Dietary baicalin supplementation enhances growth performance in fattening Hu sheep via dual modulation of immunity and gastrointestinal microbiome-metabolic crosstalk.

BACKGROUND: Baicalin is a bioactive flavonoid from Scutellaria baicalensis Georgi with antioxidant, anti‑inflammatory and antibacterial properties. However, its bitter taste and susceptibility to ruminal degradation limit its practical use in mammals. Enteric coating technology might overcome these limitations by enabling targeted intestinal release. This study investigated effects of dietary supplementation with baicalin and coated baicalin on rumen fermentation, gastrointestinal microbiota, immune function and growth performance in Hu sheep. RESULTS: Thirty-six lambs with similar body weigh…

Comparative Transcriptomic Profiling Identifies Region- and Phenotypic-Background-Associated Transcriptional Programs Linked to Coat-Color Variation in Junken Meat Sheep.

Junken meat sheep display distinct coat-color phenotypes, including a white body with a black head and an entirely black coat at birth, but the transcriptional basis of variation across skin regions and phenotypic backgrounds remains unclear. We performed bulk RNA sequencing (RNA-seq) on nine skin samples from six 1-month-old lambs: black-haired head skin (BT), white-haired dorsal trunk skin (WT), and black-haired dorsal trunk skin from lambs born entirely black (B), with three biological replicates per group. The paired BT-WT comparison characterized within-individual regional differences, wh…

Rumen-Protected Betaine Improves Meat Quality in Tibetan Sheep by Influencing Intramuscular Fat Deposition, Antioxidant Capacity, and Fatty Acid Composition.

BACKGROUND: Intramuscular fat (IMF) is a key determinant of meat sensory quality, but the effects of rumen-protected betaine (RPB) on IMF deposition in Tibetan sheep remain unclear. METHODS: Sixty male, 3-month-old Tibetan lambs (Initial weight 17.72 ± 1.36 kg) were randomly assigned to a control (basal diet, n = 30) or RPB supplementation (basal diet supplemented with 0.08% RPB, n = 30) for 90 d. RESULTS: Transcriptomic and lipidomic analyses revealed that RPB modulated glycerophospholipid metabolism-related genes (DGKI and AGPAT1) and altered lipid profiles, including PC (16:0/20:5) and PC…

Maternal intestinal and placental mitochondrial dysfunction, autophagy, and ferroptosis involving intestinal microbiota by gut microbiota transplantation from sheep to mice†.

Exposure to testosterone (T) in pregnant ewes resulted in placental dysfunction and fetal growth restriction (FGR). However, the impact of T on gut microbiota and its contribution to exacerbating intestinal and placental pathologies remains uncharacterized. Pregnant sheep received intramuscular injections of 100 mg T propionate or a control vehicle. To examine the gut microbiota' s role in T-induced FGR, gut microbiota transplantation (GMT) was conducted from T-exposed and control ewes into antibiotic-treated pregnant mice. The findings demonstrated that T exposure exacerbated mitochondrial im…

Integrated Proteomics and Metabolomics Analysis Reveals Potential Pathways Underlying Onion-Mediated Regulation of Spleen Immune Function in Liangshan Black Sheep.

Feed additives play a crucial role in boosting livestock immunity, but their underlying molecular mechanisms are often incompletely understood. This study used integrated proteomics and metabolomics to systematically investigate the immunomodulatory effects of dietary fermented onion (FO) on the spleen of Liangshan Black Sheep. Results showed that FO supplementation significantly improved systemic antioxidant capacity and immune function, indicated by a higher spleen index and increased serum concentrations of SOD, MCP-1, and IL-2 (p < 0.05). Multi-omics profiling of spleen tissues identified …

Concurrent evaluation of oxidative stress biomarkers, cytokine expression, molecular identification, and histopathological findings in sheep naturally infected with Fasciola hepatica.

Ovine fascioliasis, caused by Fasciola species, challenges livestock productivity worldwide and remains insufficiently characterized at the molecular and immunological levels in endemic regions, such as Egypt. In 150 slaughtered sheep, the prevalence was determined, mitochondrial COX1 was sequenced for species identity, hepatic cytokine transcripts were quantified via qRT-PCR, oxidative stress biomarkers (MDA, TAC, TOS, and OSI) were assayed, and histopathology was evaluated. The prevalence was 15.3 %, with non-significant sex/season effects. COX1 phylogenetics confirmed that Fasciola hepatica…

Valorization of alkaline-enzymatic hydrolysates from sheep wool as bio-input-based technologies for Trichoderma harzianum cultivation and green forages.

