Larval habitats of Anopheles species in a rural settlement on the malaria frontier of southwest Amazon, Brazil.

Rural settlements are social arrangements expanding in the Amazon region, which generate disturbances in the natural environment, thus affecting the ecology of the species of Anopheles and thus the malaria transmission. Larval habitats are important sources for maintenance of mosquito vector populations, and holding back a natural watercourse is a usual process in the establishment of rural settlements, since the formation of micro-dams represents a water resource for the new settlers. Identifying characteristics of the larval habitats that may be associated with both the presence and abundanc

MAVS Expressed by Hematopoietic Cells Is Critical for Control of West Nile Virus Infection and Pathogenesis.

UNLABELLED: West Nile virus (WNV) is the most important cause of epidemic encephalitis in North America. Innate immune responses, which are critical for control of WNV infection, are initiated by signaling through pathogen recognition receptors, RIG-I and MDA5, and their downstream adaptor molecule, MAVS. Here, we show that a deficiency of MAVS in hematopoietic cells resulted in increased mortality and delayed WNV clearance from the brain. In Mavs(-/-) mice, a dysregulated immune response was detected, characterized by a massive influx of macrophages and virus-specific T cells into the infecte

In silico analysis of MHC-I restricted epitopes of Chikungunya virus proteins: Implication in understanding anti-CHIKV CD8(+) T cell response and advancement of epitope based immunotherapy for CHIKV infection.

Chikungunya virus (CHIKV) is a mosquito-borne Alphavirus, responsible for acute febrile infection. The high morbidity and socio-economic loss associated with the recent CHIKV epidemics worldwide have raised a great public health concern and emphasize the need to study the immunological basis of CHIKV infection to control the disease. MHC-I restricted CD8(+) T cell response represent one of the major anti-viral immune responses. Accordingly, it is essential to have a detailed understanding towards CHIKV specific MHC-I restricted immunogenic epitopes for anti-viral CD8(+) CTL immunogenicity. In

Prevalence and seasonal variations of canine dirofilariosis in Portugal.

Dirofilariosis is a severe vector-borne emergent disease that is spreading worldwide and becoming a serious threat to human and veterinary public health. Portugal, a Mediterranean country, has favorable climate conditions for mosquito development and survival. At present, accurate data on the prevalence and epidemiological pattern of dirofilariosis in Portugal is scarce and outdated. To study these trends, a project was developed to assess the current prevalence of Dirofilaria immitis infection in shelter dogs as well as the prevalence of Dirofilaria species present in three coastal regions in

Survival of diurnally sub periodic Wuchereria bancrofti in Downsiomyia nivea (Diptera: Culicidae): a density dependent factor from Andaman & Nicobar Islands.

BACKGROUND & OBJECTIVES: In India, diurnally sub periodic Wuchereria bancrofti transmitted by Downsiomyia nivea is prevalent only in the Nicobar district of Andaman and Nicobar Islands. The ongoing LF elimination programme aims at transmission interruption by bringing down the microfilarie (mf) load in the community, which has implication on the parasite load in mosquito vector. Therefore, understanding density dependent constraints on transmission assumes significance from control perspective. The present study was undertaken in Teressa Island to understand the density dependent parasite mort

Challenges for the formulation of a universal vaccine against dengue.

Dengue is rapidly becoming a disease of an escalating global public health concern. The disease is a vector-borne disease, transmitted by the bite of an Aedes spp. mosquito. Dynamic clinical manifestations, ranging from asymptomatic, flu-like febrile illness, dengue fever (DF) to dengue hemorrhagic fever (DHF) with or without dengue shock syndrome (DSS), make the disease one of the most challenging to diagnose and treat. DF is a self-limited illness, while DHF/DSS, characterized by plasma leakage resulting from an increased vascular permeability, can have severe consequences, including death.

Life table characteristics of Aedes aegypti (Diptera:Culicidae) from Saudi Arabia.

Aedes aegypti (= Stegomyia aegypti) mosquito is a world vector of important arboviral diseases like dengue and Rift Valley fever. Despite its wide distribution in the western and southern regions of Saudi Arabia, where dengue outbreaks have occurred, its ecology is largely unknown. In this study we report on the main life table developmental attributes of a laboratory colony of Ae. aegypti reared from field-collected larvae from Madinah Province, west of Saudi Arabia. Females were maintained on daily blood meal and sugar. The female fecundity was ~62 eggs/female at an overall rate of 72% hatch

Travel-associated illness in older adults (>60 y).

BACKGROUND: Older individuals represent a substantial proportion of international travelers. Because of physiological changes and the increased probability of underlying medical conditions, older travelers might be at higher risk for at least some travel-associated diseases. METHODS: With the aim of describing the epidemiology of travel-associated diseases in older adults, medical data were prospectively collected on ill international travelers presenting to GeoSentinel sites from 1997 to 2009. Seven thousand thirty-four patients aged 60 years and over were identified as older travelers and w

The impact of human reservoir of malaria at a community-level on individual malaria occurrence in a low malaria transmission setting along the Thai-Myanmar border.

BACKGROUND: The probability of contracting malaria in a given individual is determined not only by the individual's characteristics, but also the ecological factors that characterize the level of human-vector contact in the population. Examination of the relationship between "individual" and "supra-individual" variables over time is important for understanding the local malaria epidemiology. This is essential for planning effective intervention strategies specifically for each location. METHODS: A retrospective cohort study was conducted, which followed a community-cohort of about 3,500 resid

Characterization of Rift Valley fever virus transcriptional terminations.

