Pregabalin synchronizes the regeneration of nerve and muscle fibers optimizing the gait recovery of MDX dystrophic mice.

Duchenne muscular dystrophy (DMD) is an X-linked genetic disorder induced by mutations in the dystrophin gene, leading to a degeneration of muscle fibers, triggering retrograde immunomodulatory, and degenerative events in the central nervous system. Thus, neuroprotective drugs such as pregabalin (PGB) can improve motor function by modulating plasticity, together with anti-inflammatory effects. The present work aimed to study the effects of PGB on axonal regeneration after axotomy in dystrophic and non-dystrophic mice. For that, MDX and C57BL/10 mouse strains were subjected to peripheral nerve

Glycerophospholipid profile alterations are associated with murine muscle-wasting phenotype.

INTRODUCTION: Phospholipids are essential components of cellular membranes and are closely associated with cellular functions, but relationships involving skeletal muscle phospholipid profiles and their physiological phenotypes have remained unclear. METHODS: We carried out comprehensive phospholipid analyses using liquid chromatography-tandem mass spectrometry to determine the phospholipid profiles of skeletal muscles derived from muscle-wasting mouse models, including denervated and Duchenne muscular dystrophy mouse models (mdx) as well as rescued mdx mice expressing truncated dystrophin.

Modifier genes: Moving from pathogenesis to therapy.

This commentary will focus on how we can use our knowledge about the complexity of human disease and its pathogenesis to identify novel appr … rticle explored protective modifiers, including plastin 3 (PLS3) and coronin 1C (CORO1C), in spinal muscular atrophy (SMA). SMA is an autosomal recessive deficit of survival motor neuron protein (SMN) caused by mutations in SMN1. However, the severity of SMA is determined primarily by the number of SMN2 copies, and this results in significant phenotypic variabilit …

Targeting RNA to treat neuromuscular disease.

The development of effective therapies for neuromuscular disorders such as Duchenne muscular dystrophy (DMD) is hampered by considerable challenges: skeletal muscle is the most abundant tissue in the body, and many neuromuscular disorders are multisystemic conditions. However, despite these barriers there has recently been substantial progress in the search for novel treatments. In particular, the use of antisense oligonucleotides, which are designed to target RNA and modulate pre-mRNA splicing to restore functional protein isoforms or directly inhibit the toxic effects of pathogenic RNAs, off

Flexion contractures possibly reflect the existence of hypocortisolism: two case reports.

The diagnosis of adrenal insufficiency is discussed in case of low blood pressure and digestive symptoms. Rare inaugural abdomino-crural muscle contracture can be a misleading symptom. Here we report two new cases. A 50-year-old woman presenting a leaning forward walking attitude and negligence for the past 2 months was referred to the neurologic unit. Abdomino-crural contracture, clinical hypogonadism, and hyponatremia directed towards a panhypopituitarism, which was confirmed by subsequent investigations. Pituitary MRI found an empty sella turcica. The outcome was dramatic after hormone repl

Disuse atrophy of human skeletal muscle: cell signaling and potential interventions.

In response to atrophic stimuli, physical alterations include decreases in fiber diameter and contractile protein content. Despite the fact that these phenotypical alterations have been well characterized, the signaling pathways that mediate these adaptations are still under investigation. There have been significant advances in the past few years delineating signal transduction pathways that regulate protein turnover. In the process of evaluating the effect of various atrophy-inducing stimuli on signal transduction pathways in skeletal muscle, it is apparent that differences do exist concerni

Pelvic tilt in neuromuscular disorders.

The pelvic tilt is part of a complex deformity in which both the spine and the hip are involved. By viewing the different planes and analyzing the deforming forces, four different types of pelvic tilt in combination with spinal and hip deformities can be identified. In spina bifida patients congenital deformities of the spine may add to the progression of scoliosis. Analyzing our own patients we found that certain types of pelvic tilt can be related to certain neuromuscular diseases. Moreover our results show that the pelvic tilt can be effectively corrected by spinal surgery, whereas no effec

Autonomic, locomotor and cardiac abnormalities in a mouse model of muscular dystrophy: targeting the renin-angiotensin system.

