Emergence and endemicity of arboviruses in Haiti: a review.

OBJECTIVES: The increase in geographical spread of arboviruses and the incidence of arboviral diseases of humans is a rising public health concern, particularly in the context of ongoing population movements, climate change, and challenges to public health surveillance systems in countries where many of these viruses are endemic. We present here a scoping review and a synopsis of the existing literature on arboviruses that have been reported in Haiti, identify existing challenges in public health surveillance, and discuss the implications for Haiti, the Caribbean, and the Americas. METHODS: A

Multiexon COL1A2 deletion as a rare mechanism in osteogenesis imperfecta: Case report and literature review.

BACKGROUND: Osteogenesis imperfecta (OI) is mostly caused by pathogenic variants in COL1A1/COL1A2; while single nucleotide variants predominate, multiexon copy number variants (CNVs) remain under-recognised contributors with incompletely defined phenotypic impacts. METHODS: We investigated a fetus presenting severe skeletal dysplasia using NGS with CNV calling and MLPA, clinical and radiologic assessments, and transcript analysis of maternal fibroblasts. We also reviewed the literature and curated databases for reported multiexon COL1A2 deletions. RESULTS: The fetus exhibited lethal OI type

Co-segregation of WFS1 p.Gly494Ser variant with recurrent vasovagal syncope in a multigenerational family: a candidate susceptibility variant needing independent validation.

INTRODUCTION: Vasovagal syncope (VVS) is the most prevalent subtype of reflex syncope. However, the genetic etiology underlying familial recurrent VVS has not been fully elucidated. METHODS: We recruited a multigenerational Chinese pedigree with three-generation recurrent early-onset VVS. Whole-exome sequencing was performed to screen pathogenic variants. Variant interpretation was conducted following the ACMG-AMP 2015 criteria, and multiple in-silico prediction tools were applied to evaluate variant pathogenicity. Clinical laboratory indicators of affected family members were collected and a

Causal effects of type 2 diabetes, obesity, gout, and hypothyroidism on carpal tunnel syndrome: a univariable and multivariable Mendelian randomization study.

BACKGROUND: Carpal tunnel syndrome (CTS) is a prevalent peripheral neuropathy with unclear pathogenesis. This study investigated the causal relationships between metabolic diseases and CTS using univariable and multivariable Mendelian randomization (MR) analyses. METHODS: Single nucleotide polymorphisms from large genetic databases served as instrumental variables. Genome-wide data on type 2 diabetes (T2D), obesity, gout, hypothyroidism, and CTS were obtained from the IEU OpenGWAS Project (Integrative Epidemiology Unit Open Genome-Wide Association Studies Project). Univariable MR analysis ass

CACNA1A Genetic Variants and Their Potential Involvement in Migraine Pathogenesis.

Migraine is a prevalent neurological disorder that affects over 1 billion individuals worldwide. The pathogenesis of migraine remains incompletely understood, though evidence suggests a multifactorial etiology involving genetic factors. The CACNA1A gene has been implicated in rare forms of Familial Hemiplegic Migraine (FHM). This study aimed to investigate the role of CACNA1A variants in individuals with and without a family history of migraine. We genotyped 150 subjects (100 migraine patients: 50 with migraine without aura (MO), 50 with migraine with aura (MA) and 50 controls) for six CACNA1A

Mood Disturbances in a Patient on Statin Therapy: A Case Report.

Statins (3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitors) are widely used to treat hyperlipidaemia and significantly reduce the risk of cardiovascular events. While generally well tolerated, emerging evidence suggests that statins may, in rare cases, be associated with neuropsychiatric adverse effects, including mood disturbances and behavioural changes. We report the case of a 54-year-old woman with hypertension, but no personal or family history of psychiatric illness, who developed mood and behavioural changes shortly after initiating atorvastatin 10 mg following the detection of

Salivaomics: New Frontiers in Studying the Relationship Between Periodontal Disease and Alzheimer's Disease.

