Serotonin system genes and obsessive-compulsive trait dimensions in a population-based, pediatric sample: a genetic association study.

BACKGROUND: Serotonin system genes are commonly studied in obsessive-compulsive disorder (OCD), but genetic studies to date have produced inconsistent results, possibly because phenotypic heterogeneity has not been adequately accounted for. In this paper, we studied candidate serotonergic genes and homogenous phenotypic subgroups as presented through obsessive-compulsive (OC) trait dimensions in a general population of children and adolescents. We hypothesized that different serotonergic gene variants are associated with different OC trait dimensions and, furthermore, that they vary by sex. M

The association between inefficient repair of oxidative DNA lesions and common polymorphisms of the key base excision repair genes as well as their expression levels in patients with rheumatoid arthritis.

INTRODUCTION: Rheumatoid arthritis (RA) is a common autoimmune heterogeneous joint disease of still unknown etiology. The pathology of RA leads to chronic inflammation of the joint tissues, which causes joint cartilage and bone destruction. One of the characteristic features of RA is oxidative stress, most likely induced and stimulated by inappropriate B- and T-cell activity. This results in accumulation of oxidative DNA lesions in peripheral blood mononuclear cells (PBMCs) isolated from RA patients, as we have shown previously. We have hypothesized that oxidative stress together with an impai

Coupling Neurogenetics (GARS™) and a Nutrigenomic Based Dopaminergic Agonist to Treat Reward Deficiency Syndrome (RDS): Targeting Polymorphic Reward Genes for Carbohydrate Addiction Algorithms.

Earlier work from our laboratory, showing anti-addiction activity of a nutraceutical consisting of amino-acid precursors and enkephalinase inhibition properties and our discovery of the first polymorphic gene (Dopamine D2 Receptor Gene [DRD2]) to associate with severe alcoholism serves as a blue-print for the development of "Personalized Medicine" in addiction. Prior to the later genetic finding, we developed the concept of Brain Reward Cascade, which continues to act as an important component for stratification of addiction risk through neurogenetics. In 1996 our laboratory also coined the te

[Individual susceptibility to occupational carcinogens: the evidence from biomonitoring and molecular epidemiology studies].

This paper reviews the literature on the influence of metabolic and DNA repair polymorphisms of biological indicators of genotoxic risk commonly used in biomonitoring occupational exposure to carcinogens. Genetic polymorphisms which influence biomarkers (urinary metabolites, protein and DNA adducts), include P450 cytochromes (CYPs) and glutathione S-transferases (GSTs) in exposure to polycyclic aromatic hydrocarbons (PAHs), and acetyltransferases (NATs) in exposure to aromatic amines (AAs). As regards exposure to benzene, also relevant is the influence of epoxydohydrolase (EPHX) and NAD(P)H qu

Mitochondrial DNA polymorphism in mitochondrial myopathy.

In order to test the hypothesis that mitochondrial myopathy may be caused by mutation of the mitochondrial (mt) genome, restriction fragment length polymorphism in leucocyte mt DNA has been studied in 38 patients with mitochondrial myopathy, 44 of their unaffected matrilineal relatives, and 35 normal control subjects. Previously unreported mt DNA polymorphisms were identified in both patients and controls. No differences in restriction fragment patterns were observed between affected and unaffected individuals in the same maternal line, and there was no evidence of major deletion of mt DNA in

PTPN22 intron polymorphism rs1310182 (c.2054-852T>C) is associated with type 1 diabetes mellitus in patients of Armenian descent.

Protein tyrosine phosphatase, nonreceptor type 22 (PTPN22), is an archetypal non-HLA autoimmunity gene. It is one of the most prominent genetic contributors to type 1 diabetes mellitus outside the HLA region, and prevalence of its risk variants is subject to enormous geographic variability. Here, we address the genetic background of patients with type 1 diabetes mellitus of Armenian descent. Armenia has a population that has been genetically isolated for 3000 years. We hypothesized that two PTPN22 polymorphisms, rs2476601 and rs1310182, are associated with type 1 diabetes mellitus in persons o

Cytochrome p450 3A4 messenger ribonucleic acid induction by rifampin in human peripheral blood mononuclear cells: correlation with alprazolam pharmacokinetics.

OBJECTIVE: There is significant interest in the assessment of the individual cytochrome p450 (CYP) 3A4 activity. We analyzed whether CYP3A4 messenger ribonucleic acid (mRNA) concentrations in leukocytes reflect CYP3A activity in the liver measured by alprazolam as an in vivo probe drug. We also wanted to identify whether genetically determined high CYP3A5 expression is associated with increased alprazolam clearance. METHODS: Alprazolam plasma concentrations were measured 10 hours after intake of 1 mg alprazolam. CYP3A4 mRNA concentrations in peripheral blood mononuclear cells were quantified

DNA polymorphisms: what they are and their role in human pigmentation.

The color of the skin, hair and eye is controlled by multiple genes and is among the most visible examples of human phenotypic variation. Genetics correlate phenotypic with genotypic variation. Recent scientific work reveals DNA polymorphisms at least partially responsible for some of the differences observed in human pigmentation. These are the focus of this review.

Mechanisms in Neisseria meningitidis for resistance against complement-mediated killing.

Bacterial meningitis and septicaemia is a global health problem often caused by Neisseria meningitidis. The complement system is the most important aspect of host defence against this pathogen, and the critical interaction between the two is influenced by genetic polymorphisms on both the bacterial and the host side; variations of the meningococcus may lead to increased survival in human sera, whereas humans with complement deficiencies are more susceptible to meningococcal infections. Here we discuss the mechanisms of meningococcal resistance against complement-mediated killing and the influe

[Can molecular genetic knowledge from studies of hereditary carcinoma be applied to sporadic colorectal carcinoma?].

