Primary metastasized extraskeletal Ewing sarcoma of the vulva: report of a case and review of the literature.

PURPOSE: Ewing's sarcoma (ES) and primitive neuroectodermal tumor (PNET) are closely related malignant neoplasms, usually affecting the skeletal system. Extraosseous ES/PNETs are uncommon, with occasional reports of tumors affecting the genitourinary tract. Only few cases of primary vulvar Ewing's sarcoma/PNET have previously been reported. METHODS: We present a patient with primary vulvar Ewing's sarcoma with pulmonary metastasis who presented at the Department of Gynecology and Obstetrics, University of Schleswig-Holstein, Campus Luebeck. RESULTS: The present report is the third case of a

PET surveillance of patients with Ewing sarcomas of the trunk: Must the lower legs be included?

AIM: FDG-PET(/CT) is frequently used in surveillance of Ewing sarcoma (ES) patients. Since ES and PNET (primitive neuroectodermal tumours) may cause peripheral metastases some centers routinely recommend whole body PET acquisition from head to toe what may necessitate repositioning of the patient and thus extending examination time. It is not clear yet whether inclusion of lower leg adds to the diagnostic accuracy of PET scanning, especially in primary tumors of the trunk. PATIENTS, METHOD: 40 patients with ES and PNET of the trunk who were referred for surveillance after primary therapy with

Multifocal Ewing's sarcoma of the brain, calvarium, leptomeninges, spine and other bones in a child.

Ewing's sarcoma (ES) is a malignant osseous neoplasm that mostly affects children and young male adults, and usually presents as a solitary bony lesion. Multifocal ES of the central nervous system is extremely rare, with an incidence ranging from 1.1% to 4.3%. Clinically, ES may mimic osteomyelitis. In this report, we describe the case of an 11-year-old boy who had multiple calvarial, leptomeningeal, spinal and various other bony lesions of ES, which were diagnosed radiologically and histopathologically. To the best of our knowledge, this is the first time that multiple brain, calvarial and sp

Local control and function after twice-a-day radiotherapy for Ewing's sarcoma of bone.

Between February 1982 and December 1987, 39 patients with Ewing's sarcoma of bone have been treated at the University of Florida with a twice-a-day radiotherapy regimen to their primary lesion, 35 with radiation alone and 4 with a combination of radiation and surgery. Although three separate systemic regimens were used (standard risk, 1982-1987 [SR-1]; high-risk, 1982-1984 [HR-2]; and high-risk, 1985-1987 [HR-3]), the radiotherapy regimen remained constant through the years of the study. Those patients whose soft-tissue mass completely regressed after induction chemotherapy received 5040 cGy (

Non-metastatic Ewing's sarcoma: results in 98 patients treated with neoadjuvant chemotherapy.

From June 1983 to December 1987, 98 patients with non-metastatic Ewing's sarcoma of bone were treated by a protocol of neoadjuvant chemotherapy consisting of two months of preoperative chemotherapy with vincristine, adriamycin, and cyclophosphamide followed by local treatment and one year of chemotherapy aimed at preventing recurrence using the same drugs plus dactinomycin. Local treatment consisted of surgery in 39 cases (40%), radiation in 34 (34%), and both surgery and radiation in 25 (26%). After an average follow-up of five years, 51 patients (52%) had remained continuously disease-free (

Pathologic features of extraosseous Ewing's sarcoma: a report from the Intergroup Rhabdomyosarcoma Study.

Eighty-four cases of extraosseous Ewing's sarcoma (EOE) were found during the pathology review of the Intergroup Rhabdomyosarcoma Study I and II. Patients commonly presented during or after adolescence with the most common primary sites including the trunk, extremities, and retroperitoneum. Males were slightly more affected. Histologic sections of 74 tumors in the pathology repository were re-reviewed with attention to rosette formation (positive in 18 cases) and glycogen deposition (++ in 21, + in 36, +/- in 11, and - in 2 of 70 cases examined). Fourteen tumors (7 with rosettes and 7 without)

UBA1 drives cell cycle to promote malignancy and serves as a therapeutic target in Ewing Sarcoma.

BACKGROUND: Ewing sarcoma (ES) is an aggressive malignancy with high metastatic potential and poor prognosis, necessitating novel biomarkers and therapeutic targets. This study investigates the role of the ubiquitin-activating enzyme E1 (UBA1) in ES pathogenesis and its clinical relevance. METHODS: Bioinformatic analysis of GEO datasets was performed. UBA1 expression was validated in clinical samples and ES cell lines (A673, RDES) via Western blot and qPCR. To investigate the functional role and therapeutic potential of UBA1 in ES, we performed siRNA-mediated knockdown followed by a series of

Elimusertib enhances cytotoxic effects of conventional chemotherapy and sensitizes to radiation in preclinical Ewing sarcoma models.

