Structural mediation of the default-mode network in children with callosal agenesis.

Agenesis of the corpus callosum is a neurodevelopmental condition characterized by the partial or complete absence of the corpus callosum, the largest white matter bundle connecting the cerebral hemispheres. The default-mode network comprises bilateral frontal, temporal, and parietal regions that exhibit correlated activity at rest. Previous studies show that individuals with agenesis of the corpus callosum show overall preserved default-mode network functional connectivity, suggesting compensatory mechanisms for maintaining bilaterally correlated activity. In this study, we aimed to explore w

Neuroanatomy and lesion networks of central poststroke pain.

Identifying lesion sites associated with central poststroke pain (CPSP) may facilitate targeted screening for early symptoms, possibly even paving the way for preventive measures and earlier treatment initiation. Here, we test the hypothesis that damage to a nociceptive pathway extending from the brainstem to the cerebral cortex, and including white matter tracts, is associated with CPSP. We investigated the lesion locations of 72 patients with CPSP relative to poststroke comparison subjects without pain (n = 123), divided into a discovery and independent validation data set. The study include

Sexual dimorphism of white-matter functional connectome in healthy young adults.

BACKGROUND: Sexual dimorphism in human brain has garnered significant attention in neuroscience research. Although multiple investigations have examined sexual dimorphism in gray matter (GM) functional connectivity (FC), the research of white matter (WM) FC remains relatively limited. METHODS: Utilizing resting-state fMRI data from 569 healthy young adults, we investigated sexual dimorphism in the WM functional connectome. We constructed both WM-WM and GM-WM FC networks and subsequently analyzed their FC strength, functional connectivity density, and network topological properties. Based on i

The Addiction Neurocircuitry and Resting-State Functional Connectivity in Cannabis Use Disorder: An fMRI Study.

Cannabis use disorder (CUD) affects ~22-million people globally and is characterised by difficulties in cutting down and quitting use, but the underlying neurobiology remains unclear. We examined resting-state functional connectivity (rsFC) between regions of interest (ROIs) of the addiction neurocircuitry and the rest of the brain in 65 individuals with moderate-to-severe CUD who reported attempts to cut down or quit, compared to 42 controls, and explored the association between rsFC and cannabis exposure and related problems, to elucidate potential drivers of rsFC alterations. The CUD group

Altered static and dynamic functional network connectivity and combined Machine learning in asthma.

Asthma is a reversible disease characterized by airflow limitation and chronic airway inflammation. Previous neuroimaging studies have shown structural and functional abnormalities in the brains of individuals with asthma. However, earlier research has primarily focused on static changes in brain activity, neglecting the effects of asthma on the dynamic characteristics of functional brain networks. This study included 31 asthma patients and 31 healthy controls (HCs). Independent component analysis (ICA) was employed to extract changes in static functional network connectivity (sFNC) and dynami

Lateral Cerebellar Theta Burst Stimulation Can Modulate Default Mode Network Connectivity in Autistic Adults.

Developing effective neuromodulation strategies for core autistic symptoms remains a critical need. The lateral cerebellum, implicated in socio-cognitive functions often affected in autism, represents a promising target for repetitive transcranial magnetic stimulation (rTMS), which has not yet been tested in this population. Here, we report a pilot investigation of the safety, feasibility, and network-level effects of one-session intermittent theta burst stimulation (iTBS, a variant of rTMS; 1200 pulses delivered at 80% active motor threshold with a 15-minute inter-train interval) targeting th

Developmental alterations of indirect-pathway medium spiny neurons in mouse models of Huntington's disease.

Huntington's disease (HD) is a complex neurodegenerative disorder with cognitive and motor symptoms that typically manifest in adulthood. However, embryonic brain development impairments leading to cortical defects in HD mutation carriers has been shown recently supporting a neurodevelopmental component in HD. Despite HD is primarily recognized as a striatal pathology, developmental alterations in this structure, particularly during the early postnatal period, remain poorly understood. To fill this gap, we examined striatal development in newborn R6/1 mice. We found that D2 receptor-expressing

Depression in Premanifest Huntington's Disease: Aberrant Effective Connectivity of Striatum and Default Mode Network.

BACKGROUND: Depression frequently precedes motor symptoms in Huntington's disease gene expansion carriers (HDGECs), yet the neural mechanisms remain poorly characterized. OBJECTIVE: We investigated effective connectivity between the default mode network (DMN) and striatal regions in HDGECs. METHODS: We analyzed 3-T resting-state functional magnetic resonance imaging data from 98 HDGECs (48.98% females; mean age, 42.82 years). Spectral dynamic causal modeling estimated subject-level connectivity, whereas parametric empirical Bayes determined group-level effective connectivity differences betw

The neural basis of resting-state fMRI functional connectivity in fronto-limbic circuits revealed by chemogenetic manipulation.