Colorectal carcinomas without a family history are considered to be "sporadic" carcinomas, however, also have a genetic basis. Within the hereditary forms there are 15-50% of patients without a family history being carriers of de novo germline mutations. In addition, non-pathogenic polymorphisms in these tumorsyndrome-genes as well as in genes involved in the carcinogen metabolism (GST, NAT, CYP, MTHFR) are associated with an increased or decreased colorectal cancer risk. Identification of these genetic risk factors will enable individually tailored surveillance and recommendations for prophyl

Study on the Genetic Diversity Characteristics of the Endemic Plant Rhododendron bailiense in Guizhou, China Based on SNP Molecular Markers.

Rhododendron bailiense was identified as a new species in 2013, with approximately 150 individuals existing globally, found only in Dafang County and Panzhou City, Guizhou Province, China. Despite its discovery, the genetic diversity and population structure of this species remain poorly understood, hindering efforts to collect and conserve wild germplasm resources. In this study, double digest restriction-site associated DNA sequencing was conducted on 26 samples from two populations of R. bailiense to identify single nucleotide polymorphism (SNP) loci. Using these data, the research explores

Variation at genes influencing facial morphology are not associated with developmental imprecision in human faces.

Facial asymmetries are commonly used as a proxy for human developmental imprecision resulting from inbreeding, and thus reduced genetic heterozygosity. Several environmental factors influence human facial asymmetry (e.g., health care, parasites), but the generalizability of findings on genetic stressors has been limited in humans by sample characteristics (island populations, endogamy) and indirect genetic assessment (inference from pedigrees). In a sample of 3215 adult humans from the Rotterdam Study, we therefore studied the relationship of facial asymmetry, estimated from nine mid-facial la

Recombination landscape shaped by inversion polymorphisms: a high-density linkage map and chromosome-level assembly of inversion-rich spruce bark beetle genome.

Understanding the recombination landscape is crucial for revealing the extent of its variation across the tree of life and for uncovering its underlying causes and evolutionary consequences. Among the many factors influencing recombination rates, polymorphic inversions are particularly important modifiers. Increasingly, complex inversion landscapes are being documented across diverse taxa, and detailed recombination rate data are essential for advancing our understanding of variation in inversion-rich genomes. Here, we combined whole-genome sequencing of 2 two-generation families with cytogene

Genome-wide assessment of genetic diversity and population structure of Capsicum in Southwestern Colombia using DArTseq markers.

Chili peppers (Capsicum spp.) are among the most widely cultivated horticultural crops worldwide due to the high demand for their pungent fruits. Despite Colombia's high diversity of Capsicum species and varieties, its genetic resources remain underrepresented in international markets and scientific studies. This study aims to characterize the genetic diversity and population structure of Capsicum accessions from southwestern Colombia. Accessions were collected through field surveys in local markets and protected natural areas and genotyped using DArTseq platform. A total of 69 accessions repr

Selection of geographical populations suitable for artificial breeding of the Northeast China Brown Frog (Rana dybowskii).

Amphibians, as a group greatly disturbed by human activities, are at increased risk of extinction. Rana dybowskii is an anuran species with both ecological and economic significance. Due to environmental changes and human overexploitation, it has been classified as Near-Threatened. This study integrates morphological and molecular immunological approaches to identify R. dybowskii populations with greater survival and disease resistance, based on 32 morphological traits and MHC class I and II polymorphism. Morphological results showed that compared with other populations, Yichun (YC) population

[Epidermolysis bullosa : Diagnosis and therapy].

Hereditary epidermolysis bullosa is characterized by mechanically induced blistering of the skin and mucous membranes. The causative mechanisms are based on mutations in genes for structural proteins of the epidermis and the dermal-epidermal junction zone. EB is genetically and clinically heterogeneous. Based on the clinical phenotype alone, the diagnosis is difficult in newborns and toddlers, and sometimes even in adults. A precise diagnosis is only possible with laboratory tests like immunofluorescence mapping, electron microscopy and mutation analysis in specialized centers. The treatment i

Genetic Variants Associated With Anxiety and Stress-Related Disorders: A Genome-Wide Association Study and Mouse-Model Study.

IMPORTANCE: Anxiety and stress-related disorders are among the most common mental disorders. Although family and twin studies indicate that both genetic and environmental factors play an important role underlying their etiology, the genetic underpinnings of anxiety and stress-related disorders are poorly understood. OBJECTIVES: To estimate the single-nucleotide polymorphism-based heritability of anxiety and stress-related disorders; to identify novel genetic risk variants, genes, or biological pathways; to test for pleiotropic associations with other psychiatric traits; and to evaluate the as

BsmI, ApaI and FokI variants of vitamin D receptor gene polymorphism as predictors of response to treatment in immune thrombocytopenia patients.

Vitamin D receptor (VDR) polymorphisms are linked with the incidence and severity of several autoimmune diseases. The current work aimed at evaluating if VDR rs1544410 (BsmI), rs7975232 (ApaI) and rs2228570 (FokI) gene polymorphisms could be predictors of response to steroid treatment in patients with immune thrombocytopenia (ITP). The study involved 75 steroid treatment responders and 75 resistant ITP patients. All participants were subjected to VDR BsmI, ApaI and FokI gene polymorphisms analysis through genotyping by RT-PCR. Carrying the FokI F allele was significantly associated with low vi
