Alterations in static and dynamic intrinsic brain local connectivity and associated molecular analysis in drug-naïve first-episode schizophrenia: Insights from resting-state functional magnetic resonance imaging.

Schizophrenia is a severe psychiatric disorder marked by widespread brain abnormalities. Recent studies suggest that pathological changes may originate from focal 'epicenter' regions and subsequently spread to other brain areas strongly connected to them. Investigating drug-naïve first-episode schizophrenia (dn-FES) patients may help characterize early-stage regional functional abnormalities and their potential neurochemical underpinnings. Resting-state functional magnetic resonance imaging data were acquired from 50 dn-FES patients and 50 age- and sex-matched healthy controls (HCs). Static re

Alpha-band fronto-limbic hyperconnectivity in adolescents with major depression and suicidal ideation.

BACKGROUND: Adolescents with major depressive disorder (MDD) are at high risk for suicide, but the neurophysiological correlates of suicidal ideation remain unclear. Resting-state EEG functional connectivity offers a promising approach to investigating brain network alterations associated with suicide risk. METHODS: We recruited first-episode, treatment-naïve adolescents with MDD with suicidal ideation (SI-MDD, n = 30), without suicidal ideation (NSI-MDD, n = 38), and healthy controls (HC, n = 33). Resting-state EEG was used to construct source-level functional connectivity networks based on t

Diencephalic integrity explains aspects of hippocampal amnesia.

Research on amnesia has been fundamental in establishing the role of the human hippocampus in memory. Even though other structures within the hippocampal-diencephalic-cingulate network also play a role in episodic memory, studies of hippocampal amnesia often ignore the importance of changes in this broader network. In a large cohort of patients (n = 38) with hippocampal damage due to autoimmune limbic encephalitis, we previously found that amnesia was predominantly explained by resting-state functional abnormalities across this network. Here, we examined the integrity of individual diencephali

Projections from Regions of the Cerebellar Nuclei Receiving Jaw Muscle Proprioceptive Signals to Trigeminal Motoneurons and Their Premotoneurons in the Rat Pons and Medulla.

The cerebellum plays a crucial role in sensorimotor control through cerebellofugal projections from the cerebellar nuclei. However, little is known about the cerebellofugal projection features involved in jaw sensorimotor control, although the dorsolateral parts of the interposed cerebellar nucleus (IntDL) and medial cerebellar nucleus (MedDL) do receive proprioceptive signals bilaterally from rat jaw-closing muscle spindles (JCMSs). This study aimed to detail the cerebellofugal projection features involved in jaw sensorimotor control. Anterograde tracer was injected into regions of the rat In

Differential functional connectivity of amygdala in drug-resistant temporal lobe epilepsy.

OBJECTIVE: Recent studies highlight the amygdala's crucial role in temporal lobe epilepsy (TLE), particularly in magnetic resonance imaging-negative cases and new TLE subtypes with structural amygdala changes. This study aims to investigate the electrophysiological properties and connectivity patterns of the amygdaloid complex in TLE patients using stereoelectroencephalography (SEEG). METHODS: From March 2020 to December 2023, we collected data from nine patients with drug-resistant TLE who underwent SEEG with dual amygdala trajectories: dorsal amygdala (DA) targeting medial and central nucle

From resting-state functional hippocampal centrality to functional outcome: An extended neurocognitive model of psychosis.

BACKGROUND: We previously proposed a neurocognitive model of psychosis in which reduced morphometric hippocampal-cortical connectivity precedes impaired episodic memory, social cognition, negative symptoms, and functional outcome. We provided support for this model in a patient subtype, and aimed to extend these findings to resting-state functional MRI to potentially explain the progression for a broader range of patients. METHODS: We used a subsample of our previous analysis consisting of 54 patients with first-episode psychosis and 52 controls and applied the machine-learning algorithm Subt

Network-wide modulation of synaptic plasticity and spike patterns in motor circuits after pallidal deep brain stimulation in a dystonia model.

