American journal of medical genetics. Part B, Neuropsychiatric genetics : the official publication of the International Society of Psychiatric Genetics2003eng
This is an initial report on a six-site collaborative project, Genetics of Recurrent Early-Onset Depression (GenRED). This is a study of a large sample of families with recurrent major depressive disorder (DSM-IV) beginning by the age 30 in probands or 40 in relatives. Evidence suggests that early onset and recurrence of depressive episodes predict substantially increased risk of depression in first-degree relatives compared with the general population, suggesting that susceptibility genes might be mapped with this phenotype. The projected sample of 800-1,000 affected sibling pairs (ASPs) and

With increasing access to individual-level data from genome-wide association studies, it is now common for researchers to have individual-level data of some traits, whereas for other traits only publicly released summary statistics are available due to privacy and safety concerns. Current genetic-correlation methods require both traits to be in the same format. When one trait has individual-level data and the other has only summary statistics, researchers often convert individual-level data to summary statistics and then apply summary-based methods, which is inefficient and can lose informatio

In populations with low genetic diversity, the capacity to adapt to environmental change is limited. While epigenetic mechanisms can generate phenotypic plasticity, their role in mediating adaptive life-history transitions in the wild is largely unknown. Here, we show that in adult Namibian cheetahs (Acinonyx jubatus jubatus), a species with remarkably low genetic diversity, the transition from a floater to a male holding a territory is associated with widespread DNA methylation changes, with a predominance of hypermethylation in territorial males. Using paired blood samples collected from the

Mate choice influences levels of inbreeding and genetic diversity in natural populations. Positive assortative mating (PAM), for instance, has been described in Ixodes ricinus L., but this behavior has yet to be investigated in the closely related Ixodes scapularis Say, 1821. By analyzing nine microsatellite loci of 30 females and 30 males removed in copula from hunter-harvested deer specimens at the Savannah River Site (South Carolina) during the hunting season of fall 2010, we found a weak but significant assortative pairing signature in a population that, otherwise, appeared genetically hom

Systemic lupus erythematosus (SLE) is a complex autoimmune condition that disproportionately impacts non-White ethnic and racial groups, particularly individuals in the African diaspora who experience heightened incidence, prevalence, and adverse outcomes. Genetic and epigenetic factors play significant roles in SLE risk, however these factors neither explain the whole of SLE risk nor the stark racial disparities we observe. Moreover, our understanding of genetic risk factors within African ancestry populations is limited due to social and environmental influences on research participation, di

Cerebrotendinous xanthomatosis (CTX) is a rare, autosomal recessive bile acid synthesis disorder caused by pathologic variants in CYP27A1, a gene involved in bile acid synthesis. Impaired function in this gene leads to accumulation of plasma cholestanol (PC) in various tissues, often in early childhood, resulting in such clinical signs as infantile diarrhea, early-onset bilateral cataracts, and neurological deterioration. The current study aimed to identify cases of CTX in a population of patients with a greater CTX prevalence than the general population, to facilitate early diagnosis. Patient

Non-syndromic cleft lip with/without cleft palate (NSCL/P) is one of the most common birth defects in humans with an overall prevalence of one per 1000 live births. Due to genetic and environmental influences, the fusion of the lips or palate may be interrupted at any stage and cause a cleft. Over decades, dozens of susceptible genes and loci have been identified using multiple genetic approaches. Our group has collected samples of NSCL/P patients since 2008 and established the biobank. We discovered numerous susceptible loci related to the occurrence of NSCL/P in the Chinese population, such

This paper reviews and summarizes three main aspects of spinocerebellar ataxias (SCA) in the Asian population. First, epidemiological studies were comprehensively reviewed. Overall, the most common subtypes include SCA1, SCA2, SCA3, and SCA6, but there are large differences in the relative prevalence of these and other SCA subtypes between Asian countries. Some subtypes such as SCA12 and SCA31 are rather specific to certain Asian populations. Second, we summarized distinctive phenotypic manifestations of SCA patients of Asian origin, for example a frequent co-occurrence of parkinsonism in some

BACKGROUND: Celiac disease (CD) is a systemic immune-mediated disorder developing in HLA genetically predisposed individuals carrying HLA-DQ2 and/or HLA-DQ8 molecules. Recent evidences supported a predominant importance of HLA-DQB1 locus and, in particular, HLA-DQB1*02 alleles. This diagnosis is poorly considered in Kazakhstan, because of the assumption that CD is not prevalent in this population.
OBJECTIVE: To demonstrate that the genetic predisposition to CD in Kazakhstan is not negligible and is actually comparable to Western populations.
METHODS: Through the analysis of HLA-DQ genotypes

Primary hyperparathyroidism is commonly caused by excess production of parathyroid hormone from sporadic parathyroid adenomas. However, the genetic architecture of sporadic primary hyperparathyroidism remains largely uncharacterized, especially in the Chinese population. To identify genetic abnormalities that may be involved in the etiology of sporadic parathyroid adenomas and to determine the mutation frequency of previously identified genes in the Chinese population, we performed whole-exome sequencing of 22 blood-tumor pairs from sporadic parathyroid adenomas. The most important finding is

