Bilirubin metabolism and its application in disease prevention: mechanisms and research advances.

The role of bilirubin, a product of heme metabolism, has evolved from a traditionally perceived metabolic waste product to a critical molecule with diverse biological roles. This article comprehensively reviews the metabolic functions of bilirubin and advances in its application for disease prevention. Bilirubin is primarily derived from hemoglobin catabolism in senescent erythrocytes. It is subsequently metabolized and excreted by the liver through tightly regulated processes involving enzymes, nuclear receptors, hormones, and pharmaceuticals. Bilirubin exhibits diverse physiological function

Impact of disease type and temporal trends on the proximity of palliative care initiation to death in older adults: a population-based study.

BACKGROUND: Chronic conditions like heart disease and dementia have surpassed cancer as leading causes of death in aging populations. Non-cancer patients, despite similar palliative care needs, often face delayed initiation or limited access. This study investigated the proximity of palliative care initiation to death among older adults with different diagnoses and influencing factors. METHODS: This cross-sectional study used Taiwan's National Health Insurance and Multiple Causes of Death databases (2010-2020), including individuals aged ≥65 who received specialist palliative care before deat

From pathophysiology to novel approaches for obesity-associated hypertension.

Obesity is a rapidly growing epidemic affecting >15% of the global adult population and has considerable clinical consequences and comorbidities, including hypertension, diabetes mellitus, cardiovascular and cerebrovascular diseases and chronic kidney disease. There is a strong association between obesity or body mass index and high blood pressure (BP) in epidemiological studies while the underlying pathophysiological events linking those conditions are not fully elucidated. Hypothetical mechanisms include a sedentary lifestyle and excess intake of processed foods that contribute to obesity, o

Cellular and molecular mechanisms of pathological tau phosphorylation in traumatic brain injury: implications for chronic traumatic encephalopathy.

Tau protein plays a critical role in the physiological functioning of the central nervous system by providing structural integrity to the cytoskeletal architecture of neurons and glia through microtubule assembly and stabilization. Under certain pathological conditions, tau is aberrantly phosphorylated and aggregates into neurotoxic fibrillary tangles. The aggregation and cell-to-cell propagation of pathological tau leads to the progressive deterioration of the nervous system. The clinical entity of traumatic brain injury (TBI) ranges from mild to severe and can promote tau aggregation by indu

Overall effects of microplastics on brain.

Microplastic (MP) and nanoplastic (NP) pollution represents a pervasive environmental issue, raising significant concerns regarding potential neurotoxicity and impacts on brain health. This review synthesizes recent research findings to provide a comprehensive overview of the effects of MPs/NPs on the brain. Evidence demonstrates that MPs/NPs can cross critical biological barriers, including the blood-brain barrier and the placenta, gaining access to the central nervous system (CNS) and the developing fetal brain, influenced by particle size, charge, and the biomolecular corona. Once present,

Pyridazine and Pyridazinone in Medicinal Chemistry: Synthesis and Antiinflammatory Pathways Targeting TxA2, TNF-α, and IL-6.

Inflammation is a fundamental biological reaction to harmful stimuli, which is crucial in the initiation and advancement of different diseases, including rheumatoid arthritis, cardiovascular conditions, neurological disorders such as Alzheimer's and Parkinson's, and multiple cancer types. Chronic inflammation, in particular, contributes to irreversible tissue damage and the progression of disease. Thus, the suppression of key inflammatory mediators has become a promising therapeutic approach. Thromboxane A2 (TxA2), tumor necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) are among the med

The Roles of EDA2R in Ageing and Disease.

Ageing is a complex biological process driven, in part, by inflammaging. Recent research identifies the ectodysplasin A2 receptor (EDA2R) as a key regulator of inflammaging and a novel biomarker of ageing, with its expression increasing with age across diverse tissues in humans and animal models. Elevated EDA2R gene expression is associated with accelerated ageing, cellular senescence, frailty, obesity, acne, radiation response and increased levels of inflammatory, renal, cardiac and vascular biomarkers. Similarly, elevated EDA2R protein levels, a critical component of the proteomic ageing clo

The Identification of Potential Treatment Targets to Reduce the Risk of Obesity-Related Complications: A Step Toward a Treat-to-Target Approach in Obesity Management.

AIMS: Obesity-related complications (ORCs) are associated with substantial health and economic burdens. Although treatment targets are routinely used for other chronic conditions, none currently exist in obesity management. We aimed to identify an adiposity measure that indicates a reduced risk of four ORCs. MATERIALS AND METHODS: This population-based cohort study of patients aged 18-60 years used data from the UK Clinical Practice Research Datalink Aurum. Associations between absolute 10-year ORC risk and baseline body mass index (BMI), waist-height ratio (WHtR), and changes in adiposity me

Platelet-Rich Plasma (PRP): Molecular Mechanisms, Actions and Clinical Applications in Human Body.

Platelet-rich plasma (PRP) is an autologous blood-derived concentrate increasingly utilized in regenerative medicine for its ability to accelerate healing and tissue repair. PRP is broadly classified by leukocyte content, fibrin architecture, and platelet concentration, with classification systems developed to standardize characterization. Preparation methods, including single- or double-spin centrifugation and buffy coat techniques, influence the final composition of PRP, determining the relative proportions of platelets, leukocytes, plasma proteins, and extracellular vesicles. These componen

Antibody ligation of HLA class II induces YAP nuclear localization and formation of cytoplasmic YAP condensates in human endothelial cells.

Antibody (Ab) crosslinking of HLA class II (HLA II) molecules on the surface of endothelial cells (ECs) triggers proliferative and prosurvival intracellular signaling, which are implicated in promoting chronic Ab-mediated rejection (cAMR). Despite the importance of cAMR in transplant medicine, the mechanisms involved remain incompletely understood. Here, we examined the regulation of yes-associated protein (YAP) nuclear cytoplasmic localization and phosphorylation in human ECs challenged with Abs that bind HLA II, which are strongly associated with cAMR. To examine changes in YAP localization

MGluR5 Agonist CHPG