Rift Valley fever virus (RVFV) (genus Phlebovirus, family Bunyaviridae) has a tripartite negative-strand genome and causes a mosquito-borne disease among humans and livestock in sub-Saharan African and Arabian Peninsula countries. Phlebovirus L, M, and N mRNAs are synthesized from the virus-sense RNA segments, while NSs mRNA is transcribed from the anti-virus-sense S segment. The present study determined the 3' termini of all RVFV mRNAs. The 3' termini of N and NSs mRNAs were mapped within the S-segment intergenic region and were complementary to each other by 30 to 60 nucleotides. The termini

Antagonism of PRDX1 and Japanese encephalitis virus via the autophagy pathway.

Japanese encephalitis virus (JEV) is a mosquito-borne zoonotic infectious agent and causes animal and human illness of the central nervous system. Japanese encephalitis virus inflicts severe harm, yet even with an approved vaccine, prevention and control remain daunting, and the molecular mechanism underlying the virus-host interplay remains incompletely understood. In this study, we found that the host peroxiredoxin 1 (PRDX1) protein inhibits JEV replication. PRDX1 targets nonstructural protein 1 (NS1) of JEV and mediates its degradation via the proteasome and autophagy pathways, thus inhibit

In-silico identification of host-key-genes associated with dengue-virus-infections highlighting their pathogenetic mechanisms and therapeutic agents.

Dengue fever (DF), a potentially fatal mosquito-transmitted viral disease caused by dengue virus (DENV) infections (DENVI), stands as the predominant arthropod-borne viral illness worldwide, presenting a significant global health challenge. DENV-mediated proteins/proteases interact with host proteins to develop the infection. Despite the severity of DENVI, the infection-causing host key-genes (hKGs), their pathogenetic processes, and inhibitors/activators are not yet rigorously investigated. This study aimed to disclose DENVI-causing hKGs, highlighting their pathogenetic mechanisms and therape

Local drivers of Rift Valley fever outbreaks in Mauritania: A one health approach combining ecological, vector, host and livestock movement data.

Rift Valley fever (RVF) is a vector-borne zoonotic disease with recurrent epidemic and epizootic outbreaks in Mauritania caused by the RVF virus (RVFV). In recent years, outbreaks have occurred with increasingly shorter inter-epidemic periods. The primary objective of this study was to utilise a high-resolution spatiotemporal model and identify the drivers and ecological suitability for RVFV infections, as well as areas for RVF outbreaks and emergence in humans and animals, respectively, in Mauritania. We used geolocated data from 2019 to 2023 for modelling, including human RVF cases confirmed

Next-generation sequencing survey of acute febrile illness in Senegal (2020-2022).

INTRODUCTION: Acute febrile illnesses (AFI) in developing tropical and sub-tropical nations are challenging to diagnose due to the numerous causes and non-specific symptoms. The proliferation of rapid diagnostic testing and successful control campaigns against malaria have revealed that non-Plasmodium pathogens still contribute significantly to AFI burden. Thus, a more complete understanding of local trends and potential causes is important for selecting the correct treatment course, which in turn will reduce morbidity and mortality. Next-generation sequencing (NGS) in a laboratory setting can

Microscopic prevalence and risk factors of asymptomatic malaria in Gorgora, western Dembia, Northwest Ethiopia: exploring hidden threats during minor transmission season.

BACKGROUND: Malaria poses a significant public health threat globally, particularly in African regions, where asymptomatic malaria is a considerable logistic problem. Individuals with asymptomatic malaria do not seek treatment, and thus they are invisible to health facilities and represent a substantial hidden reservoir of Plasmodium species. This study aimed to determine the prevalence of asymptomatic malaria and its associated factors in Gorgora, western Dembia district, Northwest Ethiopia. METHODS: A community-based cross-sectional study was conducted from May to June 2023 in the Gorgora a

Detection of Dirofilaria immitis in golden jackals (Canis aureus L.) but not in red foxes (Vulpes vulpes L.) and European badgers (Meles meles L.) in Croatia.

BACKGROUND: Dirofilariosis is a parasitic mosquito-borne disease caused by members of the genus Dirofilaria, which includes Dirofilaria immitis and Dirofilaria repens. Surveillance studies in Europe have revealed that D. immitis can also be detected in a range of wild carnivores, raising questions over the impact of infections on wild carnivore animal health but also whether these populations may act as a reservoir for infection of other species, including domestic dogs and humans. METHODS: In the current study, we conducted surveillance for the presence of D. immitis in several wild carnivor

Seroepidemiological Studies on Japanese Encephalitis: A Systematic Review.

OBJECTIVES: Japanese encephalitis (JE) is one of the major mosquito-borne infectious diseases in the Western Pacific region, accounting for 20%-30% of mortality cases. The JE virus (JEV) seroprevalence fluctuations indicate that continuous research is important for prevention and control activities. By mapping JEV seroprevalence by age stratification, the population profile for immunity and susceptibility can be identified to aid in vaccination program planning. Thus, this study aimed to determine the trend of age-specific JEV seroprevalence. METHODS: We conducted a systematic review of all s
