Spatial buffering during slow and paroxysmal sleep oscillations in cortical networks of glial cells in vivo.

The ability of neuroglia to buffer local increases of extracellular K(+) has been known from in vitro studies. This property may confer on these cells an active role in the modulation and spreading of cortical oscillatory activities. We addressed the question of the spatial buffering in vivo by performing single and double intraglial recordings, together with measures of the extracellular K(+) and Ca(2+) concentrations ([K(+)](out) and [Ca(2+)](out)) in the cerebral cortex of cats under ketamine and xylazine anesthesia during patterns of slow sleep oscillations and spike-wave seizures. In addi

["Benign" form of myoclonic epilepsy in children].

Among 62 children with myoclonic epilepsy who had first seizures between 1 and 10 years, without clinical or radiological evidence of brain lesion, we selected the 16 patients who had exhibited several types of fits and had stopped having seizures for over two years. First seizures occurred between 18 months and 4 years, and they were generalized clonic, tonic-clonic or tonic. After a mean 3 months' period, patients started also to have absence and myoclonic fits. During the period with various types of seizures, that lasted a mean 10 months, patients were ataxic and hyperkinetic, and 11 of th

[Polygraphic findings in bird-like face syndrome with favorable spontaneous evolution].

The syndrome characterized by acquired micrognathia, hypersomnia and periodic apneas during sleep is a rare consequence of mandibular underdevelopment. The pathogenesis is ascribed to incomplete obstruction of the upper airways associated with a hypoexcitability of the respiratory center. Tracheostomy, with the placement of a permanent tracheal cannula, has proved to be the only treatment producing clinical remission of the syndrome. Polygraphic findings were described in a case spontaneously recovered of hypersomnia and periodic apneas with acquired micrognathia due to a bilateral mastoiditis

Neurotoxic N-methyl-D-aspartate lesion of the ventral midbrain and mesopontine junction alters sleep-wake organization.

The dorsal regions of the midbrain and pons have been found to participate in sleep regulation. However, the physiological role of the ventral brainstem in sleep regulation remains unclear. We used N-methyl-D-aspartate-induced lesions of the ventral midbrain and pons to address this question. Unlike dorsal mesencephalic reticular formation lesions, which produce somnolence and electroencephalogram synchronization, we found that ventral midbrain lesions produce insomnia and hyperactivity. Marked increases in waking and decreases in slow wave sleep stage 1 (S1), stage 2 (S2) and rapid eye moveme

Residual effects of ethanol and chlordiazepoxide treatments for alcohol withdrawal.

Eighteen male alcoholics were randomly assigned to one of two alcohol detoxification treatments. One group received a low dose ethanol treatment while the other group received a chlordiazepoxide treatment. This study compares recovery of sleep EEG and clinical symptomatology following these two detoxification treatments. Sleep EEG and clinical measures were obtained for the final medication day and during a 6-day postmedication "recovery" period. The chlordiazepoxide treatment produced suppression of rapid eye movement (REM) sleep lasting for about 4 days and virtually eliminated delta sleep (

Spontaneous and visually driven high-frequency oscillations in the occipital cortex: intracranial recording in epileptic patients.

High-frequency oscillations (HFOs) at ≥80 Hz of nonepileptic nature spontaneously emerge from human cerebral cortex. In 10 patients with extraoccipital lobe epilepsy, we compared the spectral-spatial characteristics of HFOs spontaneously arising from the nonepileptic occipital cortex with those of HFOs driven by a visual task as well as epileptogenic HFOs arising from the extraoccipital seizure focus. We identified spontaneous HFOs at ≥80 Hz with a mean duration of 330 ms intermittently emerging from the occipital cortex during interictal slow-wave sleep. The spectral frequency band of spontan

Impaired hippocampal circuit function underlying memory encoding and consolidation precede robust Aβ deposition in a mouse model of alzheimer's disease.

