Current and novel food allergens: Cross-reactivity and co-sensitisation risks.

Food allergy currently affects approximately 10 % of the global population, representing a significant health challenge with no known cure. This chapter describes the current and emerging food allergens, mechanisms of cross-reactivity and co-sensitisation, clinical implications and management, as well as future directions and research gaps. While current allergens such as milk, eggs, peanuts or crustaceans are the primary focus of existing regulations, the rise of novel protein sources, including edible insects, plant-based alternatives, algae and fungi, introduces new risks for de novo sensit

Ethanol extract of Dysosma versipellis induces cytotoxicity in colonic epithelial cells via activation of the PI3K-Akt/MAPK signaling pathways.

BACKGROUND: Dysosma versipellis (DV) has pharmacological activity but causes toxicity, especially in nontargeted tissues such as the colon. Understanding its underlying mechanism is crucial for safety. METHODS: UPLC-MS/MS was used to identify DV extract components. Active compounds (247) and targets (1901) were screened from databases; colon toxicity targets (1872) were obtained from GeneCards. The core targets were identified via network pharmacology and validated by molecular docking. The rats received DV extract for 5 days; the colon tissues were subjected to histopathology, transcriptomic

SRSF10 promotes cisplatin resistance in bladder cancer via BIN1 Exon 12 retention and ANXA1 activation.

RNA alternative splicing is a fundamental post-transcriptional mechanism that plays a key role in generating protein diversity. Previous studies from our group have shown that overexpression SRSF10 could promote liver cancer cell proliferation and invasion. However, its involvement in bladder cancer remains poorly understood. In the present study, we investigated SRSF10 expression using data from TCGA and GEO databases, immunohistochemistry, and proteomics. Functional validation was performed through in vitro and in vivo experiments, RNA sequencing, and bioinformatics analysis. Additionally, C

Fibrinogen-like protein 2-complement C3 interaction exacerbates tubular inflammation in acute kidney injury by elevating complement C3a levels.

Renal tubular epithelial cells are among the earliest renal parenchymal cells to be injured in the context of acute kidney injury (AKI). Numerous studies have confirmed that fibrinogen-like protein 2 (FGL2) can regulate the occurrence and development of inflammation during disease progression. We found that FGL2 expression is elevated under AKI conditions. However, the role of FGL2 in AKI remains unclear. To elucidate the role of FGL2 in AKI, we employed lentiviral and adeno-associated virus for transfection in cellular and murine models. Furthermore, by leveraging datasets from the gene expre

A patent perspective of RSK inhibitors to treat cancer (2020-present).

INTRODUCTION: The Ser/Thr protein kinase family, p90 ribosomal S6 kinase (RSK), can be activated by inputs from ERK1/2, PDK1 and in some cases MK1/MK2. Dysregulated RSK activity is implicated in diseases associated with inflammation such as cancer. There are four RSK family members of which RSK1/2 are tumor promoters whereas RSK3/4 can act as tumor suppressors. AREAS COVERED: The review summarizes patents that have identified compounds that decrease RSK activity. These patents all focus on the use of RSK inhibitors as cancer therapeutics. The patents were identified using the World Intellectu

Human Peripheral Myelin Protein 2 and Charcot-Marie-Tooth Disease or Structural Missense Variants Show Different Binding to Myelin-Like Lipid Monolayers.

The peripheral myelin protein P2 (also known as PMP2/FABP8) has two basic functions in vivo: the transport of fatty acids within the cell to the target compartments and the adhesion of lipid bilayers in the peripheral nervous system myelin to each other. In this work, we focus on the membrane-binding properties of P2 and its Charcot-Marie-Tooth disease variants. Experiments were carried out on lipid monolayers at the air-water interface as a membrane model system, with the lipid composition of the cytoplasmic leaflet of myelin. Our study provides quantitative data on the membrane affinity of P

Chlorpyrifos and tamoxifen co-pretreatment promotes stem-like phenotype and upregulation of anti-estrogen therapy resistance markers in ERα breast cancer cells.

Endocrine therapy (ET) is the standard first-line treatment for hormone receptor positive breast cancer (BC) but about 30 % of patients develop resistance. Chlorpyrifos (CPF), is an endocrine disruptor pesticide that promotes a stemness phenotype and triggers molecular mechanisms involved in ET resistance. The goal of this study is to assess whether CPF induces tamoxifen (TAM) resistance by modulating estrogen receptor α (ERα) expression through histone deacetylase 1 (HDAC1) regulation. We analyzed public databases of BC patients and performed in vitro experiments using the MCF-7 cell line. We
