Extracellular vesicles (EVs) are crucial mediators of intercellular communication in the tumor microenvironment (TME) which facilitate the transfer of bioactive molecules including functional mitochondria and their integral components. This review summarizes the emerging role of EV-mediated mitochondrial transfer in cancer progression. We delineate the mechanisms governing the packaging of mitochondria and their constituents into EVs and subsequently highlight their multifaceted functions across various malignancies, including breast cancer, prostate cancer, blood malignancies, head and neck s

Ocular neovascular diseases (ONDs) are a leading cause of global blindness. While current anti-vascular endothelial growth factor (VEGF) treatments have revolutionized management, their efficacy is constrained by issues such as non-response, resistance development, and the burden of repeated intravitreal injections. Recent studies have highlighted the growing importance of transfer RNA-derived small RNAs (tsRNAs) in regulating pathological angiogenesis. This review provides a systematic analysis of tsRNAs, encompassing their origins, classification, and mechanisms of action, followed by an exp

Catocala electa is one of the secondary pests in agriculture. In this study, the whole mitochondrial genome sequence of C. electa was determined with the help of Illumina sequencing. The complete genome sequence is 15,575 bp in total length, which contains 13 protein-coding genes (PCGs), 22 transfer RNAs(tRNAs) region, 2 ribosomal RNAs (rRNAs) and 1 control region (CR). The overall base components of mitogenome of all bases was 39.58% for A, 41.72% for T, 11.38% for C and 7.33% for G. Phylogenetic analysis using the ML method showed that C. electa was clustered into one branch with Catocala sp

Phagocytosed Borrelia burgdorferi (Bb), the Lyme disease spirochete, induces a robust and complex innate immune response in human monocytes, in which TLR8 cooperates with TLR2 in the induction of NF-κB-mediated cytokine production, whereas TLR8 is solely responsible for transcription of IFN-β through IRF7. We now establish the role of Bb RNA in TLR8-mediated induction of IFN-β. First, using TLR2-transfected HEK.293 cells, which were unable to phagocytose intact Bb, we observed TLR2 activation by lipoprotein-rich borrelial lysates and TLR2 synthetic ligands but not in response to live spirochet

Developing computational methods for single-cell drug response prediction deepens our understanding of tumor heterogeneity and uncovers resistance mechanisms critical to improving cancer therapy. However, current approaches struggle to fully capture intratumoral heterogeneity, as bulk RNA sequencing (bulk RNA-seq) obscures heterogeneity across individual cells, while single-cell RNA sequencing (scRNA-seq) remains constrained by limited throughput and high cost. Current approaches integrating bulk and scRNA-seq data frequently encounter batch effects, impairing robust knowledge transfer. Moreov

We report here the characterization of a three-generation Han Chinese family with Leber's hereditary optic neuropathy (LHON). This Chinese family exhibited high penetrance and expressivity of visual impairment. The average age-of-onset was 19 years in this family. All male and 33% female matrilineal relatives in this Chinese family developed visual loss with a wide range of severity, ranging from blindness to normal vision. Sequence analysis of the complete mitochondrial DNA in this pedigree revealed the presence of the ND4 G11778A mutation and 40 other variants, belonging to the Asian haplogr

The reaction of E. coli 70s ribosomes with 0.2 M NaH-35 s03 (pH 7.1, 3.5hrs, 37 degree) led to the conversion of 4.5% of the uracil residues of the R, RNA into 5.6-dihydrouracil-6-sulfonate residues. The modified ribosomes exhibited a significant decrease in their ability to bind (14-C)-phenylalanyl-(RNA-phe and to incorporate (14-C)-phenylalanine into protein in the presence of polyuridylic acid. The ability of the modified ribosomes to form an initiation complex as measured by the A-U-G or coliphage MS2 RNA dependent binding of (14-C)-fmet-tRNA-fmet was also impaired, as was their ability to

INTRODUCTION: Distylium chinense is a flood-tolerant woody species distributed in the hydro-fluctuation zone of the Three Gorges Reservoir in the Yangtze River basin and plays an important role in regional ecological restoration. Nevertheless, its mitochondrial genome (mitogenome) has not yet been characterized.
METHODS: Here, the complete mitogenome of D. chinense was de novo assembled using a hybrid sequencing strategy integrating MGISEQ-2000 and Nanopore platforms. Codon usage bias was evaluated based on relative synonymous codon usage (RSCU). Simple sequence repeats (SSRs), mitochondrial

Trichothiodystrophy (TTD), a rare, autosomal recessive, multisystem developmental disorder, is characterized by short, brittle hair with "tiger-tailed banding" on polarized microscopy. TTD is caused by variations in 10 genes: 3 nucleotide excision repair/basal transcription factor IIH genes, 4 amino acid charging transfer RNA genes, basal transcription factor IIE, RNF113A, and an RNA-splicing gene (MPLKIP/TTDN1). We performed whole-exome sequencing to identify a candidate gene in Sabinas brittle hair syndrome, a mild form of TTD. We report 5 nonphotosensitive adult patients from 3 unrelated fa

Antimicrobial resistance develops as a major problem in infectious diseases treatment. While antibiotic resistance mechanisms are usually studied using lethal antibiotic doses, lower doses allowing bacterial growth are now considered as factors influencing the development and selection of resistance. Starting with a high-density Tn insertion library in Vibrio cholerae and following its evolution by TN-seq in the presence of subinhibitory concentrations of antibiotics, we discovered that RNA modification genes can have opposite fates, being selected or counter-selected. We, thus have undertaken