Population-based testing for BRCA1/2 mutations detects a high proportion of carriers not identified by cancer family history-based testing. We sought to determine whether population-based testing is an effective approach to genetic testing in the Bahamas, where 23% of women with breast cancer carry one of seven founder mutations in the BRCA1 or BRCA2 gene. We determined the prevalence of founder BRCA mutations in 1847 Bahamian women without a personal history of breast or ovarian cancer, unselected for age or family history. We found that 2.8% (20/705) of unaffected women with a family history

The pathogenesis of periodontal disease is not solely based on microbial dental plaque but is the result of the multifactorial and complex interaction between infection and host response. Many studies in the literature have demonstrated the differences between individuals in terms of host immune response and the presence of genetic components in numerous pathological conditions. Therefore, periodontitis may be defined as a complex genetic disorder with a phenotype formed by the genetic structure and environmental factors in the affected individual. So, determination of the genetic susceptibili

Metabolic syndrome (MetS) is a common complex trait consisting of the clustering of abdominal obesity, hypertension, dyslipidemia, and dysglycemia. MetS is found in about 25% of the population in the United States and is associated with increased risk for type 2 diabetes and cardiovascular disease. Despite research into possible genetic influences for MetS, no consistently reproducible genetic markers have been obtained, partially due to lack of agreement on the definition of the phenotype. Because phenotypic precision is essential for genomic interrogation, the evolving discipline of clinical

BACKGROUND: Fibroblast growth factor receptor 3 (FGFR3) mutation is classically known to cause a spectrum of autosomal dominant skeletal dysplasias through gain-of-function mechanisms. However, because most genetic studies are limited to symptomatic individuals or high-risk families, the unbiased variant carrier frequency, variants spectrum, and disease prevalence in newborns remain largely uncharacterized.
METHODS: We performed high-throughput targeted next-generation sequencing of the FGFR3 gene on 39,104 Chinese newborns from a multi-center cohort. Variants were classified according to the

BACKGROUND: The course of disease development during the coronavirus disease 2019 (COVID-19) pandemic has demonstrated a very wide spectrum, with the most vulnerable group of severe disease comprising > 10% of cases worldwide. Previously, several clinical and laboratory phenotypes have been suggested for the prediction of severe disease courses with different impacts in diverse populations. METHODS: Using a logistic regression model, we performed a study of 227 patients (37% with severe disease), all of whom were ethnically Uzbek, to identify predisease clinical phenotypes associated with dise

AIMS: Available data on economic consequences of carrying an inherited familial hypercholesterolaemia (FH) genetic variant are sparse. This study aims to explore the hospital and pharmaceutical resource use and costs associated with cardiovascular disease (CVD) in patients with genetically verified FH compared with age- and sex-matched controls during the period 2010 through 2019.
METHODS AND RESULTS: We included 5585 individuals with genetically verified FH from the Norwegian Unit for Cardiac and Cardiovascular Genetics registry and 111 483 age- and sex-matched controls from the general Norw

The processes of labor and birth have a complex evolutionary history, with substantial variation among species showing differences in gestational length, offspring number, anatomy, and rates of fetal development. Understanding the genomic basis of pregnancy is therefore a focus of evolutionary research, given the importance of reproductive success in processes such as natural selection, mutation, genetic drift, and migration. Disruptions to normal pregnancy processes include preterm birth, which can arise from multiple factors, including infection, anatomical variation, injury, age, parity, an

BACKGROUND: Prostate cancer (PCa) remains a leading malignancy among men worldwide, with inherited genetic susceptibility playing a pivotal role in its pathogenesis. Although numerous single nucleotide polymorphisms (SNPs) within immune response, inflammatory, and apoptotic pathways have been linked to PCa risk, these associations often exhibit substantial inconsistency across diverse populations. This study aimed to evaluate the association of selected polymorphisms within the RNASEL, ELAC2, MSR1, and KLK3 genes with PCa susceptibility in a Hungarian cohort.
METHODS: This case-control study

OBJECTIVE: Prostate cancer (PCa) is among the most prevalent malignancies in men, with a substantial genetic predisposition. Despite extensive genome-wide association studies (GWAS) identifying numerous risk loci, the causal target genes remain largely undetermined. To address this knowledge gap, we conducted the first transcriptome-wide association study (TWAS) of PCa in the Chinese population to systematically identify putative target genes.
METHODS: This study included 5048 participants, comprising 2425 individuals from the Chinese Prostate Cancer Genetics Consortium cohort (1417 PCa cases

The Apolipoprotein L1 (APOL1) innate immunity gene product represents the sole member of the APOL gene family in humans capable of secretion into circulation, thereby mediating the trypanolysis of T. brucei brucei. Gain-of-function variants of the APOL1 gene originated and spread among human population groups to extend APOL1's protective capacity to include also serum-resistant subspecies, such as T. brucei gambiense (S342G known as APOL1-G1) and T. brucei rhodesiense (N388_Y389del known as APOL1-G2). The biochemical pathways underlying the lytic activity of these evolutionary favored mutation
