Caffeic acid inhibits chromium(VI)-induced oxidative stress and changes in brush border membrane enzymes in rat intestine.

We have previously shown that a single oral dose of potassium dichromate results in a decrease in the activities of several brush border membrane enzymes, produces oxidative stress, and alters the activities of several antioxidant enzymes in the small intestine of rats. In the present study, we have investigated the effect of treatment with the dietary antioxidant caffeic acid on potassium dichromate-induced biochemical changes in the rat intestine. Adult male Wistar rats were randomly divided into four groups: control, potassium dichromate alone, caffeic acid alone, and potassium dichromate +

Metal availability in heavy metal-contaminated open burning and open detonation soil: assessment using soil enzymes, earthworms, and chemical extractions.

The effects of heavy metal contamination on soil enzyme activity and earthworm health (bioaccumulation and condition) were studied in contaminated soils collected from an formerly open burning and open detonation (OBOD) site. Soil extraction methods were also evaluated using CaCl(2) and DTPA solutions as surrogate measures of metal bioavailability and ecotoxicity. Total heavy metal content of the soils ranged from 0.45 to 9.68 mg Cd kg(-1), 8.96 to 5103 mg Cu kg(-1), 40.21 to 328 mg Pb kg(-1), and 56.61 to 10,890 mg Zn kg(-1). Elevated metal concentrations are assumed to be primarily responsib

In vitro effects of chlorpromazine on glycerol-3-phosphate acyl transferase and 1-acylglycerol-3-phosphate acyltransferase in rat liver microsomes.

The in vitro effect of chlorpromazine on rat liver glycerol-3-phosphate acyltransferase and 1-acylglycerol-3-phosphate acyltransferase was studied. Chlorpromazine decreased glycerol-3-phosphate acyltransferase activity in a concentration-dependent manner. The inhibition was competitive with respect to palmitoyl-CoA, while non-competitive with respect to sn-glycerol-3-phosphate. Ki was determined to be approximately 0.15 mM with respect to both palmitoyl-CoA and sn-glycerol-3-phosphate. From these results, together with the inhibitory effect of amphiphilic anions and neutral detergents on the e

Purification of a carbonic anhydrase from the inner ear of the guinea pig.

A soluble carbonic anhydrase (carbonate hydro-lyase, EC 4.2.1.1) has been purified to homogeneity from the membranous lateral wall (stria vascularis, spiral ligament, spiral prominence, and outer sulcus) of the guinea-pig inner ear. About 1% of the protein of the membranous lateral wall is carbonic anhydrase. The specific activity of the enzyme in homogenates of the lateral wall is 1.6-1.8 times that of whole blood; for homogenates of the components, stria vascularis and the fraction containing the spiral ligament, the specific activities are 0.9 and 2.0 times the specific activity of whole bl

Protease, Amylase, and Secretory Phospholipase A2 Levels in Human Amniotic Fluid of Fetuses with Open Spina Bifida.

INTRODUCTION: We previously demonstrated elevated levels of secretory Phospholipase A2 (sPLA2) and Amylase in the amniotic fluid (AF) of a fetal rat Retinoic Acid (RA) Induced Open Spina Bifida (OSB) model. Our objective was to determine whether these enzymes, as well as protease, were elevated in the AF of human fetuses with OSB. METHODS: AF was collected at the time of diagnosis (Early 21-22 weeks, n=9) or during fetoscopic repair (Late 26-27 weeks, n=12). Controls (21-22 weeks, n=3) were amniocentesis samples collected for advanced maternal age. Activity levels of amylase, protease, and PL

Integrated analysis of hepatic responses to deoxynivalenol in mice: Changes in cytochrome P450 and carboxylesterase expression, activity, and transcriptomic profiles.

BACKGROUND: Deoxynivalenol (DON) is among the most prevalent mycotoxins and exerts adverse effects on the liver. However, a systematic evaluation of its effects on hepatic cytochrome P450 enzymes (Cyp450) and carboxylesterases (Ces) in mice is lacking. METHODS: Using combined in vivo and in vitro models, we investigated the impact of DON on these key enzymes. C57BL/6 mice and AML12 cells were exposed to DON. Histological and ultrastructural changes were assessed by hematoxylin and eosin staining and transmission electron microscopy. The expression and activity of target enzymes were evaluated

Discovery of juglone and its derivatives as potent SARS-CoV-2 main proteinase inhibitors.

