[Familial myopathy with desmin storage seen as a granulo-filamentar, electron-dense material without mutation of the alphabeta-crystallin gene].

BACKGROUND: Desmin-related myopathy is a familial myopathy and cardiomyopathy. Three subgroups have been outlined, an autosomal dominant (AD) granulofilamentous type with cardiomyopathy, an AD cytoplasmic/spheroid inclusion body type and an autosomic recessive Mallory body-like inclusion type. Recently, in one family belonging to the first group it has been identified a mutation within a gene coding for a chaperone protein, alphabeta-crystallin (CRYAB gene). OBJECTIVE: To describe a Spanish family with a desmin-related myopathy, an AD granulofilamentous type with cardiomyopathy, with a molecu

Antigenic variation and strain heterogeneity in Borrelia spp.

Antigenic variation and strain heterogeneity have been demonstrated for the pathogenic Borrelia species, i.e. B. burgdorferi and the relapsing fever borreliae. In relapsing fever, new borrelia serotypes emerge at a high rate spontaneously, a mechanism that is caused by DNA rearrangements on linear plasmid translocating genes coding for variable major proteins from previous silent to expression sites (i.e. from inner sites to telomeric sites of the plasmid). As a result of this variation, the borreliae escape the immune response of the host, thus leading to the relapse phenomenon. In B. burgdor

Astrocyte-enriched and cilia-related genetic signatures in dementia: A whole-exome sequencing study.

BACKGROUND: Alzheimer's disease (AD), the most prevalent form of dementia, is highly heritable, yet much of its genetic basis remains unexplained. Most genetic studies have focused on European populations, with limited data from under-represented groups. Whole-exome sequencing (WES) in the Han Chinese population may help identify novel rare variants and population-specific genetic risk factors. AIMS: This study aims to use WES to identify potential candidate genes associated with dementia and AD in the Han Chinese population. METHODS: In this exploratory pilot study, we performed WES in 63 H

Artificial intelligence in forensic genetics: a multi-database bibliometric and science-mapping analysis (2003-2026).

PURPOSE: Artificial intelligence, machine learning and computational prediction are entering forensic genetic workflows, from probabilistic DNA-profile interpretation to the prediction of externally visible characteristics, biogeographical ancestry and age. This study maps the structure of that literature and interprets it in terms of translational maturity, to support research prioritisation and validation agendas. METHODS: Two complementary retrieval arms were executed between 10 and 13 September 2026: a venue-anchored search of 80 forensic and genetics journals derived from the Scopus Sour

Pathogenesis and neurotropism of a contemporary Powassan virus lineage II strain in mice.

Powassan virus (POWV) is an emerging tick-borne flavivirus that can cause neurologic disease in humans. POWV exhibits substantial genetic and phenotypic diversity, including marked variability in replication in vitro and pathogenesis in mice. However, most experimental studies have relied on historical lineage I and II strains to assess POWV pathogenesis. Here, we characterized a contemporary lineage II strain, NY.19.12, in mice and investigated the potential viral determinants of disease. NY.19.12 caused earlier onset of clinical signs and earlier detection of viral RNA (vRNA) in the spleen a

Significant variance in genetic diversity among populations of Schistosoma haematobium detected using microsatellite DNA loci from a genome-wide database.

BACKGROUND: Urogenital schistosomiasis caused by Schistosoma haematobium is widely distributed across Africa and is increasingly being targeted for control. Genome sequences and population genetic parameters can give insight into the potential for population- or species-level drug resistance. Microsatellite DNA loci are genetic markers in wide use by Schistosoma researchers, but there are few primers available for S. haematobium. METHODS: We sequenced 1,058,114 random DNA fragments from clonal cercariae collected from a snail infected with a single Schistosoma haematobium miracidium. We assem

MtDNA control region variation affirms diversity and deep sub-structure in populations from southern Africa.

BACKGROUND: The current San and Khoe populations are remnant groups of a much larger and widely dispersed population of hunter-gatherers and pastoralists, who had exclusive occupation of southern Africa before the influx of Bantu-speakers from 2 ka (ka = kilo annum [thousand years] old/ago) and sea-borne immigrants within the last 350 years. Here we use mitochondrial DNA (mtDNA) to examine the population structure of various San and Khoe groups, including seven different Khoe-San groups (Ju/'hoansi, !Xun, /Gui+//Gana, Khwe, ≠Khomani, Nama and Karretjie People), three different Coloured groups

Animal models of copper-associated liver disease.

Recent advances in molecular biology have made possible the identification of genetic defects responsible for Wilson's disease, Indian childhood cirrhosis and copper toxicosis in Long Evans Cinnamon rats, toxic milk mice, and Bedlington terriers. The Wilson's disease gene is localized on human chromosome 13 and codes for ATP7B, a copper transporting P-type ATPase. A genetic defect similar to that of Wilson's disease occurs in Long Evans Cinnamon rats and toxic milk mice. Familial copper storage disorders in Bedlington and West Highland white terriers are associated with early subclinical disea

Non-Canonical Function of ORC1: Regulating Notch-VEGFR2-Mediated Angiogenesis in Melanoma Progression.