Current therapeutic strategies for Alzheimer's disease (AD) target amyloid-beta (Aβ) fibrils and high molecular weight protofibrils associated with plaques, but molecular cascades associated with AD may drive neural systems failure before Aβ plaque deposition in AD. Employing hippocampal electrophysiological recordings and dynamic calcium imaging across the sleep-wake cycle in a freely behaving mouse model of AD before Aβ plaques accumulated, we detected marked impairments of hippocampal systems function: In a spatial behavioral task, phase-amplitude coupling (PAC) of the hippocampal theta and

Effects of long-term use of clonazepam on nonrapid eye movement sleep patterns in rapid eye movement sleep behavior disorder.

OBJECTIVE: We aim to analyze in detail the characteristics of nonrapid eye movement (NREM) sleep in drug-free patients with idiopathic rapid eye movement sleep behavior disorder (iRBD). We compare drug-free iRBD patients to both normal controls and drug-free patients with narcolepsy/RBD and evaluate the changes following the long-term use of bedtime clonazepam. PARTICIPANTS AND METHODS: Forty-six participants were recruited: 15 with iRBD (13 men, 2 women; mean age, 65.8±4.39years), 13 with narcolepsy/RBD (10 men, 3 women; mean age, 63.0±6.73years), and 18 normal controls (10 men, 8 women; mea

Sleep-Wake Disorders After Traumatic Brain Injury: Pathophysiology, Clinical Management, and Future.

After experiencing a traumatic brain injury (TBI), the majority of patients will develop sleep-wake disorders (SWD). These can include insomnia, posttraumatic pleiosomnia (increased sleep need), excessive daytime sleepiness (EDS), obstructive and/or central sleep apnea, circadian SWD, and a variety of parasomnias. Untreated SWD may impede the recovery process and can negatively impact mood, metabolic health, cognitive function, and immune function among other processes. Importantly, these patients tend to misperceive their posttraumatic sleep problems. Consequently, interviews performed in sta

The Treatment of Sleep Dysfunction in Neurodegenerative Disorders.

Sleep dysfunction is highly prevalent across the spectrum of neurodegenerative conditions and is a key determinant of quality of life for both patients and their families. Mounting recent evidence also suggests that such dysfunction exacerbates cognitive and affective clinical features of neurodegeneration, as well as disease progression through acceleration of pathogenic processes. Effective assessment and treatment of sleep dysfunction in neurodegeneration is therefore of paramount importance; yet robust therapeutic guidelines are lacking, owing in part to a historical paucity of effective t

Polysomnographic studies of Lesch-Nyhan syndrome.

Three cases of Lesch-Nyhan syndrome (LNS) were examined by polysomnography to assess the brainstem function, and to determine the causes of the neurological manifestations and sudden death in this syndrome. In the two older cases, the amount of slow wave and rapid eye movement (REM) sleep, the REM density and the frequency of REM bursts were decreased. In the youngest case, symmetrical phasic movements of all four limbs were observed at all sleep stages other than REM sleep. Although movements other than these symmetrical body movements appeared to be normal in this case, the frequency of twit

Gammahydroxybutyrate and narcolepsy: a double-blind placebo-controlled study.

We treated 24 patients with narcolepsy for 4 weeks with gammahydroxybutyrate (GHB), 60 mg/kg/night, in a randomized double-blind placebo-controlled cross-over trial. Both clinical and polysomnographic criteria were used to assess the results. Compared to placebo, GHB reduced the daily number of hypnagogic hallucinations (from 0.87 to 0.28; p = 0.008), daytime sleep attacks (from 2.27 to 1.40; p = 0.001) and the severity of subjective daytime sleepiness (from 1.57 to 1.24 on a 0-4 scale; p = 0.028). The number of daily cataplexy attacks was reduced from 1.26 at baseline to 0.56 after 4 weeks of

Differential activation within costal diaphragm during rapid-eye-movement sleep in cats.

