Enantioselective fate of dinotefuran from tomato cultivation to home canning for refining dietary exposure.

Understanding the enantioselective fate of chiral neonicotinoid dinotefuran is of vital importance for accurate dietary exposure assessment and food safety regulation. The study investigated the enantioselectivity in respect to dissipation, metabolism, and removal, of dinotefuran from tomato cultivation to tomato paste processing. The chiral analytical method of dinotefuran, UF and DN was developed in tomato using ultrahigh performance supercritical fluid chromatography/tandem mass spectrometry. Under greenhouse cultivation R-dinotefuran preferentially degraded (T1/2, 9.1-12.6 days), resulting

Plant-Microbe Interaction: Transcriptional Response of Bacillus Mycoides to Potato Root Exudates.

Beneficial plant-associated bacteria play an important role in promoting growth and preventing disease in plants. The application of plant growth-promoting rhizobacteria (PGPR) as biofertilizers or biocontrol agents has become an effective alternative to the use of conventional fertilizers and can increase crop productivity at low cost. Plant-microbe interactions depend upon host plant-secreted signals and a reaction hereon by their associated bacteria. However, the molecular mechanisms of how beneficial bacteria respond to their associated plant-derived signals are not fully understood. Asses

Evaluation of tablet punch configuration on mitigating capping by a quality by design approach.

Capping is a common problem in the manufacture of some types of tablets and unless resolved, the tableting process cannot proceed. Hence, all factors that can help to lessen the likelihood of capping without unnecessarily reduce turret speed and/or compaction force would be tenable. This study investigated the influence of tablet punch configuration on mitigation of tablet capping. Tablets were prepared from high-dose paracetamol-potato starch granules in a rotary tablet press with flat face plain (FFP), flat face bevel edge (FFBE) and flat face radius edge (FFRE) punch configurations. The dir

Effects of Lime and Organic Amendments Derived from Varied Source Materials on Cadmium Uptake by Potato.

Repeated applications of Cd-rich phosphate fertilizers have resulted in elevated concentrations of this toxic element in some New Zealand soils. Exceedance of the food safety standard for Cd (0.1 mg kg fresh weight) has been reported for potato ( L.). Composts may efficiently sorb Cd in soil and therefore reduce its phytoavailability, leading to reduced uptake by plants. We aimed to determine the potential of various composts, shredded corn stover, and lime at two different rates to reduce the transfer of Cd from a soil (containing 1.45 mg kg Cd) to potato (var. 'Nadine'). In the control, the

Toughing It Out--Disease-Resistant Potato Mutants Have Enhanced Tuber Skin Defenses.

Common scab, a globally important potato disease, is caused by infection of tubers with pathogenic Streptomyces spp. Previously, disease-resistant potato somaclones were obtained through cell selections against the pathogen's toxin, known to be essential for disease. Further testing revealed that these clones had broad-spectrum resistance to diverse tuber-invading pathogens, and that resistance was restricted to tuber tissues. The mechanism of enhanced disease resistance was not known. Tuber periderm tissues from disease-resistant clones and their susceptible parent were examined histologicall

Homocysteine over-accumulation as the effect of potato leaves exposure to biotic stress.

Homocysteine (Hcy) is a naturally occurring intermediate metabolite formed during methionine metabolism. It has been well documented that its excess can be extremely toxic to mammalian, yeast and bacterial cells. In spite of the metabolic value of Hcy known for decades, the role of this amino acid in the plant response to stress has not been recognized yet. In the presented study, using potato plant (Solanum tuberosum L.) and Phytophthora infestans as a model system, the presence and tissue localization of Hcy in leaves was examined by an immunohistochemical method. The over-production of Hcy …

Purification, crystallization and preliminary X-ray diffraction analysis of a fungal saponin-detoxifying enzyme.

Tomatinase, an extracellular enzyme belonging to family 3 of the glycosyl hydrolases, is produced by the fungal tomato-leaf pathogen Septoria lycopersici and detoxifies the saponin alpha-tomatine. An efficient strategy for purification of the enzyme from fungal culture medium has been developed. Single crystals have been grown by vapour diffusion at 289 K from 17.5%(w/v) PEG 4K, 5%(v/v) 2-propanol and 0.1 M sodium acetate pH 4.5 as precipitant. When cryoprotected at 100 K, these crystals diffract to at least 3.0 A and belong to space group P2(1)2(1)2. Based on an estimated molecular weight of

Production of thaxtomin a by two species of Streptomyces causing potato scab.

A total of nine isolates of streptomycetes were isolated from scab lesions on potato tubers. Five out of them were pathogenic on potato minitubers and four of the pathogenic isolates produced thaxtomin A in infected tubers tissues. The lesion surface areas induced by thaxtomin A were highest in treatment of the minitubers with extract of OMB inoculated with S-6 and S-7, intermediate with that inoculated with S-4 and lowest with S-3. The pathogenic isolates were identified by their colour of aerial mycelia, melanin pigment productivity (+ or -), the type of spore chains morphology and carbon ut

Effect of high-pressure treatment on the texture of cherry tomato.

