Recurrence of Treacher Collins' syndrome with sonographic findings.

Treacher Collins' syndrome, or mandibulofacial dysostosis, is a rare, autosomal-dominant disorder. We describe the recurrence of this anomaly in the fetus of a patient who previously had delivered a severely affected infant. The ultrasonographic findings of hydramnios, absence of fetal swallowing movements, and poor growth of the biparietal diameter and head circumference permitted us to suggest the diagnosis antenatally of another affected child.

Gynecomastia. A bothersome but readily treatable problem.

Although breast enlargement in boys and men can cause both psychological and physical distress, the disorder is rarely serious and is readily treatable. Several factors can lead to the estrogenic excess that causes growth of breast tissue. Dr Jacobs describes a patient with gynecomastia related to cirrhosis of the liver who responded promptly to a brief course of tamoxifen citrate therapy.

[Post-traumatic hip dysplasia].

At a traffic accident in 1970 a then 6 year old boy got an impacted fracture of the neck of the right femur with persisting subluxation. This was not particularly noticed because of the severity of other injuries. In the meantime the most recent follow-up examination (1981) of the boy (now 17 years old) showed a dysplasia like growth disorder. Various possibilities of interpretation are discussed.

Atypical myeloproliferative disorder associated with a 21 trisomic clone.

The hematological and cytogenetical studies of a case of atypical myeloproliferative disorder are reported. The patient displayed moderate leukocytosis, anemia and thrombocytopenia. Reduced or absent growth of CFU-C was observed in the bone marrow and peripheral blood. The cytogenetical analysis revealed a trisomy 21 limited to the neoplastic clone. The overall results are discussed and compared with the rare cases of acquired trisomy 21 reported in the literature.

Epigenetics Beyond Fetal Growth Restriction: A Comprehensive Overview.

Fetal growth restriction is a pathological condition occurring when the fetus does not reach the genetically determined growth potential. The etiology of fetal growth restriction is expected to be multifactorial and include fetal, maternal, and placental factors, the latter being the most frequent cause of isolated fetal growth restriction. Severe fetal growth restriction has been related to both an increased risk of perinatal morbidity and mortality, and also a greater susceptibility to developing diseases (especially cardio-metabolic and neurological disorders) later in life. In the last dec

Predicting growth and curve progression in the individual patient with adolescent idiopathic scoliosis: design of a prospective longitudinal cohort study.

BACKGROUND: Scoliosis is present in 3-5% of the children in the adolescent age group, with a higher incidence in females. Treatment of adolescent idiopathic scoliosis is mainly dependent on the progression of the scoliotic curve. There is a close relationship between curve progression and rapid (spinal) growth of the patient during puberty. However, until present time no conclusive method was found for predicting the timing and magnitude of the pubertal growth spurt in total body height, or the curve progression of the idiopathic scoliosis. The goal of this study is to determine the predictive

Early-Life Presentation and Phenotypic Variability of the IGF1R c.3595G>A (p.Gly1199Arg) Variant: A Case Report.

Most infants born small for gestational age (SGA) achieve spontaneous catch-up growth within the first two years of life; however, a subset remains growth-restricted and warrants further evaluation. Persistent short stature accompanied by microcephaly should prompt consideration of an underlying monogenic disorder. We report a term male infant born with severe SGA (birth weight -3.8 SDS; birth length -2.2 SDS) and microcephaly. At 16 months of age, his height was -2.7 SDS and head circumference -3.1 SDS. Initial insulin-like growth factor 1 (IGF-1) concentration was mildly decreased (-1.0 SDS)

Explaining the Association Between Fetal Growth and Childhood ADHD Symptoms: Cross-cohort Replication.

The association between restricted fetal growth and symptoms of attention deficit/hyperactivity disorder (ADHD) in childhood is well-replicated and robust. However, fetal growth is determined by many prenatal factors and associations with mental health may be confounded by familial and social context. In this study, we sought to quantify the relative contributions of prenatal factors and familial confounds to the association between fetal growth and ADHD symptoms. Two independent cohorts were analyzed, the Adolescent Brain Cognitive Development study (ABCD; United States) and the Growing Up in

Understanding Hippocampal Development in Young Children With Autism Spectrum Disorder.

OBJECTIVE: We examined growth trajectories of hippocampal volume (HV) in early childhood in a longitudinal cohort of male and female participants with autism spectrum disorder (ASD) and typically developing (TD) individuals, and investigated HV in those with large brains. Relations between factors potentially associated with hippocampal size and growth were investigated. METHOD: Participants received 1 to 3 structural magnetic resonance imaging scans between ages 25 and 80 months (unique participants: ASD, n =200; TD, n =110; total longitudinal scans, n = 593). HV growth during this period wa

The minor spliceosome component U4atac regulates JAK/STAT signaling to modulate hematopoiesis and immune responses in Drosophila melanogaster.

The small nuclear RNA U4atac is a core component of the minor spliceosome. In humans, homozygous or compound heterozygous point mutations in U4atac cause rare developmental disorders, such as Roifman syndrome, characterized by growth restriction, brain anomalies, and immune deficiency. To better define the pathophysiological role of U4atac mutations, we here establish a model of minor spliceosome dysfunction by generating a Drosophila melanogaster CRISPR/Cas9-induced U4atac mutant in the highly conserved stem II region. U4atac homozygous mutants exhibit growth and neurodevelopmental defects, i

Integrated Liquid Chromatography-Tandem Mass Spectrometry and Headspace Solid-Phase Microextraction With Gas Chromatography-Mass Spectrometry-based Profiling Reveals Developmental Metabolic Dynamics of Ligusticum striatum DC.

