Chemogenetic induction of CA1 hyperexcitability triggers indistinguishable autistic traits in asymptomatic mice differing in Ambra1 expression and sex.

Among the genomic alterations identified as risk factors in mice models of autism spectrum disorders (ASD), heterozygous deletion of Ambra1 (Activating Molecule in Beclin1-Regulated Autophagy) triggers an ASD phenotype associated with hippocampal hyperexcitability exclusively in the female sex although Ambra1 protein is comparably expressed in the hippocampus of symptomatic females and asymptomatic males. Given the intricate relationship between Ambra1 deficiency and sex in the etiology of ASD, we took advantage of asymptomatic mice including Ambra1+/- males and wild-type (Wt) mice of both sex

Review: Cross-breeding, advanced reproductive technologies, and genetic selection in twelve dairy production systems in Africa.

The supply and demand for milk in Africa have been rising due to factors such as population growth, urbanization, increasing incomes, and improved living standards. Dairy production significantly impacts the economic and social development of African countries, contributing over 10% to the agricultural gross domestic product in countries such as Ethiopia, Kenya, and Tanzania. The ongoing development of the African dairy industry can be attributed, to a certain extent, to the utilization of cross-breeding strategies and advanced reproductive technologies such as artificial insemination, embryo

Metagenomic insights into the wastewater resistome before and after purification at large‑scale wastewater treatment plants in the Moscow city.

Wastewater treatment plants (WWTPs) are considered to be hotspots for the spread of antibiotic resistance genes (ARGs). We performed a metagenomic analysis of the raw wastewater, activated sludge and treated wastewater from two large WWTPs responsible for the treatment of urban wastewater in Moscow, Russia. In untreated wastewater, several hundred ARGs that could confer resistance to most commonly used classes of antibiotics were found. WWTPs employed a nitrification/denitrification or an anaerobic/anoxic/oxic process and enabled efficient removal of organic matter, nitrogen and phosphorus, as

High carriage and possible hidden spread of multidrug-resistant Salmonella among asymptomatic workers in Yulin, China.

Food workers have frequent contact with unprocessed foods, but their carriage of Salmonella and potential influence on public health have not been comprehensively assessed. We investigated Salmonella carriage among food workers compared with non-food workers based on occupational health screening of 260,315 asymptomatic workers over an 8-year surveillance period in Yulin, China. We confirmed that healthy carriers serve as natural reservoirs for Salmonella, with higher carriage rates in food workers than non-food workers. The isolates from food workers also exhibited greater serovar diversity a

[Analysis of cortical network dynamics in the behavioral state of a mouse model of autism].

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by behavioral abnormalities such as poor social communication and stereotyped/repetitive behaviors. Functional dynamics among multiple cortical areas are associated with processing sensory information and planning and executing behavioral expressions. However, the reconfiguration of large-scale functional network dynamics during behaviors remains to be elucidated in ASD. In this review, we describe our virtual reality (VR) based real-time imaging system which allowed us to investigate wide-field cortical activity in

An Old Acquaintance: Could Adenoviruses Be Our Next Pandemic Threat?

Human adenoviruses (HAdV) are one of the most important pathogens detected in acute respiratory diseases in pediatrics and immunocompromised patients. In 1953, Wallace Rowe described it for the first time in oropharyngeal lymphatic tissue. To date, more than 110 types of HAdV have been described, with different cellular tropisms. They can cause respiratory and gastrointestinal symptoms, even urinary tract inflammation, although most infections are asymptomatic. However, there is a population at risk that can develop serious and even lethal conditions. These viruses have a double-stranded DNA g

Illuminating the Mechanisms Underlying Sex Differences in Cardiovascular Disease.

Sex is a key risk factor for many types of cardiovascular disease. It is imperative to understand the mechanisms underlying sex differences to devise optimal preventive and therapeutic approaches for all individuals. Both biological sex (determined by sex chromosomes and gonadal hormones) and gender (social and cultural behaviors associated with femininity or masculinity) influence differences between men and women in disease susceptibility and pathology. Here, we focus on the application of experimental mouse models that elucidate the influence of 2 components of biological sex-sex chromosome

Epigenetic Clock Analysis in Children With Fetal Alcohol Spectrum Disorder.

BACKGROUND: Fetal alcohol spectrum disorder (FASD) is characterized by severe clinical impairment, considerable social burden, and high mortality and morbidity, which are due to various malformations, sepsis, and cancer. As >50% of deaths from FASD occur during the first year of life, we hypothesized that there is the acceleration of biological aging in FASD. Several recent studies have established genome-wide DNA methylation (DNAm) profiles as "epigenetic clocks" that can estimate biological aging, and FASD has been associated with differential DNAm patterns. Therefore, we tested this hypothe

Intronic Variant in CNTNAP2 Gene in a Boy With Remarkable Conduct Disorder, Minor Facial Features, Mild Intellectual Disability, and Seizures.