The increase in sheep populations to meet the demand for meat generates large volumes of sheep wool, most of which is considered agro-industrial waste and poses a potential risk of contamination due to improper handling. Protein recovery is an attractive strategy for generating agricultural inputs, such as protein hydrolysates (PHs). This study aimed to evaluate the agrobiotechnological potential of sheep wool-derived PHs as (i) an alternative peptone source for Trichoderma harzianum and (ii) growth-promoting inputs in hydroponic green fodder (HGF) production. To this end, an alkaline-enzymati…

Theileria annulata infects B-cells in sheep, which display lower dissemination potential compared to T. lestoquardi-infected ovine B-cells.

Theileria annulata and Theileria lestoquardi are deadly tick-borne parasites of cattle and sheep, respectively. These parasites are transmitted by Hyalomma ticks, and their geographical distributions partially overlap, with T. annulata having a wider range. Theileria lestoquardi infection is highly pathogenic in its natural sheep and goat hosts while T. annulata infection usually causes a mild disease in these species. Interestingly, T. annulata does not produce merozoites/piroplasms in sheep and goats, therefore it is not tick-transmissible. The aim of the current study was to understand the …

Influence of Dietary Forage Neutral Detergent Fiber on Ruminal Fermentation, Chewing Activity, Nutrient Digestion, and Ruminal Microbiota of Hu Sheep.

As the key components of dietary carbohydrates, ensuring a balance between forage-neutral detergent fiber (FNDF) and rumen-degradable starch (RDS) is essential for ruminant health. Eight male Hu sheep equipped with rumen cannulas were randomly divided into four groups based on dietary FNDF content: low FNDF (L-FNDF, 6.08%), middle low FNDF (ML-FNDF, 9.47%), middle high FNDF (MH-FNDF, 12.48%), and high FNDF (H-FNDF, 15.68%), while the RDS levels (15.65% of DM on average) were similar among the four groups. A replicated 4 × 4 Latin square design was employed in this study. The results indicated …

Associations Between Non-Steroidal and Steroidal Anti-Inflammatory Drug Use, Welfare, and Milk Production in Dairy Sheep: A Multivariate Study.

This study examines the use of analgesic medications among sheep farmers, highlighting both their benefits and the significant gaps in their adoption. Our findings indicate that 26.7% of sheep farmers did not utilize any analgesics, a figure that contrasts sharply with high usage rates among swine (over 80%) and cattle (over 85%) farmers. The low utilization of NSAIDs and corticosteroids in sheep farming may be attributed to inadequate education and the lack of approved indications for these medications in Greece. Additionally, we found a positive correlation between farm size and the use of N…

MNQ derivative D19 alleviates LPS-induced inflammation and oxidative stress in sheep follicular granulosa cells through the GPX4-mediated ferroptosis.

INTRODUCTION: 2-methoxy-1,4-naphthoquinone (MNQ), a compound derived from Impatiens balsamina L., is recognized for its anti-inflammatory and antioxidant properties. However, the effects of D19, a derivative of MNQ, remain unexplored. This study aimed to elucidate the protective effect of D19 against lipopolysaccharide (LPS)-induced follicular granulosa cells (GCs) dysfunction in sheep and its underlying molecular mechanisms. METHODS: An in vitro model of GCs injury was established using LPS to induce inflammation and oxidative stress. The effects of D19 were evaluated by examining inflammato…

A new preclinical sheep model of medial meniscus anterior root repair: Part 2-Surgical strategy, technical considerations, pearls and pitfalls.

PURPOSE: To address a gap in translational research by developing a preclinical sheep model of medial meniscus anterior root (MAR) repair in vivo and to compare probabilities of potential pitfalls and difficulties with humans. METHODS: Preoperative planning and surgical procedures applied to patients were adapted to adult sheep. Eight healthy, skeletally mature, female Merino ewes between 2 and 4 years of age underwent a mini-open medial parapatellar approach to both stifle joints without luxating the patella. Next, the MAR was transected in 16 knees (8 sheep) resulting in a subtype 2A tear a…

High genetic variability in Taenia multiceps larvae: A mitochondrial DNA-based study of sheep isolates using CO1 and NADH1 genes.

Coenurus cerebralis, the larval stage of Taenia multiceps, is a significant parasitic threat to small ruminants, yet its molecular diversity remains underexplored in many endemic regions. This study investigated the genetic variability of C. cerebralis from sheep in Elazig province, Türkiye, by analyzing mitochondrial CO1 (812 bp) and NADH1 (498 bp) gene regions. Sequence data from 39 isolates confirmed the presence of T. multiceps, with 21 CO1 and 17 NADH1 haplotypes identified over half of which were singletons. Most polymorphic sites were parsimony-informative, indicating meaningful evoluti…

Reproductive neuroendocrine defects programmed by prenatal testosterone treatment between gestational days 60-90 are amplified by postnatal obesity in sheep.