Simultaneous recordings of the diaphragmatic electromyogram (EMG) were made from two separate regions of the costal diaphragm in six normal cats. The diaphragmatic activities were always synchronous and the amplitudes and rates of rise were similar during slow-wave sleep. In contrast, during natural rapid-eye-movement (REM) sleep, different activity was often present in the two leads. These differences were in the time of onset and offset, as well as in the amplitude and spike patterns, and occurred in approximately 5-20% of the diaphragmatic bursts averaged over the entire REM sleep period. Acute Sleep Disruption Does Not Diminish Pulsatile Growth Hormone Secretion in Pubertal Children.

CONTEXT: In children, growth hormone (GH) pulses occur after sleep onset in association with slow-wave sleep (SWS). There have been no studies in children to quantify the effect of disrupted sleep on GH secretion. OBJECTIVE: This study aimed to investigate the effect of acute sleep disruption on GH secretion in pubertal children. METHODS: Fourteen healthy individuals (aged 11.3-14.1 years) were randomly assigned to 2 overnight polysomnographic studies, 1 with and 1 without SWS disruption via auditory stimuli, with frequent blood sampling to measure GH. RESULTS: Auditory stimuli delivered du

[Regulation of sleep].

Sleep regulation calls for 3 processes: the first one is the homeostatic process increasing during wakefulness and decreasing during sleep, the second one is the circadian process depending on the circadian oscillator which controls temperature and alertness rhythms, and the third one is the ultradian process: it determines the NREM/REM periodicity. In the 2 process model of sleep regulation, the power density of the delta band (0.75-4.5 Hz) called slow wave activity (SWA) and obtained by spectral analysis, is supposed to reflect the variations of a homeostatic recovery process (process S) tha

Corpus callosum agenesis: does sleep matter?

Agenesis of the corpus callosum is a brain malformation that can lead to intellectual, behavioural, social and emotional impairments, with outcomes varying based on severity. Sleep disturbances are common in children with this disorder, and some studies have evidenced the role of corpus callosum in interhemispheric synchronisation that may influence sleep patterns. This narrative review attempted to intercept available information on sleep in patients with isolated Agenesis of the corpus callosum, partial and complete, across different ages. Since literature about this issue is underdeveloped,

Dynamic brain-heart interaction in sleep characterized by variational phase-amplitude coupling framework.

Sleep is a complex physiological state characterized by synchronized cortical and cardiac oscillations, which reflect dynamic communication and interaction between the central (CNS) and autonomic (ANS) nervous systems, crucial for maintaining homeostasis and overall health. However, the dynamic interplay between CNS and ANS rhythmicities in sleep remains unclear. Here, we present a variational phase-amplitude coupling framework that associates frequency modulations of the electroencephalogram and cardiac R-peak intervals across sleep dynamics. We validate the robustness of our method on spurio

Reorganization of sleep architecture after surgery for OSAHS.

CONCLUSION: Genioglossus advancement and hyoid suspension (GAHM) plus uvulopalatopharyngoplasty (UPPP) can decrease the apnea hypopnea index (AHI) and arousal index, especially respiratory-related arousals. Correspondingly, sleep architecture is reorganized, which is expressed postoperatively as an increase in slow wave sleep and a decrease in lighter sleep. OBJECTIVE: To explore the reorganization of sleep patterns after GAHM plus UPPP for severe obstructive sleep apnea hypopnea syndrome (OSAHS). PATIENTS AND METHODS: GAHM plus UPPP was performed on 31 patients with severe OSAHS (AHI > 40).

Influences of a light-dark profile and the pineal gland on the hypnotic activity of melatonin in mice and rats.

We have investigated the influences of the light-dark cycle and the pineal gland on the hypnotic activity of melatonin in rats and mice. The results showed that melatonin significantly shortened time to sleep onset and wakefulness time, increased slow wave sleep, paradoxical sleep, and total sleep time in rats during the light phase of a 12-h light:12-h dark cycle, by electroencephalogram recording. However, during the dark phase it had almost no significant sleep-promoting effect except shortened time to sleep onset. Melatonin exhibited more potent sleep-promoting effect in rats exposed to co