Introduction: Mutations in the contactin-associated protein-like 2 (CNTNAP2) gene (MIM#604569) encoding for CASPR2, a cell adhesion protein of the neurexin family, are known to be associated with autism, intellectual disability, and other neuropsychiatric disorders. A set of intronic deletions of CNTNAP2 gene has also been suggested to have a causative role in individuals with a wide phenotypic spectrum, including Pitt-Hopkins syndrome, cortical dysplasia-focal epilepsy syndrome, Tourette syndrome, language dysfunction, and abnormal behavioral manifestations. Case presentation: A 10-years-old

Examining the Overlap between Autism Spectrum Disorder and 22q11.2 Deletion Syndrome.

22q11.2 deletion syndrome (22q11.2DS) is a genomic disorder reported to associate with autism spectrum disorders (ASDs) in 15-50% of cases; however, others suggest that individuals with 22q11.2DS present psychiatric or behavioral features associated with ASDs, but do not meet full criteria for ASD diagnoses. Such wide variability in findings may arise in part due to methodological differences across studies. Our study sought to determine whether individuals with 22q11.2DS meet strict ASD diagnostic criteria using research-based guidelines from the Collaborative Programs of Excellence in Autism

Genetic disruption of ankyrin-G in adult mouse forebrain causes cortical synapse alteration and behavior reminiscent of bipolar disorder.

Genome-wide association studies have implicated the ANK3 locus in bipolar disorder, a major human psychotic illness. ANK3 encodes ankyrin-G, which organizes the neuronal axon initial segment (AIS). We generated a mouse model with conditional disruption of ANK3 in pyramidal neurons of the adult forebrain (Ank-G cKO). This resulted in the expected loss of pyramidal neuron AIS voltage-gated sodium and potassium channels. There was also dramatic loss of markers of afferent GABAergic cartridge synapses, resembling the cortical microcircuitry changes in brains from psychotic patients, and suggesting

Vaccination against ectoparasites.

Ectoparasites of livestock are of great economic and social importance but their effective control remains difficult. The feasibility of vaccination as a novel control measure was established over a decade ago with the commercial release of a recombinant vaccine against the cattle tick Boophilus microplus. Since then, research has continued on ticks and other ectoparasites. While some ectoparasite species will undoubtedly be refractory to immunological control, for others there has been a steady accumulation of knowledge of partially protective antigens, now accelerating through the applicatio

Poliomyelitis outbreak in an unvaccinated community in The Netherlands, 1992-93.

An outbreak of poliomyelitis occurred in the Netherlands between September, 1992, and February, 1993, after 14 years without endemic cases. The outbreak was due to poliovirus type 3 and involved 71 patients, of whom 2 died and 59 had paralysis. The patients were aged between 10 days and 61 years (median 18 years). None of the patients had been vaccinated, and all but 1 belonged to a socially and geographically clustered group of people who refuse vaccination for religious reasons. Control measures were taken within 5 days of notification of the first patient and included a wide offer of vaccin

The polio epidemic in The Netherlands, 1992/1993.

Starting September 1992, an outbreak of poliomyelitis occurred in the Netherlands. Until now a total of 67 cases have been notified. These were the first indigenous cases since 1978. None of the patients had been vaccinated before, and all but one belong to small Protestant groups that refuse vaccination for religious reasons. The age of the patients varies from several days to 61 years (mean age 19) and the male to female ratio is 1.6:1.0. The epidemic is caused by wild poliovirus type 3. Partial genomic sequencing showed closest resemblance to a virus isolated in India in 1991. The epidemiol

Challenges in Academic Authorship and Citation Metrics: Navigating Inflation, Biases, and the Rise of Evidence Synthesis.

This paper examines the challenges in academic publishing related to authorship inflation, citation bias, and the overreliance on quantitative metrics to evaluate research impact. Authorship inflation, including "gift authorship," occurs when individuals with minimal contributions are listed as co-authors, often due to institutional or political norms, inflating academic profiles without reflecting actual input. The study also discusses how citation patterns differ across disciplines, with high-citation fields like biomedical sciences benefiting from larger, faster publications, while research

Genetic Testing in Breast Cancer: Narrative Review and Clinical Insights.

BACKGROUND: Identifying hereditary breast cancer is increasingly important, as germline pathogenic variants not only influence lifetime cancer risk but also guide surveillance, preventive strategies and targeted treatments. While breast cancer genes (BRCA1 and BRCA2) remain central to hereditary breast and ovarian cancers, progress in cancer genetics has shifted clinical practice toward broader multigene testing approaches. METHODS: We reviewed the recent literature on the molecular basis of hereditary breast cancer, current genetic testing strategies, guideline-based indications, and the psy