Polycystic ovary syndrome (PCOS) is the leading cause of anovulatory infertility in women of reproductive age, and obesity can increase the severity and development of the PCOS phenotype. Prenatal testosterone (T) treatment between gestational days 30-90 advanced puberty and disrupted the reproductive and metabolic phenotype in female sheep, recapitulating attributes of women with PCOS, with postnatal obesity amplifying its severity. On the other hand, prenatal T treatment from gestational days 60-90 led to a much milder phenotype. We hypothesized that reproductive neuroendocrine defects progr…

Long-term safety and dose escalation of intracerebroventricular CLN5 gene therapy in sheep supports clinical translation for CLN5 Batten disease.

CLN5 neuronal ceroid lipofuscinosis (NCL, Batten disease) is a rare, inherited fatal neurodegenerative disorder caused by mutations in the CLN5 gene. The disease is characterised by progressive neuronal loss, blindness, and premature death. There is no cure. This study evaluated the efficacy of intracerebroventricular (ICV) delivery of an adeno-associated viral vector encoding ovine CLN5 (scAAV9/oCLN5) in a naturally occurring sheep model of CLN5 disease. CLN5 affected (CLN5-/-) sheep received low, moderate, or high doses of scAAV9/oCLN5 at three disease stages. The treatment delayed disease p…

Unfavorable genetic correlations between fecal egg count and milk production traits in the French blond-faced Manech dairy sheep breed.

BACKGROUND: Genetic selection has proven to be a successful strategy for the sustainable control of gastrointestinal parasitism in sheep. However, little is known on the relationship between resistance to parasites and production traits in dairy breeds. In this study, we estimated the heritabilities and genetic correlations for resistance to parasites and milk production traits in the blond-faced Manech breed. The resistance to parasites of 951 rams from the selection scheme was measured through fecal egg counts (FEC) at 30 days post-infection under experimental conditions. Six milk production…

Risk assessment of three sheep stocking modes via identification of bacterial genomes carrying antibiotic resistance genes and virulence factor genes.

In order to protect the prairie ecological environment, intensive farming has become a prevalent method of sheep stocking. However, the link between captivity stocking mode and ecological risk of sheep feces is still poorly understood. In this study, metagenomics was used to identify the environmental risk of sheep feces among three stocking modes. Our results showed that captivity mode (C) elevated antibiotic resistance in feces, with the abundance of antibiotic resistance genes (ARGs) (5.381 copies/cell) higher than that of half-pen stocking (Fh) (1.093 copies/cell) and grazing mode (Fr) (0.…

Brief hypoxia in late gestation sheep causes prolonged disruption of fetal electrographic, breathing behaviours and can result in early labour.

KEY POINTS: Brief episodes of severe fetal hypoxia can arise in late gestation as a result of interruption of normal umbilical blood flow Systemic parameters and blood chemistry indicate complete recovery within 1-2 hours, although the long-term effects on fetal brain functions are unknown Fetal sheep were subjected to umbilical cord occlusion (UCO) for 10 min at 131 days of gestation, and then monitored intensively until onset of labour or delivery (<145 days of gestation) Normal patterns of fetal behaviour, including breathing movements, episodes of high and low voltage electorcortical activ…

Connexin Hemichannel Mimetic Peptide Attenuates Cortical Interneuron Loss and Perineuronal Net Disruption Following Cerebral Ischemia in Near-Term Fetal Sheep.

Perinatal hypoxia-ischemia is associated with disruption of cortical gamma-aminobutyric acid (GABA)ergic interneurons and their surrounding perineuronal nets, which may contribute to persisting neurological deficits. Blockade of connexin43 hemichannels using a mimetic peptide can alleviate seizures and injury after hypoxia-ischemia. In this study, we tested the hypothesis that connexin43 hemichannel blockade improves the integrity of cortical interneurons and perineuronal nets. Term-equivalent fetal sheep received 30 min of bilateral carotid artery occlusion, recovery for 90 min, followed by a…

Circulating catecholamines partially regulate T-wave morphology but not heart rate variability during repeated umbilical cord occlusions in fetal sheep.

Fetal heart rate (FHR) variability (FHRV) and ST segment morphology are potential clinical indices of fetal well-being during labor. β-Adrenergic stimulation by circulating catecholamines has been hypothesized to contribute to both FHRV and ST segment morphology during labor, but this has not been tested during brief repeated fetal hypoxemia that is characteristic of labor. Near-term fetal sheep (0.85 gestation) received propranolol (β-adrenergic blockade; n = 10) or saline (n = 7) 30 min before being exposed to three 2-min complete umbilical cord occlusions (UCOs) separated by 3-min reperfusi…