Ewing Sarcoma (EwS) is a highly aggressive malignancy predominantly affecting young individuals, with poor prognosis in metastatic or relapsed cases. This study investigates the therapeutic potential of ataxia telangiectasia and Rad3-related protein (ATR) inhibition using the selective small-molecule inhibitor elimusertib, both as a monotherapy and in combination with cytotoxic drugs or radiotherapy. Elimusertib significantly inhibited cell proliferation and induced apoptosis in EwS cell lines. In a chorioallantoic membrane (CAM) model, elimusertib suppressed tumor initiation and reduced tumor

CuET inhibits Ewing sarcoma of bone progression through modulation of the ESM1-MAPK/ERK signaling axis.

BACKGROUND: Ewing's sarcoma of bone is the second most common primary malignant bone tumor in children and adolescents. Although multimodal therapy has improved 5-year survival to 60-70%, outcomes for relapsed or metastatic disease remain poor (<30%). Notably, copper(II) diethyldithiocarbamate (CuET), a copper-ligand complex generated through the in vivo metabolism of the FDA-approved anti-alcoholism drug disulfiram, has been reported to exhibit antitumor activity across a range of solid malignancies, including colorectal and breast cancers; however, its biological relevance and therapeutic po

Extraosseous Ewing sarcoma of the distal pancreas: diagnostic pitfalls and the critical role of molecular testing.

Primary pancreatic Ewing sarcoma (ES) is an exceedingly rare malignancy, with fewer than 100 confirmed cases reported worldwide. We report the case of a young adult woman presenting with a large solid-cystic pancreatic body-tail mass, initially diagnosed as solid pseudopapillary neoplasm and subsequently misinterpreted as metastatic endometrial stromal sarcoma following resection. Lack of adjuvant therapy and loss to follow-up preceded metastatic recurrence, whereupon targeted immunohistochemistry and molecular analysis established the definitive diagnosis of extraosseous ES with t(11;22)(q24;

StagX1, an isoquinolinone compound, selectively targets CES1-positive Ewing sarcoma cells as a potential therapeutic agent.

StagX1 {ethyl 2-[[2-[2-[(2,3-dihydro-1,4-benzodioxin-6-yl)amino]-2-oxoethyl]-1,2-dihydro-1-oxo-5-isoquinolinyl]oxy]propanoate} is a derivative of isoquinolinone, possessing an ethyl propionate. StagX1 exhibits growth-inhibitory activity in multiple Ewing sarcoma cell lines. To advance StagX1 as a potential lead, we conducted experiments to examine its metabolism and stability in tissue culture media, plasma, liver microsomes, cells and mice. Our studies demonstrate that StagX1 is metabolically unstable and undergoes rapid hydrolysis to its corresponding acid metabolite (StagX1-acid) through cl

Unexpected ¹⁸F-DOPA Uptake in Pediatric Posterior Mediastinal Ewing Sarcoma: A Diagnostic Pitfall in Pediatric Oncology.

¹⁸F-DOPA PET/CT is a well-established imaging modality in pediatric nuclear medicine, with applications in both oncologic and nononcologic conditions. Common indications include brain tumors, neuroendocrine neoplasms, neuroblastoma, and congenital hyperinsulinism. Here, we present an unusual case of ¹⁸F-DOPA uptake in Ewing sarcoma. This case illustrates a potential diagnostic pitfall, emphasizing that ¹⁸F-DOPA avidity is not specific for neuroendocrine or neuroblastic tumors and may occur in non-neuroendocrine malignancies.

Ubiquitin-Specific Protease 6 mRNA Lipid Nanoparticles Ignite Antitumor Immunity and Suppress Tumorigenesis in Ewing Sarcoma.

Ewing sarcoma is an aggressive pediatric cancer that has remained refractory to current therapeutics. Immunotherapy has been unsuccessful in Ewing sarcoma, largely due to poor understanding of how its immune tumor microenvironment is regulated. We recently demonstrated that ubiquitin-specific protease 6 (USP6) can remodel the Ewing sarcoma immune landscape to engender an antitumorigenic tumor microenvironment. USP6 expression in Ewing sarcoma cells enhances surface expression of immunostimulatory ligands and receptors and induces production of multiple chemokines, driving recruitment and activ

Primary extraskeletal intradural Ewing sarcoma with acute hemorrhage: a case report and review of the literature.

BACKGROUND: Spinal cord tumors present a challenge in diagnosis and treatment due to their varied histopathological characteristics. While Ewing sarcoma is a rare malignant tumor typically originating from skeletal bone, cases of primary intradural extraskeletal Ewing sarcoma are exceptionally rare. The similarity of its presentation to other spinal tumors further complicates its identification and management. CASE PRESENTATION: We report a case of a 58-year-old Palestinian male with intradural extraskeletal lumbar Ewing sarcoma. The patient initially presented with lower back pain and bilate