Ligusticum striatum DC. (Ligusticum chuanxiong Hort. or Ligusticum wallichii) is a traditional medicinal herb in East Asia used for cardiovascular, neurological, and inflammatory disorders, but developmental phytochemical changes remain unclear. Here, targeted liquid chromatography-tandem mass spectrometry (LC-MS/MS) and untargeted headspace solid-phase microextraction with gas chromatography-MS were applied to examine five bioactive compounds and volatile organic compounds (VOCs) in aerial and root tissues across seven growth periods. Five compounds, including ferulic acid, icariside F2, ligu

MATR3 is essential for oocyte growth and maturation quality through a dual molecular mechanism.

The molecular mechanisms governing mRNA accumulation during oocyte growth, essential for developmental competence, remain poorly understood. This study investigates the role of Matrin-3 (MATR3), a highly expressed RNA-binding protein in growing oocytes (GOs), using oocyte-specific knockout mouse models and human oocyte maturation arrest (OMA) samples. The results showed that MATR3 was more abundant in GOs than fully grown oocytes (FGOs), highly expressed in the nucleus of non-surrounded nucleolus (NSN) oocytes, and exited the nucleus during the NSN-to-surrounded nucleolus (SN) transition. In O

Sleep matters: rethinking its role in growth hormone deficiency treatment.

PURPOSE: This letter has the purpose of alerting the medical community to the impact of sleep debt on growth hormone (GH) therapy, mostly applied to pediatric and puberty ages. METHODS: We discussed the role of sleep in GH release, especially N3 sleep. RESULTS: During sleep, hypothalamus is stimulated to secrete growth hormone–releasing hormone (GHRH) which activates synthesis and secretion of GH by anterior pituitary gland. Studies focusing on the interaction of sleep and GH release are scarce and have controversial results. Some studies have found a direct effect of N3 sleep in GH pulse, ind

Open, Hybrid and Endovascular Management of Aortic Arch Aneurysms: Recent Updates and Future Directions.

Once considered a surgical frontier fraught with risk, aortic arch aneurysms now represent a domain of evolving innovation. Despite their rarity, they pose severe risks of dissection, rupture, and mortality if not adequately managed. Primarily caused by prior aortic dissections, atherosclerosis, or connective tissue disorders, these aneurysms are often found incidentally on CTA or MRA imaging. Medical management focuses on reducing aortic wall stress through blood pressure control, risk factor modification, and regular imaging to monitor growth. Surgical intervention is typically indicated whe

Tyrosine Supplementation Rescues a Growth Defect in a Humanized S. cerevisiae Model of YARS1 Associated with CMT-DI.

Dominant pathogenic mutations in tyrosyl-tRNA synthetase (YARS1) are associated with Charcot-Marie-Tooth disease (CMT), a progressive peripheral neuropathy for which no disease-modifying therapies currently exist. While recent advances in amino acid supplementation therapies suggest potential benefit for recessive aminoacyl-tRNA synthetase disorders, their applicability to dominant YARS1-associated neuropathies remains unclear. Here, we investigated the pathogenic mechanisms underlying the dominant YARS1 variants G41R, D81I, and E196Q. Using biochemical and functional analyses, we identified i

Transient receptor potential channels in hair follicle biology: physiological significance and translational relevance.

Transient receptor potential (TRP) channels are a family of versatile ion channels that act as sensor for a wide range of chemical and physical stimuli. These cation-permeable channels are widely expressed in various tissues and play pivotal roles in numerous biologic processes. Studies have demonstrated that six specific TRP channels -TRPV1, TRPV3, TRPV4, TRPM5, TRPML1 and TRPML3 - are actively involved in hair follicle (HF) cell proliferation, modulation of hair growth-related signalling pathways, regulation of hair cycle progression, and control of hair growth and pigmentation in humans and

Rickets.

Rickets remains one of the most common nontransmissible pediatric diseases, especially in low- to middle-income countries. Rickets is characterized by impaired mineralization of growing bone resulting in bone frailty, deformities, impaired growth and development, and pain with standing or walking. Nutritional rickets is the most common cause of bone disease in the world. The most frequent etiology of rickets is vitamin D deficiency, with pathognomonic biochemical and radiologic characteristics. However, even in the context of appropriate vitamin D levels, other causes for rickets need to be co

A Potent Neutralizing Monoclonal Antibody to Human Growth Hormone Suppresses Insulin-Like Growth Factor-1 in Female Rats.

Acromegaly and gigantism are disorders caused by hypersecretion of growth hormone (GH), usually from pituitary adenomas. Although somatostatin analogues (SSA), dopamine agonists, and GH receptor antagonists are important therapeutic agents, all of these have issues with their effectiveness, safety, and/or convenience of use. To overcome these, we developed a GH-specific potent neutralizing a mouse monoclonal antibody (mAb) named 13H02. 13H02 selectively bound both to human and monkey GH with high affinity, and strongly inhibited the biological activity of GH in the Nb2 rat lymphoma cell prolif
