Transcranial magneto-acoustic-electrical stimulation is a new non-invasive neuromodulation technology, in which the induced electric field generated by the coupling effect of ultrasound and static magnetic field are used to regulate the neural rhythm oscillation activity in the corresponding brain region. The purpose of this paper is to investigate the effects of transcranial magneto-acoustic-electrical stimulation on the information transfer and communication in neuronal clusters during memory. In the experiment, twenty healthy adult Wistar rats were randomly divided into a control group (fiv

A nanocomposite based on chitosan and montmorillonite was developed as carrier to improve oral bioavailability of oxytetracycline. The nanocomposite was prepared by simple solid-liquid interaction and loaded with the drug. The loaded nanocomposite was characterized by X-ray powder diffraction, thermal analysis, FTIR spectroscopy and zeta potential. Caco-2 cell cultures were used to evaluate in vitro cytotoxicity and drug permeation. Confocal laser scanning microscopy was also performed to evaluate the eventual entrapment of drug into the Caco-2 cells. Results showed that the nanocomposite was

The effect of brief trains of electrical stimulation, at 2, 3 and 20 x threshold (T), of cutaneous afferents in the medial plantar nerve on the discharges of single medial gastrocnemius static and dynamic gamma-efferents has been investigated at rest and during locomotion in a decerebrate cat preparation. The units were classified as dynamic (10 units) or static (10 units) indirectly on the basis of their resting and locomotor discharge characteristics. Responses were assessed by calculating the change in mean gamma-rate during the 100 ms after stimulus onset compared with a control period. At

BACKGROUND: Major depressive disorder (MDD) is a common and intractable mood disorder. Electroconvulsive therapy (ECT) is a common means of brain electrical stimulation for the treatment of MDD, but the neurobiological mechanism of its clinical symptom relief effect is still to be explored. This study aims to explore how ECT plays a role in depression remissions by investigating the changes of static and dynamic brain network characteristics in MDD patients before and after ECT.
METHODS: Resting-state functional magnetic resonance imaging (fMRI) scans were obtained from nine MDD patients twic

Large-conductance Ca2+- and voltage-activated potassium (BK) channels display a near-linear current-voltage relationship in excised membrane patches but exhibit inward rectification in intact cells due to block by intracellular cations. This rectification arises from electrostatic interactions between endogenous cationic blockers and negatively charged residues lining the inner pore. Here, we examined how the introduction of an additional negative charge within the inner vestibule affects BK channel permeation. Phenylalanine 315 in the S6 segment of the BK α-subunit was substituted with aspart

BACKGROUND: Chimeric antigen receptor T-cell (CAR-T) therapy has achieved remarkable success in hematologic malignancies, yet demonstrates limited efficacy in solid tumors, including hepatic cancers. T-cell exhaustion and insufficient persistence represent major obstacles. We hypothesized that optimizing both manufacturing processes and CAR structural design could reduce exhaustion and enhance therapeutic outcomes in immunocompetent mouse models of primary and metastatic liver malignancies.
METHODS: We systematically compared antibody-based versus bead-based activation methods, evaluating the

Enzymes use electrostatic interactions to recognize their substrates, preorganize active sites, and stabilize reaction transition states. Formamidopyrimidine-DNA glycosylase (Fpg) is a bacterial enzyme that repairs a pro-mutagenic DNA lesion, 8-oxoguanine; its human homologues are important for cancer prevention. General acid-base catalysis in the active site of Fpg requires a finely tuned proton transfer between Pro1 and Glu2 residues. To assess the protonation state of the active site experimentally, we used EPR spectroscopy with a novel imidazolidine nitroxyl spin label responsive in the ph

Nisin, a GRAS-approved antimicrobial peptide, suffers from limited aqueous solubility hindering practical applications. This study engineered two modified chitin nanocarriers-surface-deacetylated chitin nanofibers (S-ChNF) and oxidized chitin nanowhiskers (O-ChNW)-to improve nisin delivery. At pH 4 and a mass ratio of 1:2, both types of nanochitin achieved their highest encapsulation efficiency (EE) and loading capacity (LC): S-ChNF (EE 92.23 %, LC 30.74 %) and O-ChNW (EE 79.62 %, LC 26.53 %). Spectroscopic analyses confirmed nisin binding via hydrogen bonding and electrostatic interactions. M

This study characterizes the steroidal saponin diosgenin by theoretical and experimental spectroscopic techniques. Theoretical simulations were performed using the DFT/B3LYP/6-311++G(d,p) basis set to simulate spectroscopic, structural and other properties. Optimized geometries from simulations and experiments showed strong agreement, with R2 value of 0.99846 for bond lengths and 0.88092 for bond angles. Vibrational spectra revealed distinctive peaks for the methyl, methylene, and methine groups in diosgenin. Solvent-solute interactions on the Frontier Molecular Orbitals (FMO), Molecular Elect

