Developing and validating a neonatal screening tool for congenital anomalies to be used in low- and middle-income country settings.

BACKGROUND: Congenital anomalies are among the common health problems faced by children in low- and middle-income countries, contributing substantially to infant mortality rates. Due to limited access to newborn screening programmes in most of the resource-limited settings, many congenital anomalies go undiagnosed and untreated, leading to adverse outcomes. This study aimed to develop and validate a newborn screening tool for congenital anomalies for use in resource-limited settings. METHODS: A Delphi approach was used to assemble a group of experts and develop the screening tool. Tool valida

Damage Accrual in Patients with Systemic Lupus Erythematosus Predicts Mortality and Is Associated Primarily with Antiphospholipid Syndrome and Hypertension.

Background/Objectives: Long-term outcomes in systemic lupus erythematosus (SLE) are largely driven by irreversible organ damage, yet the relative contribution of comorbid conditions remains insufficiently characterized. We aimed to characterize damage accrual and identify comorbidities associated with damage severity and mortality. Methods: A retrospective study of adult patients with SLE followed at a single-center (2014-2023). The Systemic Lupus International Collaborating Clinics/American College of Rheumatology Damage Index (SDI), was used to assess damage at last follow-up. Damage was cat

Hybrid movement strategies in orthopedic oncology: Mechanistic integration of musculoskeletal and tumor-host interactions.

Exercise is widely recommended in oncology; however, its role in orthopedic oncology remains insufficiently defined within a clinical context where tumor biology, skeletal fragility, muscle loss, systemic therapy, and reconstructive biomechanics converge. Existing rehabilitation frameworks primarily emphasize functional recovery, while the biological integration between mechanical loading and tumor-host systems remains incompletely characterized in clinical decision-making contexts. This narrative review synthesizes mechanistic evidence across tumor mechanobiology and its integration with bone

Autonomic Dysfunction as a Potential Pathophysiological Link Between Chronic Low Back Pain and Coronary Artery Disease: A Systematic Review.

Chronic low back pain (CLBP) is a leading cause of disability worldwide and is increasingly recognized as a condition associated with systemic physiological alterations beyond musculoskeletal dysfunction. Emerging evidence suggests that autonomic nervous system dysregulation may be associated with impaired cardiovascular regulation and increased cardiovascular risk in individuals with CLBP. This systematic review aimed to evaluate the current evidence regarding autonomic dysfunction in CLBP and its potential association with cardiovascular disease. A comprehensive literature search was conduct

Expanding the genotypic and phenotypic spectrum of PGAP1 deficiency: clinical and functional insights from 15 patients.

Glycosylphosphatidylinositol-anchored proteins (GPI-APs) are essential for neuronal development, synaptic organization and signaling. Defects in GPI-anchor biosynthesis or remodeling cause rare neurodevelopmental disorders, including post-GPI attachment to proteins 1 (PGAP1) deficiency. PGAP1 encodes an inositol deacylase required for GPI-anchor remodeling and appropriate trafficking and membrane localization of GPI-APs. Loss of PGAP1 function disrupts GPI-AP processing, but the clinical spectrum remains incompletely defined because reported cohorts are small. We report 15 individuals with bia

Demographics and clinical manifestations in patients with Behçet's disease: a retrospective observational study from a non-endemic country.

Behçet's disease (BD) is a systemic vasculitis of unknown origin affecting both arterial and venous vessels, resulting in diverse clinical features. Its manifestations vary due to ethnic, geographic, and individual differences. Although the highest prevalence of BD is reported along the ancient Silk Road route, nowadays, due to population migrations, the disease can be detected worldwide. Our study aimed to evaluate the demographic and clinical characteristics of patients with BD in a cohort from a non-endemic country and compare it with other cohorts from endemic and non-endemic countries wor

Sexual Dimorphism in Systemic Inflammatory Responses to Femur Fracture in Mice Infected With SARS-CoV-2-Like Virus.

Patients with femur fractures who are concurrently infected with COVID-19 face a threefold increase in mortality, likely due to a compounded inflammatory response. Furthermore, sex-specific differences in immune responses to COVID-19 are well documented, implicating biological sex as a factor that can modulate disease severity in these comorbid conditions. Understanding the inflammatory interplay underlying this association is critical for the development of effective, targeted therapies to mitigate mortality. In this study, we investigated the systemic, sex-specific inflammatory response in m

A case series of neuro-chikungunya: unmasking the neurotropic potential of chikungunya virus.

BACKGROUND: Chikungunya is a mosquito-borne viral infection, which classically presents with the triad of acute onset of fever, rash, and debilitating arthralgia. Although musculoskeletal symptoms are predominant, it is increasingly recognized that chikungunya virus infection can present with neurological complications involving both the central and peripheral nervous systems via mechanisms that include direct viral invasion and immune-mediated responses. CASE PRESENTATION: In this report, we describe five adult patients with confirmed chikungunya infection who presented with a wide spectrum

Personalized rehabilitation approach for reaching movement using reinforcement learning.

Musculoskeletal disorders challenge significantly the performance of many daily life activities, thus impacting the quality of life. The efficiency of the traditional physical therapy programs is limited by ecological parameters such as intervention duration and frequency, number of caregivers, geographic accessibility, as well as by subjective factors such as patient's motivation and perseverance in training. The implementation of VR rehabilitation systems may address these limitations, but the technology still needs to be improved and clinically validated. Furthermore, current applications g