AIMS: We aimed to investigate the functional network effects of long-term deep brain stimulation (DBS) of the globus pallidus pars interna (GPi) on synaptic activity in motor thalamic and motor cortical neurons in an animal model of generalised dystonia (dtsz hamster) with a focus on understanding the underlying mechanisms of GPi-DBS in modulating neural activity. METHODS: Whole-cell recordings were performed on motor thalamic and motor cortical neurons from DBS-treated and sham-treated dtsz hamsters. Spontaneous synaptic activity was pharmacologically characterised and analysed by examining

Correlation of prefrontal-based resting state functional connectivity with the degree of cognitive impairment in Alzheimer's disease: a functional near-infrared spectroscopy study.

BACKGROUND: This study aims to investigate the differences in neural network connectivity within the prefrontal cortex (PFC) among elderly individuals with normal cognition (NC), mild cognitive impairment (MCI), and Alzheimer's disease (AD) using functional near-infrared spectroscopy (fNIRS). RESULTS: Significant differences in functional connectivity (FC) strength were observed between the NC, MCI, and AD groups in several Brodmann areas (BA) pairs, including BA46.L-BA45.R and BA9.L-BA1.L. The most pronounced FC strength differences between the NC and MCI groups occurred at the 2nd -minute m

The Secondary Motor Cortex-External Globus Pallidus Pathway Regulates Auditory Feedback of Volitional Control.

Effective use of brain-computer interfaces (BCIs) requires the ability to suppress a planned action (volitional inhibition) for adaptable control in real-world scenarios, but their mechanisms are unclear. Here, we used fiber photometry to monitor external globus pallidus (GPe) and subthalamic nucleus (STN) neurons' activity in mice during a volitional stop-signal task (67% GO, 33% NO-GO). GPe/STN neurons (receiving M2 projections) responded to auditory cues, feedback, and rewards in both trials. Importantly, chemogenetic activation of the M2-GPe pathway enhanced volitional inhibition by modula

Mapping hippocampal-cerebellar functional connectivity across the human adult lifespan.

The hippocampus and cerebellum are traditionally considered to support distinct memory systems, yet evidence from nonhuman species indicates a close relationship during spatial-mnemonic behaviour, with hippocampal projections to and from several cerebellar regions. However, little is known about this relationship in humans. To address this, we applied seed-based functional connectivity analysis to resting-state fMRI data from 479 cognitively normal participants aged 18-88 years. We identified significant functional correlations between the hippocampus and widespread areas of cerebellar cortex,

Aberrant functional gradient of thalamo-cortical circuitry in major depressive disorder and generalized anxiety disorder.

BACKGROUND: Functional gradient analysis provides insights into the brain's macroscale organization; however, the differences in thalamo-cortical gradients between major depressive disorder (MDD) and generalized anxiety disorder (GAD) remain unclear. Investigating these heterogeneities may uncover disorder-specific neural mechanisms and enhance diagnostic precision, addressing the distinct yet overlapping features of these affective disorders. METHODS: Resting-state functional MRI data were acquired from 88 healthy controls, 53 patients with MDD, and 28 patients with GAD. Functional gradient

Using in vivo MRI to reveal circuits that are sensitive to traumatic stress in male and female rats.

BACKGROUND: Post traumatic stress disorder (PTSD) is debilitating and more prevalent in women than men. While this suggests there are sex differences in the way neural systems respond to traumatic stress, identifying these systems are challenging. As such, studies designed to identify neural systems that are differentially sensitive to traumatic stress are needed. METHODS: We examined the impact of traumatic stress within a rat model (single prolonged stress (SPS)) on resting-state functional connectivity (rs-FC) and anatomical volume using in vivo MRI. Rs-FC and anatomical volume were measur

Resting-State Cortico-Cerebellar Connectivity Correlates with Post-Stroke Motor Recovery - A Prospective Functional MRI Study.

Cerebellar functional and structural connectivity are likely related to motor function after stroke. Less is known about motor recovery, which is defined as a gain of function between two time points, and about the involvement of the cerebellum. Fifteen patients who were hospitalized between 2018 and 2020 for a first cerebral ischemic event with persistent upper limb deficits were assessed by resting-state functional MRI (rsfMRI) and clinical motor score measurements at 3, 9 and 15 weeks after stroke. Age- and sex-matched healthy subjects (n = 15) were assessed once. The objectives were (1) to

Impulsivity and neuroticism share distinct functional connectivity signatures with alcohol-use risk in youth.