Measures of fMRI resting-state functional connectivity (rs-FC) are an essential tool for basic and clinical investigations of fronto-limbic circuits. Understanding the relationship between rs-FC and the underlying patterns of neural activity in these circuits is therefore vital. Here we introduced inhibitory designer receptors exclusively activated by designer drugs (DREADDs) into the amygdala of two male macaques. We evaluated the causal effect of activating the DREADD receptors on rs-FC and neural activity within circuits connecting amygdala and frontal cortex. Activating the inhibitory DREA

Unveiling the axonal connectivity between the precuneus and temporal pole: Structural evidence from the cingulum pathways.

Neuroimaging studies have consistently demonstrated concurrent activation of the human precuneus and temporal pole (TP), both during resting-state conditions and various higher-order cognitive functions. However, the precise underlying structural connectivity between these brain regions remains uncertain despite significant advancements in neuroscience research. In this study, we investigated the connectivity of the precuneus and TP by employing parcellation-based fiber micro-dissections in human brains and fiber tractography techniques in a sample of 1065 human subjects and a sample of 41 rhe

Trait reward sensitivity modulates connectivity with the temporoparietal junction and Anterior Insula during strategic decision making.

Many decisions happen in social contexts such as negotiations, yet little is understood about how people balance fairness versus selfishness. Past investigations found that activation in brain areas involved in executive function and reward processing was associated with people offering less with no threat of rejection from their partner, compared to offering more when there was a threat of rejection. However, it remains unclear how trait reward sensitivity may modulate activation and connectivity patterns in these situations. To address this gap, we used task-based fMRI to examine the relatio

Static and temporal dynamic alterations of local functional connectivity in chronic insomnia.

Several studies have revealed altered intrinsic neural activity in chronic insomnia (CI). However, the temporal variability of intrinsic neural activity in CI is rarely mentioned. This study aimed to explore static and temporal dynamic alterations of regional homogeneity (ReHo) in CI and excavate the potential associations between these changes and clinical characteristics. Eighty-seven patients with CI and seventy-eight healthy controls (HCs) were included. Resting-state functional magnetic resonance imaging was performed on all subjects and both static and dynamic ReHo were used to detect lo

Arcuate dopaminergic/GABAergic neurons project within the hypothalamus and to the median eminence.

Cotransmission, meaning the release of multiple neurotransmitters from one synapse, allows for increased diversity of signaling in the brain. Dopamine (DA) and γ-aminobutyric acid (GABA) are known to coexpress in many regions such as the olfactory bulb and the ventral tegmental area. Tuberoinfundibular dopaminergic neurons (TIDA) in the arcuate nucleus of the hypothalamus (Arc) project to the median eminence (ME) and regulate prolactin release from the pituitary, and prior work suggests dopaminergic Arc neurons also cotransmit GABA. However, the extent of cotransmission, and the projection pat

Stimulation of the ventromedial prefrontal cortex and the perception of affective faces. An effective connectivity analysis.

The ventromedial prefrontal cortex is widely linked with emotional phenomena, including appraisal, modulation, and reward processing. Its perigenual part is suggested to mediate the appetitive value of stimulation. In our previous study, besides changes in evoked MEG responses, we were able to induce an apparent behavioral bias toward more positive valence while interpreting the ambiguous, morphed faces after the effect of excitatory tDCS stimulation of the perigenual ventromedial cortex (pgVM). In the present study, we reanalyze these data to reveal the importance of functional links between

Galaninergic and hypercapnia-activated neuronal projections to the ventral respiratory column.

In mammals, the ventral respiratory column (VRC) plays a pivotal role in integrating neurochemically diverse inputs from brainstem and forebrain regions to generate respiratory motor patterns. VRC microinjection of the neuropeptide galanin has been reported to dampen carbon dioxide (CO2)-mediated chemoreflex responses. Additionally, we previously demonstrated that galaninergic neurons in the retrotrapezoid nucleus (RTN) are implicated in the adaptive response to hypercapnic stimuli, suggesting a link between RTN neuroplasticity and increased neuronal drive to the VRC. VRC neurons express galan

Divergent suicidal symptomatic activations converge on somato-cognitive action network in depression.

Individuals with depression have the highest lifetime prevalence of suicide attempts (SA) among mental illnesses. Numerous neuroimaging studies have developed biomarkers from task-related neural activation in depressive patients with SA, but the findings are inconsistent. Empowered by the contemporary interconnected view of depression as a neural system disorder, we sought to identify a specific brain circuit utilizing published heterogeneous neural activations. We systematically reviewed all published cognitive and emotional task-related functional MRI studies that investigated differences in

Altered structure and functional connection of subcortical gray matter in female patients with classical trigeminal neuralgia.

The etiology of classical trigeminal neuralgia (CTN) is still unclear. A better understanding of the cerebral structural and functional changes in female patients with CTN may provide important novel insights into the pathophysiologic mechanisms of female CTN. A total 37 female CTN patients were included and referred to MRI scans, comprising with 19 left CTN and 18 right CTN patients. We analyzed the volume and shape of subcortical gray matter (GM), and the functional connectivity (FC) between the accumbens nucleus (NAc) and whole brain in right and left CTN patients respectively.
