Changing characteristics of rotavirus diarrhea in children younger than five years in urban Bangladesh.

BACKGROUND: Childhood rotavirus diarrhea is still one of the major public health challenges. The present study aimed to determine changing characteristics of rotavirus diarrhea in under-5 children at two periods of time. METHODS: We enrolled 5,357 under-5 children with rotavirus positive in two different time periods; i) 1993-1997 (n = 2,493), and ii) 2008-2012 (n = 2,864) considering beginning and ending of two decades. These children were enrolled in the urban Dhaka Hospital of icddr,b. RESULTS: Overall, proportion of rotavirus was about 25% in 1993-97, which was 42% in 2008-12 (68% rise;

Rotavirus NSP1 inhibits interferon induced non-canonical NFκB activation by interacting with TNF receptor associated factor 2.

TNF receptor associated factor 2 (TRAF2) plays a very important role in cellular innate immune as well as inflammatory responses. Previous studies have reported TRAF2 mediated regulation of TNF and Interferon (IFN) induced canonical and non-canonical activation of NFκB. In this study, we show that rotavirus NSP1 targets TRAF2 to regulate IFN induced non-canonical NFκB activation. Here we found that rotavirus Non-Structural Protein-1 (NSP1) interacts with TRAF2 and degrades it in a proteasome dependent manner. C-terminal part of NSP1 was sufficient for interacting with TRAF2 but it alone could

[Rotavirus and malabsorption. Immunofluorescence microscopy studies of small intestine specimens].

Only 20% of the total of diarrheas in childhood are caused by bacteria . Compared to it, very often the causes for such diseases are viruses, especially rotavirus. The virus - localisation is known to be in enterocytes of the duodenal or jejunal mucosa. So it is probable that a destruction of villous structure linked with diminishing of enzyme activities-especially of lactase--can occur. 53 mucosa specimens were investigated from children by means of fluorescence microscopy. Cryo-cut-slices (8-10 mu) were fixed in acetone and marked with self-prepared rotavirus-hyperimmuneserum (against SA11-v

[Virus and gastrointestinal infections].

Numerous viruses found in the gut are not associated with primary infection or disease of the gastrointestinal tract. Other groups or viruses are not classically associated with infection of the gut but can infect the gastrointestinal tract in immunocompromised individuals (herpes simplex virus, cytomegalovirus, papillomavirus ....). The viruses associated with gastroenteritis represent a large number of taxonomic group. Because these viruses have in general been difficult to cultivate, most members of this group were firstly detected by electron microscopic examination (adenovirus, astrovirus

Improving the vaccine efficacy gap with microbial-derived therapies.

Oral polio and rotavirus vaccines underperform in low-resource settings, a failing linked to distinct microbiome compositions. This Forum examines why empiric probiotics and broad taxonomic approaches are largely ineffective in boosting mucosal vaccine immunity and proposes mechanism-driven microbial therapies that modulate epithelial barriers, utilize immune-modulating metabolites, or mitigate viral interference.

Genetic diversity and evolution of G12P[6] DS-1-like and G12P[9] AU-1-like Rotavirus strains in Brazil.

In the early 2000s, the global emergence of rotavirus (RVA) G12P[8] genotype was noted, while G12P[6] and G12P[9] combinations remained rare in humans. This study aimed to characterize and phylogenetically analyze three Brazilian G12P[9] and four G12P[6] RVA strains from 2011 to 2020, through RT-PCR and sequencing, in order to enhance our understanding of the genetic relationship between human and animal-origin RVA strains. G12P[6] strains displayed a DS-1-like backbone, showing a distinct genetic clustering. G12P[6] IAL-R52/2020, IAL-R95/2020 and IAL-R465/2019 strains clustered with 2019 Nort

Isolation and Characterization of Distinct Rotavirus A in Bat and Rodent Hosts.

Rotavirus A (RVA) causes diarrheal disease in humans and various animals. Recent studies have identified bat and rodent RVAs with evidence of zoonotic transmission and genome reassortment. However, the virological properties of bat and rodent RVAs with currently identified genotypes still need to be better clarified. Here, we performed virus isolation-based screening for RVA in animal specimens and isolated RVAs (representative strains: 16-06 and MpR12) from Egyptian fruit bat and Natal multimammate mouse collected in Zambia. Whole-genome sequencing and phylogenetic analysis revealed that the

Duration of Postdiarrheal Enteric Pathogen Carriage in Young Children in Low-resource Settings.

