The potential carcinogenic activity of 5,6-dimethoxysterigmatocystin (DMSC) was examined by oral administration in rats. In Experiment I, all of eight effective ACI/N rats given DMSC in the diet at a concentration of 50 p.p.m. developed neoplastic nodules of the liver. Five rats developed hepatocellular carcinomas and four rats had hemangioendothelial sarcomas of the liver. Two other rats developed osteosarcomas. Cerebral blood flow (CBF) was measured and cerebrovascular resistance (CVR) was calculated in anesthetized spontaneously hypertensive rats (SHR) and normotensive rats (NR) following the administration of incremental dosages of i.v. flunarizine or papaverine. CBF and CVR changes following papaverine were the same in both groups of rats irrespective of the dose of the drug. The effect of flunarizine was much more pronounced in SHR than in NR. The results point out the greater dependency of basal cerebrovascular tone in SHR upon Ca2+ influx into vascular smooth muscle cells.

Eustachian salpingitis was induced with type II collagen in rats. The inflammatory reaction spread from the tubal submucous layer toward the surrounding tissue. Lymphocytes and plasmacytes were the major cells found in the inflammatory infiltrates. Bone resorption with many osteoclasts and mononuclear cells infiltrating into the muscle were also observed. These findings suggest that an immune response against type II collagen can induce eustachian salpingitis.

Pathological evaluations were done in 205 rats with mononuclear cell leukemia. Leukemia was diagnosed in 22.2% of males and 20.4% of females with significant risk beginning at 20 months of age. The Fischer 344 rat is being used increasingly in toxicology studies. There have been few reports in which rats of this strain were used in teratology and reproduction studies, but comparison of teratologic data with other toxic and points and kinetic information would be greatly facilitated by using the same strain. Therefore, the embryotoxic effects of two positive teratogens, aspirin and hydroxyurea, were compared in Fischer rats and in the commonly used Wistar rats. Aspirin was administered in single oral doses of 500 and 625 mg/kg on d 10; hydroxyurea was injected ip at 500 mg/kg on d 11.
