases of SE at their institution prior to December 31, 2009. A multivariate analysis was performed to determine factors associated with cessation of seizures.

A combination of valproates in dose 15-25 mg per day and levetiracetam in dose 20-25 mg per day was used in the treatment of 23 patients with different forms of epilepsy (juvenile myoclonic epilepsy, juvenile absence epilepsy, convulsive idiopathic generalized epilepsy and children absence epilepsy). The use of these drugs was effective in all forms of epilepsy. There were positive changes in the frequency of seizures and epileptic activity in the EEG.

OBJECTIVE: Conventional low-dose levetiracetam (LEV) for post-neurosurgical seizure prophylaxis often yields subtherapeutic serum levels. This study characterized the population pharmacokinetics (PPK) of intravenous LEV to optimize dosing in neurosurgical patients.
METHODS: We conducted a retrospective PPK analysis in 87 neurosurgical patients (131 concentrations). Nonlinear mixed-effects modeling was used. Monte Carlo simulations identified regimens achieving ≥ 90% probability of target attainment (PTA) for trough concentrations (Cmin) of 12-46 μg/mL.