The effect of high-pressure treatment (200-600 MPa for 20 min) on the texture of cherry tomatoes and on the key softening enzymes (pectinmethylesterase and polygalacturonase) was investigated. When subjected to high-pressure treatment whole cherry tomatoes showed increasing textural damage with increasing pressures up to 400 MPa. However, treatment at pressures above 400 MPa (500-600 MPa) led to less apparent damage than treatment at 300 and 400 MPa; the tomatoes appearing more like the untreated samples. These visual changes were reflected in the texture (firmness) and amount of cell rupture

Chlorophyll fluorescence imaging for early visual detection and measurement of postharvest discoloration: Potato greening and broccoli yellowing.

Chlorophyll serves as both physiological basis and fundamental pigment underlying healthy coloration of numerous fruits and vegetables. Yet conventional visual inspection, point-level color measurement, and destructive chlorophyll assays lack sensitivity for early, non-destructive detection of discoloration driven by abnormal chlorophyll metabolism in postharvest produce. This study developed a simple chlorophyll fluorescence imaging system to enable early detection and measurement of two representative postharvest discoloration phenomena: light-induced greening in potatoes and senescence-driv

More is not always better: phosphorus context shapes the effectiveness of single-strain and consortium plant growth-promoting bacteria in tomato.

Phosphorus availability determines whether single or consortium PGPB maximize tomato performance, demonstrating that rational inoculant design should prioritize functional complementarity over increasing microbial complexity. Phosphorus (P) availability is a major constraint for crop development and productivity, and beneficial bacteria are increasingly proposed to enhance plant P-acquisition efficiency. However, their comparative performance remains variable across studies and experimental contexts. This study investigated whether single plant growth-promoting bacteria (PGPB) strains and a de

Thiamin addition to soil increases potato tuber thiamin content under greenhouse conditions.

Thiamin is essential for human health, but humans do not synthesize it and must consume it through regular dietary intake. Plants synthesize thiamin in photosynthetic tissues to support various primary metabolic pathways. In addition, plants can also absorb thiamin from the soil. Interestingly, plant growth can be improved by supplying exogenous thiamin, but this effect has not been investigated in potato (Solanum tuberosum). Here, we report the effect of soil thiamin amendment on yield and tissue thiamin content of three potato varieties grown in a greenhouse. We watered plants with different…

Comprehensive evaluation of the novel purple-fleshed tomato germplasm black 'pearl' and analysis of anthocyanin synthesis mechanisms and components.

Anthocyanins endow tomatoes with richer nutritional characteristics. This study provides a comprehensive evaluation of the anthocyanin-rich tomato germplasm black 'Pearl,' which features an appealing purple flesh. The high levels of anthocyanins, antioxidant enzymes, vitamin C, and vitamin E confer a strong total antioxidant capacity. The organic acid and soluble sugar content in flesh are slightly lower, and it has a mild bitterness, with storage characteristics inferior to red-fleshed tomatoes. The correlation analysis between transcriptomics and metabolomics indicates that multiple structur

Structure guided development of phenoxyethanol based N-aryloxyethyl heterocycles for the control of Phytophthora infestans.

BACKGROUND: Late blight caused by the oomycete Phytophthora infestans remains one of the most devastating diseases threatening global potato and tomato production. Overuse of a narrow range of fungicides has sped up resistance, making it urgent to develop new agents that work differently and are safer. RESULTS: In this study, a series of N-aryloxyethyl heterocycles were rationally designed by integrating the aryloxyethyl scaffold of phenoxyethanol with bioactive heterocyclic frameworks. Among the 40 synthesized derivatives, compound 8c exhibited the highest anti-oomycete activity against P. i

Assessment of health effects of potato crop phytopharmaceuticals and storage products in a murine model.

Potatoes are among the most widely consumed staple foods worldwide, but their cultivation and storage frequently involve multiple phytopharmaceutical products (PPPs), raising concerns about the health risks of dietary pesticide residues. The health effects of multi-residue PPPs used in potato cultivation were assessed in an in vivo murine model, involving 36 mice, and evaluated through omics analyses. Two field cultivation methods (conventional and organic) and two post-harvest storage conditions (using 1,4-dimethylnaphthalene and mint essential oil as sprout inhibitor treatments) were conside

Assessing food by-products macrocomposition by FTIR microspectroscopy.

Food by-products offer a promising opportunity for extracting valuable compounds that can be used in the food, pharmaceutical, and polymer industries. However, unpredictable variations in the chemical composition and spatial distribution of the various components within these biological matrices create challenges for new valorization processes. These inconsistencies can lead to variable recovery efficiency and differing quality of extracts. Understanding the chemical composition and spatial distribution of these components is essential, as it will facilitate the effective valorization of these