BACKGROUND: Prolonged enteropathogen shedding after diarrhea complicates the identification of etiology in subsequent episodes and is an important driver of pathogen transmission. A standardized approach has not been applied to estimate the duration of shedding for a wide range of pathogens. METHODS: We used a multisite birth cohort of children 0-24 months of age from whom diarrheal and monthly nondiarrheal stools were previously tested by quantitative polymerase chain reaction for 29 enteropathogens. We modeled the probability of detection of the etiologic pathogen before and after diarrhea

Rotavirus vaccination and intussusception - Science, surveillance, and safety: A review of evidence and recommendations for future research priorities in low and middle income countries.

As of January 2016, 80 countries have introduced rotavirus vaccines into their national immunization programs. Many have documented significant declines in rotavirus-specific and all-cause diarrheal illnesses following vaccine introduction. Two globally licensed rotavirus vaccines have been associated with a low risk of intussusception in several studies. In July 2014, the Rotavirus Organization of Technical Allies Council convened a meeting of research and advocacy organizations, public health experts, funders, and vaccine manufacturers to discuss post-marketing intussusception surveillance a

[Digestive diseases, intestinal infections and intussusception].

There is an improvement in the knowledge of intussusception's pathophysiology. It is due to the development of new rotavirus vaccines, while initially Rotashield was taken off the market because of several cases of intussusception. We report etiopathogenic and epidemiological data, and we describe the diseases associated with an increase risk of intussusception. Intestinal infections (viral or bacterial) are suspected in idiopathic intussusception. Secondary intussusceptions can develop in various intestinal diseases because of intestinal motility abnormalities.

Rotavirus as an important cause of diarrhoea in a hospital for children in Dammam, Saudi Arabia.

A major cause of morbidity and mortality among children in developing countries is acute diarrhoea and rotavirus, a reovirus-like agent, is found to be a leading causative agent. We report here the incidence of rotavirus infection among infants and young children with gastro-enteritis treated at the Children's Hospital in Dammam, Kingdom of Saudi Arabia. One hundred and fifty children, aged between 1 and 60 months, suffering from diarrhoeal illness, were selected as the study cohort; another 150 hospitalized children of the same age group, but not suffering from diarrhoea, served as controls.

Two decades of GSK rotavirus vaccine (RV1): a global analysis to estimate vaccination completion and deaths averted in children under 5 years.

BACKGROUND: Rotavirus vaccination plays a key role in reducing rotavirus gastroenteritis (RVGE) related deaths. This analysis estimated the number of children under 5 years of age fully vaccinated with GSK rotavirus vaccine (RV1) since launch (2004) and its impact on RVGE-related deaths worldwide. RESEARCH DESIGN AND METHODS: To estimate the number of children under 5 years fully vaccinated with RV1 since launch, we used unpublished RV1 market supply data from GSK. The data were extrapolated until the time point when 1 billion doses will have been supplied. The number of RVGE-related deaths a

Isolation and Genomic Characterization of the G6P[1]-Type Sheep Rotavirus in China.

Rotavirus A (RVA) is a prevalent cause of enteric diarrhea in infants, bovine, pigs, and sheep globally. Currently, the G6P[1]-type rotaviruses are prevalent in sheep or goat in Bangladesh, Turkey, and Uganda. However, this genotype has not been reported in Chinese sheep or goat. Therefore, 12 anal swabs were collected from diarrheal sheep in Gansu Province, China, in 2023 and tested for rotavirus using reverse transcription-polymerase chain reaction (RT-PCR). Pathological sections and immunohistochemistry were used to observe pathological changes and rotavirus antigens in the duodenum, respec

Improving rotavirus vaccine coverage: Can newer-generation and locally produced vaccines help?

There are two internationally available WHO-prequalified oral rotavirus vaccines (Rotarix and RotaTeq), two rotavirus vaccines licensed in India (Rotavac and Rotasiil), one in China (Lanzhou lamb rotavirus vaccine) and one in Vietnam (Rotavin-M1), and several candidates in development. Rotavirus vaccination has been rolled out in Latin American countries and is beginning to be deployed in sub-Saharan African countries but middle- and low-income Asian countries have lagged behind in rotavirus vaccine introduction. We provide a mini-review of the leading newer-generation rotavirus vaccines and c