Substrate specificity of an enzyme is an important characteristic of its mechanism of action. Investigation of the nucleotide specificity of Plasmodium falciparum succinyl-CoA synthetase (SCS; PfSCS) would provide crucial insights of its substrate recognition. Charged gatekeeper residues have been shown to alter the substrate specificity via electrostatic interactions with approaching substrates. The enzyme kinetics of recombinant PfSCS (wild-type), generated by refolding of the individual P. falciparum SCSβ and Blastocystis SCSα subunits, demonstrated ADP-forming activity (KmATP = 48 µm). Fu

Protein-nanoparticle interactions are garnering attention due to their potential impacts on human health and environmental contamination. The colloidal properties of nanoparticles (NPs) in aqueous media may differ in the presence of natural materials such as salts and proteins. In this study, the interactions between bovine serum albumin (BSA) and fumed hydrophilic silicon dioxide (SiO2) NPs were studied in aqueous solutions under variable pH or ion composition. Investigation of hydrodynamic diameter and zeta potential changes to nanoparticles upon addition of BSA, the adsorption of BSA to the

In order to reveal the essence of the pharmaceutical incompatibility, the cooperativity effects of the drug-drug intermolecular π∙∙∙π and H∙∙∙O H-bonding interactions involving hydration were evaluated in the phenobarbital∙∙∙paracetamol∙∙∙H2O complex at the M06-2X/6-311++G** and MP2/6-311++G** levels. The thermodynamic cooperativity effects were also investigated by the statistical thermodynamic method. The results show that the π∙∙∙π stacking ternary complexes with the moderate anti-cooperativity effects are dominant in controling the aggregation process of phenobarbital, paracetamol, and H2O

Therapeutic agents aimed at inhibiting a single molecular target have not been successful in cancer therapy, but rather they impart resistance. However, multi-target inhibitors have shown promising results in circumventing the development of resistance and inducing apoptosis in cancer cells/tissues. In this study, we encapsulated doxorubicin and celecoxib in a single liposome at a ratio of 1:10. These dual drug-encapsulated liposomes showed excellent anticancer activity compared to individually encapsulated liposomes. The expression of key proteins such as AKT and COX-2 was suppressed, which s

Host jump can result in deadly pandemic events when avian influenza A viruses broaden their host specificity and become able to infect mammals, including humans. Haemagglutinin-the major capsid protein in influenza A viruses-is subjected to high rate mutations, of which several occur at its "head": the receptor-binding domain that mediates specific binding to host cell receptors. Such surface-changing mutations may lead to antigenically novel influenza A viruses hence in pandemics by host jump and in vaccine escape by antigenic drift. Changes in haemagglutinin surface electrostatics have been

Although phospholipidosis (PLD) often affects drug development, there is no convenient in vitro or in vivo test system for PLD detection. In this study, we developed an in silico PLD prediction method based on the PLD-inducing mechanism. We focused on phospholipid (PL)-compound complex formation, which inhibits PL degradation by phospholipase. Thus, we used some molecular interactions, such as electrostatic interactions, hydrophobic interactions, and intermolecular forces, between PL and compounds as descriptors. First, we performed descriptor screening for intermolecular force and then develo

Porous silicon (pSi) is a prosperous biomaterial, biocompatible, and biodegradable. Obtaining regularly functionalized pSi surfaces is required in many biotechnology applications. Silane-PEG-NHS (triethoxysilane-polyethylene-glycol-N-hydroxysuccinimide) is useful for single-molecule studies due to its ability to attach to only one biomolecule. We investigate the functionalization of pSi with silane-PEG-NHS and compare it with two common grafting agents: APTMS (3-aminopropylotrimethoxysilane) as electrostatic linker, and APTMS modified with glutaraldehyde as covalent spacer. We show the arrange

The influenza virus has received extensive attention due to the recent H1N1 pandemics originating from swine. This study reports a label-free, highly sensitive, and selective electrical immunosensor for the detection of influenza virus H1N1 based on dielectrophoretically deposited single-walled carbon nanotubes (SWCNTs). COOH-functionalized SWCNTs were deposited on a self-assembled monolayer of polyelectrolyte polydiallyldimethyl-ammonium chloride (PDDA) between two gold electrodes by dielectrophoretic and electrostatic forces, which resulted in reproducible, uniform, aligned, and aggregation-