Melanoma is an aggressive skin cancer characterized by high metastatic potential and substantial genetic heterogeneity. Although genome-wide association studies (GWAS) have identified numerous susceptibility loci, the functional roles of risk variants, most of which reside in non-coding regions, remain poorly understood. This study aimed to identify genetic susceptibility genes involved in melanoma progression and investigate their non-canonical roles in the tumor microenvironment. GWAS summary statistics from the GWAS Catalog, FinnGen, and UK Biobank were integrated using MAGMA and FUSION to

Adenosine-to-Inosine (A-to-I) RNA Editing by ADAR1 to Control RNA Sensing in Cardiovascular Disease.

Across biology, organisms have retained a mechanism to diversify the RNA transcriptome through RNA editing. Mediated by ADAR (adenosine deaminase acting on RNA) enzymes, Adenosines in double-stranded RNA (dsRNA) structures can be edited to Inosines (adenosine-to-inosine edit). Although this can change the amino acid sequence if it occurs in a coding sequence of mRNA, the majority of RNA editing in mammalian cells is found in noncoding repetitive elements. These repetitive elements have a predisposition to form long dsRNA structures that can mimic a dsRNA virus. Since the initial discoveries of

Pangenomes aid accurate detection of large insertions and deletions from targeted sequencing: the case of cardiomyopathies.

BACKGROUND: Gene panels represent a widely used strategy for genetic testing in a vast range of Mendelian disorders. While this approach aids reliable bioinformatic detection of short coding variants, it often fails to detect many larger variants. Recent studies have recommended the adoption of pangenome references (as opposed to linear reference genomes like GRCh38) to augment detection of large variants from targeted sequencing, potentially providing diagnostic laboratories with the possibility to streamline diagnostic work-ups and reduce costs. METHODS: Here, we analyze 1969 cardiomyopathy

The MEG3-miRNA regulatory axis could potentially modulate response to biological therapies in psoriatic arthritis.

BACKGROUND: The long non-coding RNA - microRNA (lncRNA-miRNA) regulatory axis is a key modulator of immune and inflammatory pathways, and growing evidence supports its contribution to therapeutic response variability. In our previous study, we identified the lncRNA MEG3 as potentially involved in the response to biological drugs, specifically TNFα- and IL17A-inhibitors, in psoriatic arthritis (PsA). METHODS: To further characterize its role, we performed a bioinformatic analysis to identify MEG3-targeted miRNAs, followed by an exploratory expression profiling analysis using qRT-PCR in a cohor

Vestibular schwannoma: genetic and epigenetic mechanisms, hearing loss, and emerging therapies.

BACKGROUND: Although most vestibular schwannomas (VS) occur sporadically, both sporadic and hereditary tumors share common molecular features beyond the loss of NF2. New evidence highlights the role of interconnected signaling pathways and epigenetic regulation in Schwann cell tumorigenesis, pointing toward potential molecularly targeted therapeutic strategies. METHODS: This review synthesizes preclinical, molecular, and clinical evidence to examine genetic and epigenetic mechanisms underlying VS, therapeutic strategies, and contributors to hearing loss. A structured search of ClinicalTrials.

A Novel Mutation in ACTG1 as the Probable Cause of Nonsyndromic Hearing Loss in Chinese Han Population.

BACKGROUND: Hearing loss is the most common sensory nervous system defect in humans. Approximately half of hearing loss cases have a genetic etiology. At present, more than 300 genes and 1000 mutations have been identified that cause hereditary hearing loss (HHL). However, there are still a large number of unknown genes related to hearing loss. OBJECTIVE: The purpose of this study was to analyze the clinical audiological characteristics of a family with hereditary nonsyndromic hearing loss (NSHL) and identify the mutation of deafness-related genes. MATERIAL AND METHODS: A Chinese Han-dominan

Endocrine features and hormone-related pathways in Wilms' tumor: From developmental biology to clinical implications.

Wilms' tumor (WT) is the most common primary renal malignancy in children and it is a paradigmatic developmental kidney tumor arising from disrupted nephrogenesis. Classically framed as a developmental disorder of the nephron progenitor compartment, WT also exhibits clinically relevant endocrine features, most notably renin-mediated hypertension and, less commonly, selected paraneoplastic syndromes including Cushing syndrome, erythrocytosis, hypercalcemia, and acquired von Willebrand disease. Biochemically, atypical elevations of α-fetoprotein (AFP) and β-human chorionic gonadotropin (β-HCG) m

The Role of MicroRNA in Diagnosis of Huntington's Disease: A Systematic Review.

Huntington's disease (HD) is an autosomal dominant genetic neurodegenerative disorder with features of progressive motor, cognitive, and psychiatric dysfunction. Current diagnosis relies largely on clinical presentation and genetic testing but lacks sensitivity to early disease diagnosis or progression monitoring. MicroRNAs (miRNAs) are short non-coding RNA molecules that regulate gene expression post-transcriptionally. They are increasingly being investigated as potential diagnostic biomarkers, especially given their detectability in biofluids like plasma and cerebrospinal fluid. This review

Distinct mutational landscapes for germline and somatic cancer variants in forty tumor suppressor genes.

Germline and somatic cancer variants in tumor suppressor genes (TSGs) share loss-of-function mechanisms, but studies of a few genes (DICER1 and CEBPA) have demonstrated differences in variant consequence and location. To systematically assess whether TSGs display distinct mutational patterns, we leveraged large public genetic databases and compared 32,941 high-quality pathogenic/likely pathogenic (P/LP) germline variants in ClinVar, with 12,907 oncogenic/likely oncogenic (O/LO) somatic tumor variants from cBioPortal across 40 TSGs. Only 3,863 (9.2%) variants were shared. Eighteen TSGs showed s