SARS-CoV-2 as a positive-sense single-stranded RNA coronavirus caused the global outbreak of COVID-19. The main protease (Mpro) of the virus as the major enzyme processing viral polyproteins contributed to the replication and transcription of SARS-CoV-2 in host cells, and has been characterized as an attractive target in drug discovery. Herein, a set of 1,4-naphthoquinones with juglone skeleton were prepared and evaluated for the inhibitory efficacy against SARS-CoV-2 Mpro. More than half of the tested naphthoquinones could effectively inhibit the target enzyme with an inhibition rate of more

Phenylbutyrate increases pyruvate dehydrogenase complex activity in cells harboring a variety of defects.

OBJECTIVE: Deficiency of pyruvate dehydrogenase complex (PDHC) is the most common genetic disorder leading to lactic acidosis. PDHC deficiency is genetically heterogenous and most patients have defects in the X-linked E1-α gene but defects in the other components of the complex encoded by PDHB, PDHX, DLAT, DLD genes or in the regulatory enzyme encoded by PDP1 have also been found. Phenylbutyrate enhances PDHC enzymatic activity in vitro and in vivo by increasing the proportion of unphosphorylated enzyme through inhibition of pyruvate dehydrogenase kinases and thus, has potential for therapy of

Demonstration of collagenase and elastase activities in the blister fluids from bullous skin diseases. Comparison between dermatitis herpetiformis and bullous pemphigoid.

We have investigated potential mechanisms for blister formation by assaying proteolytic enzymes in the blister fluids of patients with various bullous diseases. Blister fluids were obtained from patients with dermatitis herpetiformis (DH), bullous pemphigoid (BP), chronic bullous disease of childhood (CBDC), and pemphigus vulgaris (PV). The cells were recovered by centrifugation, and the supernatants as well as the cell pellets were assayed first for collagenase activity using [3H]proline-labeled type I collagen as substrate. Collagenase activity could be detected in most cases with DH, BP, an

Cardiomyocyte-enriched OTUD5 alleviates septic cardiomyopathy by promoting NLRP3 deubiquitination and inhibiting NLRP3 inflammasome activation.

BACKGROUND: Septic cardiomyopathy (SCM) is the leading cause of mortality among patients diagnosed with sepsis. Nevertheless, the precise mechanisms underlying its pathogenesis remain poorly understood. Deubiquitinating enzymes (DUBs) play a vital role in various cardiovascular diseases. METHODS: This study investigated deubiquitinating enzymes in septic myocardial injury via public RNA-Seq. Pyroptosis was modelled in primary neonatal rat cardiomyocytes using lipopolysaccharide (LPS) and nigericin, with levels assessed by IL‑1β ELISA, Western blot, PI staining, CCK‑8 and LDH assays. Neurotropic alphavirus infection induces PARP-1 hyperactivation-mediated energy collapse in motor neurons.

UNLABELLED: Motor neurons are highly vulnerable to metabolic stress, yet the mechanisms driving their degeneration during neurotropic alphavirus infections remain unclear. Venezuelan equine encephalitis virus (VEEV) causes motor neuron injury, but the intrinsic pathways underlying this susceptibility are not fully defined. Previous work suggests alphavirus-infected motor neurons may die through caspase-independent mechanisms. Here, we show that VEEV infection induces sustained activation of the DNA repair enzyme poly(ADP-ribose) polymerase-1 (PARP-1), leading to depletion of NAD+ and ATP in mu

Preoperative activation of the renin-angiotensin system and myocardial injury in noncardiac surgery: exploratory mechanistic analysis of the SPACE randomised controlled trial.

BACKGROUND: Hypertension therapy in older adults is often suboptimal, in part because of inadequate suppression of the renin-angiotensin-aldosterone system (RAAS). We hypothesised that distinct endotypes of RAAS activation before noncardiac surgery are associated with increased risk of myocardial injury. METHODS: This was a prespecified exploratory analysis of a multicentre randomised controlled trial (ISRCTN17251494) which randomised patients ≥60 yr old undergoing elective noncardiac surgery to either continue or stop RAAS inhibitors (determined by pharmacokinetic profiles).
