Hematopoietic Antigen Presenting Cells Fine-Tune the Effector Phenotype of Intrathymic Unconventional αβ T Cells.

The development of unconventional αβ T cells, including invariant natural killer T (iNKT) and mucosal-associated invariant T (MAIT) cells, in the thymus is distinct from conventional T cells. Unconventional αβ T cells adopt a memory phenotype, acquire effector functions, and can reside in the thymus long-term. It is well-established that positive selection of these unconventional T cells from CD4+CD8+ double-positive (DP) precursors depends on interaction with other DP cells. However, postselection, the regulation of their maturation and effector differentiation is less well understood. Profes

Immune priming and haemolymph-mediated protection in Mytilus edulis against Vibrio splendidus.

Improving disease resistance in farmed bivalves is essential for the long-term sustainability of aquaculture, yet practical immune-based approaches remain scarce. In a few species, immune priming has shown encouraging potential, where a first encounter with a pathogen enhances the host's ability to cope with later infections by improving recognition and clearance of encountered microbes. In this study, we report the first evidence of immune priming in the blue mussel Mytilus edulis, a species of major commercial value. We show that mussels previously exposed to Vibrio splendidus, whether live

[Use of vaccines to prevent the development of bacterial resistance].

Vaccinations against bacterial pathogens play a crucial role in infection prevention, especially given the growing prevalence of antibiotic resistance. Historically, vaccines against cholera, diphtheria, and pertussis (whooping cough) marked the first significant advances in the fight against bacterial diseases. Later, vaccines against meningococcus and tetanus, among others, were added. These active immunizations expose the immune system to components of the pathogen, stimulating the production of antibodies, T helper cells, B cells, and memory cells, thereby providing long-term protection. Pooled screening for CAR function identifies novel IL-13Rα2-targeted CARs for treatment of glioblastoma.

BACKGROUND: Chimeric antigen receptor (CAR) therapies have demonstrated potent efficacy in treating B-cell malignancies, but have yet to meaningfully translate to solid tumors. Nonetheless, they are of particular interest for the treatment of glioblastoma, which is an aggressive form of brain cancer with few effective therapeutic options, due to their ability to cross the highly selective blood-brain barrier. METHODS: Here, we use our pooled screening platform, CARPOOL, to expedite the discovery of CARs with antitumor functions necessary for solid tumor efficacy. We performed selections in pr

Combined CD25-targeted near infrared photoimmunotherapy (NIR-PIT) and intratumor IL-15 enhance the effectiveness of anti-PD-1 immunotherapy.

Immunotherapies aimed at enhancing anti-cancer immunity are now in wide clinical use. For instance, anti-programmed cell death protein 1 (PD-1) monoclonal antibody (mAb) therapy has demonstrated clinical efficacy in many tumor types, but overall, only a minority of patients respond. Near-infrared photoimmunotherapy (NIR-PIT) is a novel cancer treatment that selectively destroys targeted cells. CD25-targeted NIR-PIT is a strategy that selectively depletes regulatory T cells in the tumor microenvironment, thereby enhancing the T-cell response of PD-1 mAbs. Cytokines, such as interleukin-15 (IL-1

Evaluating Machine Learning-Based Soft Sensors for Effluent Quality Prediction in Wastewater Treatment Under Variable Weather Conditions.

Maintaining effluent quality in wastewater treatment plants (WWTPs) comes with significant challenges under variable weather conditions, where sudden changes in flow rate and increased pollutant loads can affect treatment performance. Traditional physical sensors became both expensive and susceptible to failure under extreme conditions. In this study, we evaluate the performance of soft sensors based on artificial intelligence (AI) to predict the components underlying the calculation of the effluent quality index (EQI). We thus focus our study on three ML models: Long Short-Term Memory (LSTM),

Synbiotic intervention reverses alcohol drinking-induced cognitive deficits in adolescent male mice by modulating the microbiota-gut-brain axis.

Adolescence is characterized by an increased vulnerability to substance abuse, including alcohol consumption. We investigated the effects of a synbiotic intervention on disruptions of the microbiota-gut-brain axis induced by a drinking in the dark model of intermittent alcohol exposure in adolescent mice. We found that alcohol drinking induced specific shifts in gut microbiota, namely it increased Erysipelotrichaceae and reduced fecal butyric and isovaleric acids. In adulthood, other types of gut bacteria were affected such as Rhodospirillales uncultured family and Entrorhabdus uncultured bact

Cross-Layer Analysis of Machine Learning Models for Secure and Energy-Efficient IoT Networks.

The widespread adoption of the Internet of Things (IoT) raises significant concerns regarding security and energy efficiency, particularly for low-resource devices. To address these IoT issues, we propose a cross-layer IoT architecture employing machine learning (ML) models and lightweight cryptography. Our proposed solution is based on role-based access control (RBAC), ensuring secure authentication in large-scale IoT deployments while preventing unauthorized access attempts. We integrate layer-specific ML models, such as long short-term memory networks for temporal anomaly detection and deci

A single dose of lamotrigine induces a positive memory bias in healthy volunteers.

BACKGROUND: Lamotrigine has been shown to be effective in the long-term treatment and relapse prevention of depression in bipolar disorder. However, the neuropsychological mechanisms underlying these effects are unclear. We investigated the effects of lamotrigine on a battery of emotional processing tasks in healthy volunteers, previously shown to be sensitive to antidepressant drug action in similar experimental designs. METHODS: Healthy volunteers (n = 36) were randomized in a double-blind design to receive a single dose of placebo or 300 mg lamotrigine. Mood and subjective effects were mon
