Prunella vulgaris Polysaccharide Ameliorates DSS-Induced Ulcerative Colitis by Increasing the Abundance of Ligilactobacillus agilis to Inhibit Inflammation and Preserve Intestinal Barrier Integrity.

Prunella vulgaris polysaccharide (PVP) possesses multiple bioactivities, yet its role in ulcerative colitis (UC) remains undefined. In this study, the effects of PVP were examined in mice with a DSS-induced colitis. In this colitis model, PVP treatment markedly reduced colonic inflammation and maintained the tight junction integrity. Meanwhile, PVP-enriched beneficial gut microbiota. Further research has revealed that PVP-modulated microbiota alone are sufficient to relieve UC symptoms. Mechanistically, PVP-enriched Ligilactobacillus agilis (Lig. agilis) subsequently enhanced tight junction pr

Prophage induction contributes to alterations in the gut phageome during intestinal inflammation.

Bacteriophages (phages) are abundant members of the gut microbiota and regulators of bacterial communities. During homeostasis, gut phage communities are longitudinally stable and lysogenic replication is dominant. In chronic gut inflammatory disorders, such as inflammatory bowel diseases (IBDs), there are alterations in phage diversity, which may result from changes in phage replication cycles. Here, we use a combination of in vitro, simplified community, and whole-community bioinformatics approaches to investigate whether prophage induction contributes to these alterations. We identify sever

Ganoderic Acid A alleviates ulcerative colitis by inhibiting the interleukin17 signaling pathway via targeting retinoic acid-related orphan receptor alpha.

ETHNOPHARMACOLOGICAL RELEVANCE: Ulcerative colitis (UC), a chronic inflammatory bowel disease, poses a significant global public health burden, a challenge compounded by the limited availability of effective treatments. Ganoderic acid A (GAA), which is a unique secondary metabolite in Ganoderma lucidum, has demonstrated anti-inflammatory, anti-tumor, antioxidant, and lipid-lowering activities. However, the role and mechanism of GAA in UC are not fully understood. AIM OF THE STUDY: In this study, we aimed to explore the role and involved mechanisms of GAA in UC. MATERIALS AND METHODS: We esta

Interpretation of the anti-colitis active ingredients and potential mechanisms of Wumei Wan based on spectrum-effect relationships, network pharmacology, molecular docking and experimental validation.

Wumei Wan (WMW), a renowned classic traditional Chinese medicine formula, is traditionally indicated for regulating cold-heat imbalance, exerting intestinal astringent effects, and arresting diarrhea. Clinically, it is widely employed in the treatment of ulcerative colitis (UC), which primarily manifests as chronic diarrhea and mucopurulent bloody stools. However, its material basis and pharmacological mechanisms involved remain obscure. This study employed spectrum-effect relationships, network pharmacology, molecular docking and experimental validation to identify the bioactive components of

Squid cartilage-derived chondroitin sulfate from Uroteuthis chinensis: a novel marine prebiotic modulating gut microbiota and ameliorating ulcerative colitis.

This study evaluated the prebiotic potential of chondroitin sulfate (CS) derived from the cartilage of the squid Uroteuthis chinensis, and its protective effects against dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice. Dietary intervention with squid-derived CS significantly attenuated characteristic UC symptoms, including body weight loss, colon shortening, and histopathological damage. Mechanistic analyses revealed that squid-derived CS exerted potent anti-inflammatory effects by downregulating key pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) and alleviated oxidative

Microbial metabolite Enterobactin impairs mitochondrial respiration and alleviates colitis.

BACKGROUND AND AIMS: Host-microbe interactions critically shape cellular metabolism and immune responses. The bacterial siderophore enterobactin (Ent) is known for pilfering iron from the host, but recent evidence suggests that it may also deliver iron to host mitochondria. Its impact on mitochondrial respiration, however, remains poorly understood. Here, we assess the interplay among Ent, iron, and lipocalin-2 (Lcn2, which sequesters Ent) on mitochondrial function. We also examine the effects of the 2, 3-dihydroxybenzoic acid (2, 3-DHBA), the monomeric derivative of Ent, and the mammalian sid

Glucose-derived ultra-small carbon nanozymes for treating inflammatory bowel disease.

Inflammatory bowel disease (IBD) encompasses a group of common inflammatory intestinal disorders that are currently challenging to cure. Nanozymes, as an emerging class of therapeutics, hold significant promise in treating IBD. In this study, an ultrasmall, metal-free carbon nanozyme (GNZ) was synthesized from glucose through a one-step hydrothermal method. GNZ exhibited quantitatively characterized superoxide dismutase (SOD)-mimicking catalytic activity and broad-spectrum antioxidant capacity against multiple reactive oxygen and nitrogen species. In LPS-stimulated macrophages, GNZ reduced int

Eleutheroside E alleviates ulcerative colitis by suppressing Th17 immune response via the MAPK signaling pathway and remodeling gut microbiota.

BACKGROUND: Ulcerative colitis (UC) is a long-term, recurrent inflammatory condition marked by bloody feces and mucosal damage. Its rising global incidence and socioeconomic burden necessitate safer long-term interventions. Edible and medicinal herbs (EMHs) are prospective alternatives due to their multi-target profiles and safety. Eleutheroside E (EE), a key component of the well-known EMH Eleutherococcus senticosus, exhibits robust inflammation-suppressing capacities. Nonetheless, its anti-inflammatory protective efficacy and associated mechanisms in acute colitis require further exploration

Postbiotic delivery via alginate encapsulation alleviates DSS-induced colitis by modulating gut microbiota and SCFA-mediated immune signaling.

Probiotic therapies for IBD are limited by low gastrointestinal viability and inconsistent efficacy. Postbiotics, non-viable microbes or their metabolites, offer a safer, more stable option, yet their distinct functions remain unclear, largely due to the lack of standardized delivery systems for direct comparison. In this study, we have developed a dual-delivery sodium alginate encapsulation system designed to facilitate the controlled gastrointestinal release of Lactiplantibacillus plantarum dy-1 in both its live (Sdy-1) and heat-inactivated (SHdy-1) forms. Employing colitis mouse model, we e
