Crucial involvement of transforming growth factor beta (TGF-β)/Smad signaling regulation by miR-204-5p in cataract and posterior capsular opacification.

UNLABELLED: Worldwide, cataracts remain the leading cause of irreversible blindness, primarily driven by age-related changes and free radical-mediated lens damage. Posterior capsular opacification (PCO) is a common complication following cataract surgery, arising from aberrant proliferation and epithelial-mesenchymal transition (EMT) of residual lens epithelial cells (LECs). The transforming growth factor-β (TGF-β)/Smad signaling cascade is among the most critical pathways underlying both cataractogenesis and PCO, as it drives EMT and subsequent fibrotic alterations. MicroRNAs have emerged as

Unilateral Lattice Corneal Dystrophy Associated With c.1864A>T Variant in the Transforming Growth Factor Beta-Induced Gene: A Rare Case Report.

We describe a patient with unilateral lattice corneal dystrophy (LCD) associated with a previously unreported pathogenic transforming growth factor beta-induced (TGFBI) variant (c.1864A>T, p.(Asn622Tyr)), contributing to the expanding phenotypic spectrum of non-R124C TGFBI-related LCD. A 58-year-old male patient presented with complaints of recurrent epithelial erosions and progressive vision loss in his right eye for the past 20 years. On slitlamp biomicroscopic examination of the patient's right eye, thick, branched lattice lines extended from the limbus to the central cornea. The lesions ex…

Direct and indirect effects of transforming growth factor-beta on osteoclast-mediated bone remodeling using a new in vitro bone matrix model.

Transforming growth factor-beta (TGF-β), a cytokine embedded in the bone matrix, is released during bone resorption, influencing osteoclast differentiation and coupling factor production, which affect osteoblasts and osteocytes. This study investigates the role of TGF-β in bone remodeling using an in vitro model with calcium phosphate-coated plates covalently bonded to latent TGF-β (LTGF-β(+)-CaP plates). This model replicates the natural release of TGF-β and its effects on RAW264 macrophage-like cells, which differentiate into osteoclasts upon stimulation of RANKL. Cells cultured on LTGF-β(+)

The Association Between the Transforming Growth Factor Beta-1 -509C>T Gene Polymorphism and Primary Open Angle Glaucoma in North Eastern Iran.

BACKGROUND: Glaucoma is a common cause of irreversible blindness. Transforming growth factor beta-1(TGF-β1) is the main isoform of TGF-β superfamily in the eye. Overexpression of TGF-β1 is shown to be related with the glaucoma. Studies have shown that the presence of mutant T allele of TGF-β1 -509C>T polymorphism (rs1800469) is associated with increased gene expression. So, in present study, association of the TGF-β1-509C>T gene polymorphism and primary open angle glaucoma (POAG) in patients from north east of Iran was investigated. METHODS: A case-control study was conducted on 112 POAG pati…

Epithelial-mesenchymal transition via transforming growth factor beta in pancreatic cancer is potentiated by the inflammatory glycoprotein leucine-rich alpha-2 glycoprotein.

We previously showed that an inflammation-related, molecule leucine-rich alpha-2 glycoprotein (LRG) enhances the transforming growth factor (TGF)-β1-induced phosphorylation of Smad proteins and is elevated in patients with pancreatic ductal adenocarcinoma (PDAC). As TGF-β/Smad signaling is considered to play a key role in epithelial-mesenchymal transition (EMT), we attempted to clarify the mechanism underlying LRG-related EMT in relation to metastasis in PDAC. We cultured LRG-overexpressing PDAC cells (Panc1/LRG) and evaluated the morphology, EMT-related molecules and TGF-β/Smad signaling path

The effect of ultraviolet radiation on the transforming growth factor beta 1/Smads pathway and p53 in actinic keratosis and normal skin.

Ultraviolet (UV) radiation is considered to be essential for the progression of actinic keratosis (AK) to squamous cell carcinoma (SCC); however, the mechanisms have not been fully elucidated. To understand this process, the effects of UV radiation on the transforming growth factor beta 1 (TGFβ1)/Smads pathway and p53 in normal skin and AK were studied. Normal human skin and AK tissues were cultured and divided into the following four groups according to the UV radiation dose: 0 (control group), 5, 10, and 20 J/cm2. The tissues were radiated for four consecutive days; 24 h after radiation, the…

Transforming growth factor-beta 1 (TGF-B1) liberation from its latent complex during embryo implantation and its regulation by estradiol in mouse.

Transforming growth factor-beta (TGF-B) plays an important role in embryo implantation; however, TGF-B requires liberation from its inactive latent forms (i.e., large latent TGF-B complex [LLC] and small latent TGF-B complex [SLC]) to its biologically active (i.e., monomer or dimer) forms in order to act on its receptors (TGF-BRs), which in turn activate SMAD2/3. Activation of TGF-B1 from its latent complexes in the uterus is not yet deciphered. We investigated uterine latent TGF-B1 complex and its biologically active form during implantation, decidualization, and delayed implantation. Our stu

Effect of transforming growth factor-beta receptor I kinase inhibitor 2,4-disubstituted pteridine (SD-208) in chronic allergic airway inflammation and remodeling.

Transforming growth factor (TGF)-beta is a multifunctional regulator of cell growth and differentiation with both pro- and anti-inflammatory properties. We used an inhibitor of TGF-beta receptor I (TGF-betaRI) kinase, SD-208 (2,4-disubstituted pteridine, a ATP-competitive inhibitor of TGF-betaRI kinase), to determine the role of TGF-beta in airway allergic inflammation and remodeling. Brown-Norway rats sensitized and repeatedly exposed to ovalbumin (OVA) aerosol challenge were orally administered SD-208 twice daily, before each of six OVA exposures to determine the preventive effects, or only

Altered expression of angiotensin II receptor subtypes and transforming growth factor-beta in the heart of nitrofen-induced diaphragmatic hernia in rats.

The renin-angitensin system (RAS) plays an important role as a growth factor in cardiac development. Angiotensin converting enzyme is involved in converting angiotensin I to angiotensin II (Ag-II). The effects of Ag-II are mediated by two primary receptors, type 1 (AT1) and type 2 (AT2). Ag-II stimulates transforming growth factor-beta1(TGF-beta1) and acts as a potent stimulant of myocyte growth and fetal contractile protein gene transcription. The aim of this study was to determine the expression of Ag-II receptor subtypes and TGF-beta1 in the hypoplastic heart of nitrofen-induced congenital

Transforming growth factor-beta-mediated apoptosis in the Ramos B-lymphoma cell line is accompanied by caspase activation and Bcl-XL downregulation.

Upon transforming growth factor-beta (TGF-beta) treatment, Ramos cells, a B-cell lymphoma cell line, undergo apoptosis, as measured by annexin V labeling, DNA fragmentation, and propidium iodide staining. Apoptosis could be observed by 24 h after TGF-beta exposure and occurred before the development of a significant blockage of cell cycle progression. TGF-beta-mediated apoptosis was also accompanied by a strong induction of caspase-3 subfamily activity. Incubation of cells with the caspase inhibitor Z-VAD.FMK at 20 microM, but not at 10 microM, prevented TGF-beta-induced apoptosis from occurri