New Findings What is the topic of this review? This symposium report summarizes autonomic, cardiac and skeletal muscle abnormalities in sarcoglycan-δ-deficient mice (Sgcd-/-), a mouse model of limb girdle muscular dystrophy, with emphasis on the roles of autonomic dysregulation and activation of the renin-angiotensin system at a young age. What advances does it highlight? The contributions of the autonomic nervous system and the renin-angiotensin system to the pathogenesis of muscular dystrophy are highlighted. Results demonstrate that autonomic dysregulation precedes and predicts later develo

LMNA c.1622G>A mutation and myopathic changes in a family with limb-girdle muscular dystrophy: A case report.

The present study reports a case of limb-girdle muscular dystrophy (LGMD) associated with a pathogenic lamin A/C (LMNA) mutation (c.1622G>A). Notably, this case expands the phenotypic and genotypic spectrum of LMNA-related LGMD, and provides novel familial clinical and genetic evidence for this rare mutation. Genetic sequencing revealed a heterozygous mutation in both the proband and the mother of the proband, suggesting autosomal dominant inheritance. Electromyography (EMG) revealed reduced nerve conduction velocity and abnormal potentials in the proband and mother, indicating muscle weakness

Targeted Molecular Therapies for SBMA.

Spinal and bulbar muscular atrophy (SBMA) is a late-onset neuromuscular disease caused by a polyglutamine expansion in the androgen receptor gene which results in progressive spinal and bulbar motor neuron degeneration, and muscle atrophy. Although the causative genetic defect is known, until recently, the molecular pathogenesis of the disease was unclear, resulting in few, if any, targets for therapy development. However, over the past decade, our understanding of the pathomechanisms that play a role in SBMA has increased dramatically, and several of these pathways and mechanisms have now bee…

Hypomorphic Smn knockdown C2C12 myoblasts reveal intrinsic defects in myoblast fusion and myotube morphology.

Dosage of the survival motor neuron (SMN) protein has been directly correlated with the severity of disease in patients diagnosed with spinal muscular atrophy (SMA). It is also clear that SMA is a neurodegenerative disorder characterized by the degeneration of the alpha-motor neurons in the anterior horn of the spinal cord and atrophy of the associated skeletal muscle. What is more controversial is whether it is neuronal and/or muscle-cell-autonomous defects that are responsible for the disease per se. Although motor neuron degeneration is generally accepted as the primary event in SMA, intrin…

The mouse mutation muscle deficient (mdf) is characterized by a progressive motoneuron disease.

Muscle deficient (mdf) is an autosomal-recessive mutation mapped to mouse chromosome 19. The clinical phenotype and the muscle histopathology, briefly described in 1980, and the nervous system histopathology are detailed in the present study. Homozygotes develop a posterior waddle at 4 to 8 weeks of age. Soon thereafter, the hindlimbs become paralyzed and weakness appears in forelimbs, leading to a serious disability. The disease progresses slowly and the mean lifespan is reduced to 8 months. Skeletal muscles exhibit a neurogenic atrophy with signs of reinnervation. Peripheral nerves display a

Why muscle strengthening exercises should target the quadriceps and gluteus maximus in patients with knee osteoarthritis?: Effects of knee pain on muscle volume and fatty degeneration based on AI-assisted cross-sectional analysis.

BACKGROUND: Sarcopenia, characterized by age-related muscle loss, traditionally presents as systemic muscle atrophy. However, knee osteoarthritis (OA) patients often exhibit localized quadriceps muscle atrophy, suggesting a potential pain-related mechanism distinct from classical sarcopenia. This study aimed to investigate the relationship between knee OA pain and selective muscle atrophy, particularly focusing on its role as a potential aggravating factor for sarcopenia. METHODS: Eighty-nine patients scheduled for unilateral primary total knee arthroplasty were analyzed. Preoperative compute