Increases in impulsivity and negative affect (e.g., neuroticism) are common during adolescence and are both associated with risk for alcohol-use initiation and other risk behaviors. Whole-brain functional connectivity approaches-when coupled with appropriate cross-validation-enable identification of complex neural networks subserving individual differences in dimensional traits (hereafter referred to as 'neural signatures'). Here, we analyzed functional connectivity data acquired at age 19 from individuals enrolled in a multisite European study of adolescent development (N ~ 1100) using connec

Regulation of craving and underlying resting-state neural circuitry predict hazard of smoking lapse.

Among individuals with substance use disorders, clinical outcomes may be improved by identifying brain-behavior models that predict drug re/lapse vulnerabilities such as the ability to regulate drug cravings and inhibit drug use. In a sample of nicotine-dependent adult cigarette smokers (N = 213), this laboratory study examined associations between regulation of craving (ROC) efficacy and smoking lapse, utilized functional connectivity multivariate pattern analysis (FC-MVPA) and seed-based connectivity (SBC) analyses to identify resting-state neural circuitry underlying ROC efficacy, and then

Distinguishing resilience from susceptibility: Brain network alterations during fear learning in response to childhood unpredictability.

Not all individuals exposed to childhood unpredictability develop psychopathology. This study breaks new ground by identifying the neural network signatures that distinguish resilience from susceptibility following such adversity. Using fear learning paradigms, we acquired fMRI data from 213 participants and constructed whole-brain functional connectivity networks. Innovatively, we employed dual resilience metrics-psychological (depressive symptoms) and biological (inflammatory TNF-alpha levels)-to classify participants into resilient and susceptible groups. Graph theory analysis found that th

Nicotine engages a VTA-NAc feedback loop to inhibit amygdala-projecting dopamine neurons and induce anxiety-like behaviors.

Nicotine activates ventral tegmental area (VTA) dopaminergic (DA) neurons projecting to the nucleus accumbens (NAc) to drive its reinforcing effects. Simultaneously, nicotine inhibits those projecting to the amygdala (Amg) to mediate anxiety-like behavior through a process that remains unknown. Here, we show that in male mice, NAc- and Amg-projecting DA neurons respond with similar polarities to ethanol and nicotine, suggesting a shared network-based mechanism underlying the inhibitory effect of these otherwise pharmacologically-distinct drugs. Selective activation of NAc-projecting DA neurons

Regulation of sociability by the cortico-habenula pathway in an animal model of depression.

Impaired sociability is a hallmark behavioral symptom frequently associated with depression. The medial prefrontal cortex (mPFC) is known to regulate both social behaviors and stress responses. Given the mPFC's projections to the lateral habenula (LHb) and the abnormal hyperactivity of the LHb observed in depression, the mPFC-LHb pathway may play a pivotal role in mediating impaired social behaviors in depressive disorders. Recent studies have reported increased activity of the mPFC-LHb pathway in depressive animal models. However, how this pathway responds to social stimuli and the synaptic d

Δ9-Tetrahydrocannabinol Alters Limbic and Frontal Functional Brain Connectomes Among Young Adult Cannabis Users.

BACKGROUND: Cannabis use among young adults has reached the highest levels ever recorded. Evidence indicates that acute Δ9-tetrahydrocannabinol (THC) disrupts brain connectivity. Few studies have examined this on a whole-brain level. We examined the effects of a single moderate dose of THC on resting-state functional brain networks among young adult cannabis users. METHODS: In a within-subject, double-blind, randomized study, 33 healthy occasional cannabis users received THC (7.5 mg, oral) and placebo before completing resting-state functional magnetic resonance imaging (rs-fMRI) during peak

Structural covariance network in the striatum is modulated by non-suicidal self-injury in adolescents with major depressive disorder.

BACKGROUND: Adolescents with major depressive disorder (MDD) significantly affect the development and well-being of this group. Approximately one-third of these adolescents engage in non-suicidal self-injury (NSSI) behavior, which notably increases their risk of suicide. Striatal abnormalities, particularly in the corticostriatal circuit, have been implicated in depression. The striatum is a crucial part of the reward system, and its abnormal connectivity with other brain regions within the system has been linked to repetitive NSSI behavior in adolescents. This study aims to investigate struct