OBJECTIVE: The functional role of tRNA-derived small RNAs (tsRNAs) in CD3+ T lymphocytes in rheumatoid arthritis (RA) remains unclear. This study aimed to characterize their expression profile and evaluate their biomarker potential in RA.
METHODS: Small RNA sequencing of CD3+ T lymphocytes was performed to profile tsRNA expression. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were employed to explore associated pathways. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to verify the differentially expressed tsRNAs. Receiver operating charact

Toxins of toxin-antitoxin systems use diverse mechanisms to inhibit bacterial growth. In this study, we characterize the translation inhibitor toxin MenT3 of Mycobacterium tuberculosis, the bacterium responsible for tuberculosis in humans. We show that MenT3 is a robust cytidine specific tRNA nucleotidyltransferase in vitro, capable of modifying the aminoacyl acceptor ends of most tRNA but with a marked preference for tRNASer, to which long stretches of cytidines are added. Furthermore, transcriptomic-wide analysis of MenT3 targets in M. tuberculosis identifies tRNASer as the sole target of Me

Selenoproteins are defined as proteins containing the 21st proteinogenic amino acid, selenocysteine (Sec). Sec is encoded by UGA (STOP) codons which are re-coded to Sec by the presence of a selenocysteine insertion sequence (SECIS) element in the 3'-untranslated region of selenoprotein mRNAs. The SECIS element is bound by several proteins, including SECIS-binding protein 2 (SBP2). Translation of selenoproteins critically depends on the integrity of the SECIS element - SBP2 interaction. Mutations in a SECIS element can abrogate expression of the respective selenoprotein. Mutations in SBP2 impin

BACKGROUND: Many species of filarial nematodes depend on Wolbachia endobacteria to carry out their life cycle. Other species are naturally Wolbachia-free. The biological mechanisms underpinning Wolbachia-dependence and independence in filarial nematodes are not known. Previous studies have indicated that Wolbachia have an impact on mitochondrial gene expression, which may suggest a role in energy metabolism. If Wolbachia can supplement host energy metabolism, reduced mitochondrial function in infected filarial species may account for Wolbachia-dependence. Wolbachia also have a strong influence

Reverse transcription is an essential step in the retroviral life cycle, as it converts the genomic RNA into DNA. In this review, we describe recent developments concerning the initiation step of this complex, multi-step reaction. During initiation of reverse transcription, a cellular tRNA primer is placed onto a complementary sequence in the viral genome, called the primer binding site or PBS. The viral enzyme reverse transcriptase (RT) recognizes this RNA-RNA complex, and catalyzes the extension of the 3' end of the tRNA primer, with the viral RNA (vRNA) acting as template. The initiation st

Queuosine (Q nucleoside) is a highly modified guanosine analog and is present only in the first position of the anticodons of tRNA(Tyr), tRNA(His), tRNA(Asn) and tRNA(Asp). The levels of Q in normal human brain and two different types of human brain tumors (meningiomas and astrocytomas) were determined by using an enzymatic assay method. tRNAs isolated from tumor tissues contained decrease amounts of Q when compared to tRNAs from normal (i.e. non-tumor) brain tissue. There was a relationship between the levels of Q nucleoside and the tumor malignancy in the sense that the deficiency was greate

A method is described to rapidly select and classify many independent near-UV irradiation-resistant Escherichia coli mutants, which include tRNA modification and RNA synthesis control mutants. One class of these mutants was found to be simultaneously deficient in thiamine biosynthesis and in the ability to modify uridine in tRNA to 4-thiouridine, known to be the target for near-UV irradiation. These mutants were found to be unable to make thiazole, a thiamine precursor. The addition of thiazole restores the thiamine deficiency but does not render the cells near-UV irradiation sensitive. BACKGROUND: Camellia tachangensis F. C. Zhang is an endemic Camellia species of the junction of Yunnan, Guizhou and Guangxi Provinces in China. It is characterized by a primitive five-chambered ovary morphology and serves as the botanical source of the renowned "Pu'an Red Tea". Unfortunately, the populations of the species have declined due to the destruction of their habitats by human activities. The lack of mitochondrial genomic resources has hindered research into molecular breeding and phylogenetic evolution of C. tachangensis.

Extracellular vesicles (EVs) are an attractive delivery vehicle with biological activity, intrinsic homing, low immunogenicity, and engineerability; however, challenges remain regarding loading and functional delivery of mRNA. Here, we developed a novel approach to load mRNA through low pH-induced fusion of EVs with lipid nanoparticles (LNPs) to generate hybrid EVs (HEVs). Conventional characterization showed that HEVs preserved classical features of EVs. Single particle analysis revealed successful loading of mRNA and incorporation of LNP components into HEVs. The combined properties from EV

Diagnosing systemic juvenile idiopathic arthritis (sJIA) poses significant challenges. Accumulating evidence has indicated that tRNA-derived fragments (tRFs) play integral roles in the pathogenesis of numerous diseases. Plasma samples were collected from individuals diagnosed with sJIA and healthy controls (HCs) from two medical centers and divided into training and validation cohorts. Small-RNA high-throughput sequencing was employed to investigate the expression profiles of tRFs in the plasma of patients. Aberrantly expressed tRFs in sJIA were validated using quantitative reverse-transcripti