BACKGROUND: This study explores the link between oral biofluids, microbial dysbiosis, and Alzheimer's disease (AD), highlighting saliva and gingival crevicular fluid (GCF) as non-invasive diagnostic sources. AD onset and progression appear to be influenced not only by genetic and environmental factors but also by changes in the oral microbiome and related inflammatory and metabolic alterations. As global populations age, the incidence of AD is projected to rise significantly. Emerging evidence implicates the oral microbiome and salivary metabolites in neurodegenerative pathways, suggesting tha

Valproate Exposure as an In vitro Model for Studying Morpho-Molecular Features of ASD: A Systematic Review.

INTRODUCTION: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder with a strong genetic and environmental basis. It frequently causes social and communication deficits, as well as repetitive behaviours. Valproic acid (VPA) has been shown to induce autisticlike features in animal models when administered during critical development periods. However, not much is known about its effect on cells to replicate ASD characteristics in vitro. OBJECTIVE: This review explores in vitro VPA models to elucidate the molecular and morphological characteristics of ASD, emphasizing their po

Global epidemiology and burden of interstitial lung disease.

The global incidence and prevalence of interstitial lung disease (ILD) are difficult to determine due to the rarity of the condition, inconsistent case ascertainment and reporting methods, and differences in risk factors, burden of exposures, and access to health-care systems across geographical areas. With these caveats, several studies have reported an increased incidence and prevalence of ILD over time. Idiopathic pulmonary fibrosis is the most common and extensively studied ILD, whereas connective tissue disease-associated ILD and hypersensitivity pneumonitis display the most geographical

Testing the Level of Cortisol in Dogs.

This paper focuses on cortisol levels in different dog matrices, emphasising the importance of non-invasive sampling for animal welfare and research ethics. The study reviewed over 18,000 articles from various databases, identifying 78 relevant studies that met the inclusion criteria. The findings reveal a growing trend for non-invasive matrices like saliva, hair, and urine, reliably reflecting acute and chronic stress responses while minimising animal stress during sampling. Other sample types, such as blood, are becoming less and less used due to their stress-inducing factors. Such an approa

Next-Generation Sequencing Reveals the Potential Role of RET Protooncogene in Metastasis Progression in Medullary Thyroid Cancer.

Background: Medullary thyroid carcinoma (MTC) has a high rate of local and distant metastases. In particular, the RET protooncogene appears to be the predominant driver mutation for oncogenesis. The German S3 thyroid carcinoma guidelines recommend molecular genetic analysis of the tumour without specifying the site of the tissue sampling. Whether there is difference in RET protooncogene between the primary tumour, lymph node, and distant metastasis has not yet been investigated. However, differences could be important with regard to biopsy localization, and also, thus, the choice of single- or

Genetic evidence linking gastroesophageal reflux disease to chronic kidney disease and kidney failure: a two-step Mendelian randomization study.

To investigate the causal association between gastroesophageal reflux disease (GERD) and chronic kidney disease (CKD) progression and its potential metabolic mediators. Summary-level data were extracted from the overall genome-wide association studies of the FinnGen and UK Biobank databases. This study employed two-sample, bidirectional, two-step, and multivariable Mendelian randomization (MR) techniques, utilizing single-nucleotide polymorphisms (SNPs) as genetic instruments for exposure and mediators, thereby minimizing bias due to confounders and reverse causation. We harnessed summary-leve

Proteome Signature of Alzheimer-Like Phenotypes in Frontal Cortices From Young and Old Individuals With Down Syndrome.

Down syndrome (DS) stands out as the most prevalent genetic contributor to intellectual disability, marked by the presence of an extra copy of chromosome 21 (HSA21). Notably, individuals with DS exhibit significant neuropathological changes for a diagnosis of Alzheimer's disease (AD), typically by the age of 50 years. To search for and identify biomarkers crucial for detecting and understanding the mechanisms involved in DS neuropathology, we conducted a protein expression analysis of post-mortem brain samples. We evaluated the frontal cortex of post-mortem brain samples from patients with DS
