INTRODUCTION: Pneumococcal multiple serotype carriage is important for evolution of the species and to understand how the pneumococcal population is changing with vaccination. We aimed to determine the impact of the 13-valent pneumococcal conjugate vaccine (PCV13) on multiple serotype carriage.
METHODS AND MATERIALS: Nasopharyngeal samples from fully vaccinated pneumococcal carriers (4 doses of PCV13, n=141, aged 18-72months) or from non-vaccinated pneumococcal carriers (0 doses of any PCV, n=140, same age group) were analyzed. Multiple serotype carriage was evaluated by DNA hybridization wit

We investigated the influence of molecular status on disease characteristics and clinical outcome in young patients (⩽ 40 years) with World Health Organization (WHO)-defined essential thrombocythemia (ET) or early/prefibrotic primary myelofibrosis (early-PMF). Overall, 217 patients with ET (number 197) and early-PMF (number 20) were included in the analysis. Median follow-up time was 10.2 years. The cumulative incidence of thrombosis, hemorrhages and disease evolution into myelofibrosis/acute leukemia were 16.6%, 8.6% and 3% at 15 years, respectively. No differences were detectable between ET

Advances in mouse neural circuit genetics, brain atlases, and behavioral assays provide a powerful system for modeling the genetic basis of cognition and psychiatric disease. However, a critical limitation of this approach is how to achieve concordance of mouse neurobiology with the ultimate goal of understanding the human brain. Previously, the common marmoset has shown promise as a genetic model system toward the linking of mouse and human studies. However, the advent of marmoset transgenic approaches will require an understanding of developmental principles in marmoset compared to mouse. In

Understanding the evolution of the human immunodeficiency virus type 1 (HIV-1) envelope during disease progression can provide tremendous insights for vaccine development, and simian-human immunodeficiency virus (SHIV) infection of non-human primate provides an ideal platform for such studies. A newly developed clade C SHIV, SHIV-1157ipd3N4, which was able to infect rhesus macaques, closely resembled primary HIV-1 in transmission and pathogenesis, was used to infect several pig-tailed macaques. One of the infected animals subsequently progressed to AIDS, whereas one remained a non-progressor.

The present study reports the evolution of the demographic characteristics and the molecular epidemiology of community-acquired methicillin-resistant Staphylococcus aureus (CA-MRSA) in Belgium from 2005 to 2009. Four hundred and ten CA-MRSA isolates were prospectively collected and screened for the presence of Panton-Valentin leucocidin (PVL) and toxic shock syndrome toxin 1 (TSST-1) encoding genes, while clinical information were recorded. PVL- and TSST-1-positive isolates were genotyped by pulsed-field gel electrophoresis (PFGE). Staphylococcal cassette chromosome mec (SCCmec) type, spa type

BACKGROUND: Budd-Chiari syndrome (BCS) is associated with parenchymal changes leading to major architecture remodelling. In order to gain further insight into the pathogenesis of BCS, we investigated expression of a set of genes involved in the course of chronic liver diseases.
METHODS: Quantitative expression of 35 selected genes involved in extracellular matrix regulation, growth factors, and angiogenesis was investigated in 13 cases of BCS and compared with 10 normal livers and 13 cirrhosis cases by real time reverse transcription-polymerase chain reaction. Differential gene expression was

Hypertrophic cardiomyopathy (HCM), the most common cause of sudden cardiac death in the young, is characterized by a diverse array of cardiac phenotypes evolving over several decades. We have developed transgenic rabbits that fully recapitulate the phenotype of human HCM and provide for the opportunity to delineate the sequence of evolution of cardiac phenotypes, and thus, the pathogenesis of HCM. We determined evolution of biochemical, molecular, histological, structural and functional phenotypes at 4 age-periods in 47 beta-myosin heavy chain-glutamine (MyHC-Q)-403 transgenic rabbits. Ca(+2)

Steroid hormones perform many essential roles in vertebrates during embryonic development, reproduction, growth, water balance, and responses to stress. The estrogens are essential for normal reproductive activity in female and male vertebrates and appear to have direct actions during sex determination in some vertebrates. To begin to understand the molecular mechanisms of estrogen action in alligators, we have isolated cDNAs encoding the estrogen receptors (ER) from the ovary. Degenerate PCR primers specific to ER were designed and used to amplify alligator ovary RNA. Two different DNA fragme

Background. Descriptions of genomic characters and dynamics related to Pseudomonas aeruginosa (PA) adaptation and survival in hospital water networks remain scarce but necessary for sustainable water management in hospitals.Methods. A new copper water network in an intensive care unit (ICU) was chronically colonized by a genotype sequence type (ST) 299 of PA and sporadically by a genotype ST2685. Sixty-eight ST299-PA and four ST2685-PA strains from ICU-water samples collected over 29 months and 4 months, respectively, were studied for genomic adaptation to copper water network. Deep mutational scans across receptor-binding domains (RBDs) of diverging SARS-CoV-2 variants reveal ongoing changes to the effects of mutations, a phenomenon known as epistasis. Careful accounting for these altered mutational effects is important in viral surveillance and forecasting, and more broadly, for understanding the impacts of epistasis on real-world viral evolutionary trajectories. Using a yeast-display RBD deep mutational scanning (DMS) platform, we measure the impacts of virtually all single amino acid mutations and single-residue deletions in the Omicron KP.3.1.1 and LP.8.1 RBDs o

Indigenous peoples of America represent the last principal expansion of humans across the globe1, yet their genetic history remains one of the least explored2. Although these populations have inhabited the continent for thousands of years3, their evolutionary history remains largely unresolved4,5, owing to the limited availability of genomic data. Here we present data on 128 high-coverage Indigenous American genomes and show they harbour extensive and previously uncharacterized genetic diversity, reflecting at least three dispersals into South America, followed by regional differentiation and

Class I homeodomain-leucine zipper (C1HDZ) transcription factors regulate diverse developmental and stress-related processes in land plants. While extensively studied in angiosperms, their functions in other lineages are poorly understood. In the liverwort Marchantia polymorpha, the sole C1HDZ orthologue (MpC1HDZ) controls herbivore defense by regulating oil body cell formation. This suggests that the function of C1HDZ genes may have diverged during the evolution of land plants. Here, we further investigated the function of MpC1HDZ in M. polymorpha and its integration into Arabidopsis (Arabido

Seasonal influenza circulation was profoundly disrupted during the COVID-19 pandemic, resulting in an unprecedented global decline in viral activity and genetic diversity. As non-pharmaceutical interventions gradually relaxed, influenza viruses re-emerged in multiple regions in 2022; however, genomic data from Latin America remain limited, particularly for the first post-pandemic season. In this study, we analyzed the epidemiological, genomic, and evolutionary patterns of influenza A and B viruses circulating in Mexico between June 2018 and June 2023, with a specific focus on the first post-pa

The Plasmodium falciparum multidrug resistance-1 (pfmdr1) gene, which encodes the ABC P-glycoprotein homolog PgH1, accumulates mutations that result in amino acid changes, namely single-nucleotide polymorphisms at codons N86Y, Y184F, and D1246Y, associated with altered parasite susceptibility to multiple antimalarial drugs. Among these antimalarials are compounds used in artemisinin-based combination (ACT) therapies, which combine a fast-acting but rapidly clearing artemisinin with a longer-lasting partner drug. The in vitro susceptibility of each of these drugs varies with the pfmdr1 genotype

OBJECTIVES: Prolonged severe acute respiratory syndrome coronavirus 2 infection in immunocompromised individuals creates an environment conducive to intrahost viral evolution. We report persistent infection in a patient with advanced human immunodeficiency virus (HIV), aiming to delineate how partial immune reconstitution shapes viral evolution.
METHODS: Nasopharyngeal swabs were obtained at 12 time points over a period of 154 days. Viral RNA was extracted and analyzed using whole-genome sequencing (WGS). Viral genome dynamics were analyzed longitudinally.
RESULTS: All sequenced isolates wer

A commensal organism has transformed into the leading multidrug resistant (MDR) hospital acquired pathogen, with serious implication on global public health. The ability of Enterococcus faecium to rapidly adapt through its great genomic plasticity, horizontal gene transfer and under selective pressures created by heavy antibiotic application both clinically and non-clinically is responsible for the phenomenon. Comparative genome studies have reported the presence of different community acquired and hospital acquired clades; hospital acquired clones being particularly the clonal complex 17 have

High-risk lineages of Pseudomonas aeruginosa pose a serious threat to public health, causing severe infections with high mortality rates and limited treatment options. The emergence and rapid spread of the high-risk lineage ST463 in China have further exacerbated this issue. However, the basis of its success in China remains unidentified. In this study, we analyzed a comprehensive dataset of ST463 strains from 2000 to 2023 using whole genome sequencing to unravel the epidemiological characteristics, evolutionary trajectory, and antibiotic resistance profiles. Our findings suggest that ST463 li

Understanding the relationship between antibiotic use and the evolution of antimicrobial resistance is vital for effective antibiotic stewardship. Yet, animal models and in vitro experiments poorly replicate real-world conditions1. To explain how resistance evolves in vivo, we exposed 60 human participants to ciprofloxacin and used longitudinal stool samples and a new computational method to assemble the genomes of 5,665 populations of commensal bacterial species within participants. Analysis of 2.3 million polymorphic sequence variants revealed 513 populations that underwent selective sweeps.

Highly pathogenic avian influenza A(H5) viruses globally impact wild and domestic birds, and have caused severe infections in mammals, including humans, underscoring their pandemic potential1-5. The antigenic evolution of the A(H5) haemagglutinin (HA) poses challenges for pandemic preparedness and vaccine design6. Here the global antigenic evolution of the A(H5) HA was captured in a high-resolution antigenic map. The map was used to design immunogenic and antigenically central vaccine HA antigens, eliciting antibody responses that broadly cover the A(H5) antigenic space. In ferrets, a central
